Control of 4-8S RNA transcription at the midblastula transition in Xenopus laevis embryos.

Transcription of Xenopus laevis U1 snRNA genes is subject to a precise program with respect both to the timing of activation at the midblastula transition (MBT) and to the relative levels of the two embryonic U1 RNAs (xU1b1 and b2) that are made. Here, we demonstrate that exogenous xU1b genes injected into developing X. laevis embryos come under the same controls as the endogenous genes. Injected U1 genes, unlike exogenous RNA polymerase III genes, remain quiescent until MBT and their activation at MBT requires protein synthesis during the early cleavage stages. Significantly, the onset of 4-8S RNA transcription occurs at the normal time, even when the DNA content of the embryo has been increased by injection of exogenous DNA or reduced through cleavage arrest, indicating that transcriptional activation at MBT is independent of the ratio of DNA (nucleus) to cytoplasm. In cleavage-arrested (coenocytic) embryos, the reduced level of DNA at MBT results both in a decrease in snRNA and tRNA synthesis (reflecting the lower gene dosage) and in a prolonged synthesis of large amounts of unusual RNA polymerase III transcripts, OAX RNAs. In normally cleaving embryos, small amounts of these unstable OAX RNAs (encoded by satellite I DNA) are synthesized only briefly at MBT. Our demonstration that RNA and DNA metabolism is aberrant in cleavage-arrested embryos requires reevaluation of previous experiments on transcriptional activation that utilized such coenocytic embryos.

Nuclear processing of the 3'-terminal nucleotides of pre-U1 RNA in Xenopus laevis oocytes.

U1 small nuclear RNA is synthesized as a precursor with several extra nucleotides at its 3' end. We show that in Xenopus laevis oocytes, removal of the terminal two nucleotides occurs after the RNA has transited through the cytoplasm and returned to the nucleus. The activity is controlled by an inhibitor of processing, which we call TPI, for 3'-terminal processing inhibitor. This inhibitor is sensitive to both micrococcal nuclease and trypsin treatment, indicating that it is a nucleoprotein. TPI inhibits the 3' processing of pre-U1 RNAs that have 5' ends containing m7G caps but not mature m2,2,7G caps; this finding suggests that TPI interacts directly or indirectly with the 5' end of pre-U1 RNA. The inhibition of processing by TPI, almost complete at 19 degrees C, is reversibly inactivated at slightly higher temperatures. TPI activity is solely in the soluble fraction of oocyte nuclear extracts, in contrast to the 3'-terminal processing activity, which is present in both the particulate and soluble fractions. We propose that the differential processing of the 3'-terminal nucleotides of pre-U1 RNA after its return from the cytoplasm, but not before its exit from the nucleus, may be due to the association of TPI with the m7G cap on the newly synthesized pre-U1 RNA.

3'-end-dependent formation of U6 small nuclear ribonucleoprotein particles in Xenopus laevis oocyte nuclei.

We have identified and characterized a U6 small nuclear (sn) ribonucleoprotein particle (RNP) present in the nuclei of Xenopus laevis oocytes. The structure of this U6 snRNP was investigated by native gel shift analysis and a combination of RNA-protein UV cross-linking, RNase T1 fingerprinting, and immunoprecipitation assays. These analyses demonstrate that certain forms of U6 snRNA associate with the 50-kDa nuclear antigen La both in vivo and in vitro. The La protein binds the stretch of uridylates at the 3' hydroxyl end of newly synthesized U6 snRNA. La does not bind to mature U6 snRNAs that have 2',3'-cyclic phosphate (greater than p) groups at their 3' ends (E. Lund and J. E. Dahlberg, Science 255:327-330, 1992) or to U6 snRNAs in anti-Sm-precipitable U4/U6 snRNPs. We propose that 3'-end modification, including posttranscriptional UMP addition, modulates the binding of La protein to U6 snRNA which, in turn, may affect the function of this RNA.

Childbearing and use of oral contraceptives: impact of educational level. The Nordland Health Study.

STUDY OBJECTIVE: The aim was to correlate the level of education to childbearing pattern and previous use of oral contraceptives in middle aged women. DESIGN AND SETTING: The study was a cross sectional survey of 3608 women aged 40-42 years in a county in northern Norway and involved 72% of all women in the age bracket living in the county. Information about childbearing and use of oral contraceptives was obtained from a self administered questionnaire. MAIN RESULTS: Level of education was inversely associated with parity and positively related to age at first and last birth, to the use of oral contraceptives at any time, and particularly to the use of oral contraceptives before the first birth. CONCLUSIONS: Women with high level of education have relatively low parity and postpone childbearing by using oral contraceptives.

Oral contraceptives and breast cancer. A review with some comments on mathematical models.

The relationship between oral contraceptive use and breast cancer is discussed on the basis of information given in review articles, meta-analyses and editorials emphasizing methodological problems related to bias and confounding. Over the last few years a shift in opinion has taken place. Most reviewers now consider that long-term use of oral contraceptives is associated with an increased risk of premenopausal breast cancer and no effect among postmenopausal breast cancer. This result is compatible with an additive effect (in rate measure scale) of oral contraceptive use on breast cancer risk.

SV40 T-antigen-binding sites within the 5'-flanking regions of human U1 and U2 genes.

