Cell cycle regulated synthesis of stable mouse thymidine kinase mRNA is mediated by a sequence within the cDNA.

The cDNA for mouse thymidine kinase (TK) was isolated from a cDNA library in lambda-gt11 and sequenced. It was used as a probe to follow the time course of TK mRNA expression in growth stimulated mouse fibroblasts. Linked to the HSV-TK promoter the cDNA was able to transform LTK-cells to the TK+ phenotype. The transformed cells expressed the TK mRNA and enzyme activity in a growth dependent fashion suggesting that the regulatory element is localized on the cDNA.

Increased levels of dihydrofolate reductase mRNA can be measured in normal, growth-stimulated mouse fibroblasts.

Levels of mRNA for the enzyme dihydrofolate reductase (EC 1.5.1.3) were determined in growth-stimulated 3T6 cells which contained wild-type dosage of the gene coding for this enzyme. As in the case of methotrexate-resistant cells having highly amplified levels of genes for dihydrofolate reductase, an increase in dihydrofolate reductase mRNA by a factor of 2-4 can be determined when cells enter the S phase. This increase is inhibited by sodium butyrate (which inhibits growth-stimulated 3T6 cells in mid G1 phase) but not by hydroxyurea (which inhibits in early S phase). We conclude that with the available methods it is possible to study the regulation of S phase-specific enzymes after growth stimulation at the level of the mRNA, even if gene amplification is not possible or desirable.

Effect of sodium butyrate on induction of cellular and viral DNA syntheses in polyoma virus-infected mouse kidney cells.

Sodium butyrate inhibited initiation of viral and cellular DNA replication in polyoma virus-infected mouse kidney cells. Ongoing viral or cellular DNA replication, however, was not affected by the presence of the substance. Butyrate had no effect on T-antigen synthesis and on the stimulation of transcription, one of the earliest reactions of the infected cells to the appearance of T-antigen, nor did it inhibit expression of late viral genes (synthesis of viral capsid proteins). In addition to blocking the onset of DNA synthesis, butyrate also inhibited stimulation of the activities of enzymes involved in DNA synthesis. When butyrate was removed, viral and cellular DNA syntheses were induced in parallel after a lag period of approximately 4 h. At the same time, the activities of enzymes involved in DNA synthesis increase. If protein synthesis was inhibited during part of the lag period, the initiation of DNA synthesis was retarded for the same time interval, suggesting that the proteins involved in the initiation of DNA replication had to be made. We have developed an in vitro system for measuring DNA synthesis in crude nuclear preparations which mimics the status of DNA replication in intact cells and may help in future experiments to study the requirements for initiation of cellular and viral DNA synthesis and the possible involvement of T-antigens in this reaction.

[Automatic recognition of age - specific development of the EEG in infancy and early childhood - a five year follow-up study (author's transl)].

This study was initiated with a view to early detection of dysfunctions of the central nervous system (CNS) and to observe the physiological development of the CNS by means of EEG obtained in the age-groups: new-born, half, 1, 2, 3, 4 and 5 years. This was supported by standardized clinical examinations for neurological and somatic findings and by standardized tests of psychomotoric and intellectual development. In this investigation the data as used for automatic processing consisted of 110 EEGs in the age-groups: half, 1, 2, 3 and 4 years. Bipolar EEGs were recorded using "ten-twenty" method with the following leads: F4--C4, P4--O2, F3--C3 and P3--O1. For feature extraction three methods of data reduction were applied, i.e. interval-amplitude, spectral analysis and the autoregressive model. From the features thus obtained, age-specific frequency parameters were selected by statistical methods; further evaluation was performed by cluster and discriminant analysis. The former showed an unequivocal case grouping for each age-group. Using unmatched samples of only clinically healthy children from the three age-groups 1/2, 1 and 2 years, linear discriminant analysis were applied to the parameters of the three methods of data reduction. This procedure yields in mean recognition rates of 95%, 98% resp. 98% in hold-one-out classification. Similar results are obtained with samples from four or five age groups (1/2 to 4 years). The results of this type of automatic EEG analysis show that discriminant analysis can be used to allocate EEGs to specific age-groups; hence, it may readily be ascertained whether differences between chronological age and development age as evidenced by the EEG exist.

Relationship between background activity and subclinical seizure pattern.

Open- and closed-eyed EEG records were made in frontal, temporal and occipital regions, in order to examine the time relationships between various of their characteristics, in connection with the appearance of subclinical seizure patterns. 1. In the range of alpha frequency, both with open and closed eyes, the standard deviation of frequency over all brain areas decreased both before and after subclinical seizure patterns as compared to periods without pattern, and the number of temporal waves was reduced. Frontally, the mean amplitude was increased only in open-eyed records. The time relationship of the appearance of alpha maxima differed over all regions, but was closest in frontal records in both open- and closed-eyed records, and independent of the pathological pattern. In closed-eyed frontal records, a changeover in the time lead between the hemispheres could be observed, with one side leading during the period preceding the pattern, and the other after the pattern. 2. In the range of theta frequency, occipital open-eyed records showed slowing of the frequency both before and after the subclinical seizure pattern, with an increase in the standard deviation. In closed-eyed records made in the period preceding the pattern, an increase of the mean amplitude was observed frontally and occipitally, as well as an increase of the standard deviation of the amplitude in all regions. Slowing of the theta frequency was encountered in the frontal regions. Differences in the periods preceding and following subclinical seizures were to be seen only in the theta range.

[Re-integration of children into the biological (partial) family].

The article deals with some aspects of a study about reintegration of children from Austrian SOS Children's Villages into their original families. The knowledge that children actually not always return to their original families is of outstanding importance. In the meantime, very often the family has substantially changed because family members joined or left the family. Therefore the term 'reintegration into the original family' reflects the situation insufficiently. Ignoring these changes, many times lead to difficulties within family life.
