Phylogenetic distribution of bicuculline-sensitive gamma-amino-butyric acid (GABA) receptor binding.

Bicuculline-sensitive [3H]GABA receptor binding was studied in membrane fractions prepared from vertebrate whole brain or invertebrate cephalic ganglia. In tissue not treated with Triton X-100, a significant amount of bicuculline-displaceable [3H]GABA binding was detected in the brains of all vertebrates studied, with the hagfish brain binding over twice as much [3H]GABA as the spiny dogfish, the next oldest species. All other vertebrates bound similar amounts of [3H]GABA, being one-third to one-fourth that observed in the hagfish. In contrast, after Triton treatment, the hagfish displayed the least amount of bicuculline-sensitive [3H]GABA binding and, under those conditions, the amount of binding observed increased in an evolutionary fashion. No measurable bicuculline-sensitive GABA receptor binding was noted in any invertebrate studied. These results suggest that bicuculline-sensitive GABA receptors are present in the brains of all vertebrates and that during the course of evolution there developed a Triton-sensitive substance(s) whose presence modifies the kinetic properties of this receptor site.

Cardiovascular effects of therapeutic doses of tricyclic antidepressants: importance of blood level monitoring.

The cardiovascular effects of therapeutic doses of tricyclic antidepressants are reviewed and their clinical significance discussed. At standard therapeutic doses these antidepressant drugs produced small changes in heart rate and several intervals of the EKG in some patients, but clinically significant effects on the EKG are usually seen only in patients with high blood levels of the drugs; these high plasma levels are often above those needed for therapeutic antidepressant effects. In contrast, the orthostatic hypotension induced by antidepressants usually occurs at dosages and blood levels within the therapeutic range, and, therefore, this side-effect may be of greater clinical significance in standard clinical treatment with antidepressants. A review of recent studies suggest that adequate doses of antidepressants may be used safely in geriatric depressed patients and in many patients with stable preexisting cardiac disease. However, more frequent monitoring of plasma levels of antidepressants and EKGs may be particularly important in these groups of patients.

[Materials testing demonstration of the suitability of a viscous plastic silicone elastomer for long-term functional impressions].

Comparative laboratory tests with elastomers as well as clinical trials showed that the visco-elastic silicone elastomer "Stefanat zähplastisch" (manufacturer: VEB Arzneimittelwerk Dresden, Leipziger Arzneimittelwerk) is suited for long-term dynamic impression. It is necessary to use a special, exactly dosed cross-linking agent and to apply the reactive material carefully.

Automated treatment for flight phobia: a 3 1/2 year follow-up.

The authors contacted 43 patients who had been given audiovisual treatment for flight phobia 3 1/2--5 1/2 years previously. Of the 26 subjects who had flown successfully by the end of treatment, 23 had maintained their ability to fly at follow-up. Eighteen said their attitude toward flying was more positive since treatment, and 21 rated their comfort level while flying as "much better" or "better"; however, only 3 said they were "cured." Eleven patients said the treatment had helped to alleviate other fears or anxieties.

[Studies on the setting behavior of dental gypsum. 1. Experimental characterization of the hardening process].

Using penetration tests, the authors studied the setting of dental gypsums (alpha- and beta-hemihydrate) under different conditions of processing (temperature of the mixing water, mixing time, addition of electrolytes). The suitability of penetrometry as a standardizable testing procedure for characterizing the setting process of gypsums is demonstrated and the effects of different materials and processing parameters on setting are discussed.

Rhegmatogenous retinal detachment in infancy.

A 5-week-old male infant had small stature, microphthalmia, bilateral rhegmatogenous retinal detachments, ventricular septal defect, and fundus lesions suggestive of previous chorioretinitis. Diagnostic workup failed to disclose a cause of the intraocular disorder. Bilateral retinal detachment surgery, performed when the boy was 2 and 3 months of age, has appeared successful 12 months postoperatively. This may be a description of the youngest patient with rhegmatogenous retinal detachments that were subsequently repaired.

Arthritis associated with intestinal-bypass procedure for morbid obesity. Complement activation and characterization of circulating cryoproteins.

Five patients were studied after intestinal-bypass operations for morbid obesity to determine if the arthritis observed in three of these patients was associated with circulating cryoproteins. Only serum from the three patients with acute arthritis contained cryoprotein complexes. The complexes comprised IgG, IgM, IgA, complement components C3, C4, C5 and IgG antibody against Escherichia coli and Bacillus fragilis. The C3-activator fragment of the properdin complex was identified in fresh serum samples obtained from two of the three patients with circulating cryoprotein complexes and arthritis. These observations suggest that systemically absorbed intestinal bacterial antigens may lead to (or contribute to) the formation of circulating cryoprotein complexes. Circulating cryoprotein complexes activated both the classical and alternate complement pathways and may be important in the pathogenesis of the arthritis associated with the intestinal-bypass procedure for morbid obesity.

RNA-dependent DNA-polymerase activity in human milk.

A simple method is described for testing milk specimens from nursing mothers for the presence of RNA-dependent DNA-polymerase activity. Positive results were obtained in five of 137 women (3.6%) without a family history of breast cancer, and in six of 31 women (19.3%) with a family history of breast cancer.

The pathogenesis of arthritis associated with acute hepatitis-B surface antigen-positive hepatitis. Complement activation and characterization of circulating immune complexes.

Circulating immune complexes were identified in cryoproteins isolated from serial samples of serum from six patients with acute viral hepatitis with and without arthritic symptoms. Cryoprecipitates were analyzed for the presence of hepatitis-B surface antigen (HBsAg) and hepatitis-B surface antibody (anti-HBs) by hemagglutination inhibition and hemagglutination. Complement components were detected by counter electrophoresis, and immunoglobulins were detected by gel diffusion. HBsAg, IgG, and IgM were identified in cryoprecipitates from all hepatitis patients, but were higher in concentration in patients with arthritis. Only cryoprecipitates from hepatitis patients with arthritis contained IgA and complement components C3, C4, and C5 as well as IgG and IgM, which disappear with resolution of the arthritis. The subtypes of IgG in these cryoprecipitates were predominantly the complement-fixing IgG1 and IgG3, HBsAg and anti-HBs were concentrated several-fold in the cryoprecipitates when compared to the serum concentration. Sequential studies in two patients demonstrated that the initial appearance of anti-HBs in the cryoprotein complex was associated with the detection in the complex of IgM suggesting a primary immune response to HBsAg. The C3 activator fragment (C3A) of the properdin complex was found in fresh serum obtained from three hepatitis patients with arthritis and not in uncomplicated hepatitis. The cryoprecipitable immune complexes from patients with arthritis converted C3PA in fresh normal sera to C3A in vitro whereas cryoprotein isolated from patients with uncomplicated hepatitis had no such effect. Thus, the transient appearance of circulating complement-fixing immune complexes in patients with the arthritis of acute hepatitis is associated with activation of both classical and alternate complement pathways and suggests that they play an important role in the pathogenesis of these serum sickness-like extrahepatic symptoms.