The 5' flanking regions of the genes (U1 and U2) encoding the human U1 and U2 small nuclear RNAs (snRNAs) each contain sequences that bind specifically to the simian virus (SV40) large tumor antigen (T.Ag). Substitution of these sites with sequences that lack T.Ag-binding sites did not block accumulation of U1 or U2 snRNA in a variety of cell types, but deletion of these regions resulted in the total loss of expression. Thus, these sequences may serve only a spacing function, and the T.Ag-binding sites appear not to be necessary for expression. However, coexpression of T.Ag markedly reduced expression of a U1 gene containing a high-affinity T.Ag-binding site (from the SV40 genome) in place of the U1 T.Ag-binding site. In contrast, coexpression of T.Ag enhanced synthesis of U2, but not U1, snRNA, independent of the presence of the T.Ag-binding sites. Thus, while the consensus T.Ag-binding sites within the U1 and U2 promoter regions do not appear to influence expression, the binding of SV40 T.Ag to a high-affinity site can lead to significant repression of a strong snRNA promoter, and T.Ag can enhance expression of another in the absence of a known binding site.

[Vaginal cytological smears--experiences from general practitioners in Nordland].

A survey was conducted among 142 general practitioners in Nordland county, Norway, on their use of a Pap smear. About 75% of the general practitioners took a Pap smear more often than every third year from women who used either oral contraceptives or IUD. The doctors reported taking a Pap smear less frequently from elderly women. It is concluded that the present pattern of use of Pap smears is not in conformity with the national guidelines.

Near diploid large bowel carcinomas have better five-year survival than aneuploid ones.

One hundred patients who underwent surgery for large bowel carcinoma between 1978 and 1982 were examined by flow cytometric DNA quantitation of fresh tumor specimens and divided into an aneuploid (AN) group of 63 and a near diploid (ND) one of 37. All patients were followed until death (n = 63) or until December 31, 1988. Forty-one patients (65%) with AN tumors died of cancer, as did 12 patients (32%) with ND carcinomas. Thus patients with ND tumors had a better survival rate (P = 0.04) than did those with AN ones. The difference was apparent in Dukes' Stages A, B, and C, but not in Stage D. All patients with tumors in this stage died from their carcinomas irrespective of ploidy group. Multiple regression analyses (Cox) of prognostic factors revealed that the most important prognostic variables were (in descending order) Dukes' Stage D, Dukes' Stage C, and DNA ploidy pattern. Histologic grade was not significant as an independent prognostic variable. These results indicate that the presence of a distinctly aneuploid DNA ploidy pattern in large bowel carcinoma is an important prognostic variable that worsens survival rates significantly.

Symptomatic treatment versus combination chemotherapy for patients with extensive non-small cell lung cancer.

In a randomized clinical trial, 87 patients with inoperable, extensive non-small cell lung cancer (NSCLC) were randomized to receive either combination chemotherapy (cisplatin at 70 mg/m2 intravenously [i.v.] on day 1 and etoposide at 100 mg/m2 i.v. on day 1 and 200 mg/m2 orally on days 2 and 3) or symptomatic treatment. No statistically significant differences in survival time were found between the two treatment techniques. A major problem in the interpretation of the results was the use of semicurative radiation therapy (3000 to 4200 cGy) to the primary tumor and mediastinum, which was given with symptomatic intent. Three long-term survivors were seen in the latter group.

[Disability pensions in Nordreisa in 1980-87].

During the years 1980-87 a total of 287 persons received disability pensions in the municipality of Nordreisa in northern Norway. The incidence rate among men increased to 2.1% of all economically active men aged 16-66 years in 1984 with a subsequent decrease to 0.8% in 1987. The incidence rate among women increased slowly during the whole period. In the males a strong correlation was found between incidence of disability pension and unemployment the year before. Inter-observer validation of the diagnoses showed considerable discrepancies between the evaluations of two independent doctors and between the local doctor and the official statistics.

Formation of novel C21-bile acids from cholesterol in the rat. Structure identification of the major Di- and trihydroxylated species.

We recently showed that previously unknown di- and trihydroxylated C21-bile acids are major degradation products of sitosterol and campesterol in bile-fistulated female Wistar rats. Using a mixture of 4-14C- and 22-3H-labeled cholesterol it was shown that such C21-bile acids are formed also from cholesterol in amounts up to about 25% of the total formation of bile acids. The C21-bile acids were formed from labeled cholesterol also in perfused rat liver, demonstrating that the liver is the site of synthesis. The major trihydroxylated C21-bile acids in bile were identified, by means of mass spectrometry, NMR, stereospecific dehydrogenases, and reagents, as 5 beta-pregnan-3 alpha, 11 beta, 15 beta-triol-21-oic acid and 5 beta-pregnan-3 alpha, 11 beta, 15 alpha-triol-21-oic acid. The corresponding 11-oxo-isomers were also present. A minor trihydroxylated C21-bile acid was identified as 5 beta-pregnan-3 alpha, 11 beta, 16-triol-21-oic acid. The major dihydroxylated C21-bile acid was identified by the same means as 5 alpha-pregnan-3 alpha, 12 alpha-diol-21-oic acid. Male rats converted 4-14C-cholesterol into C21-bile acids less efficiently than did female rats. None of the C21-bile acids from male rats contained a 15-hydroxyl group. It is speculated that the novel C21-bile acids are formed both from cholesterol and from plant sterols by an initial hydroxylation at C21 followed by peroxisomal or mitochondrial beta-oxidation. The presence of a hydroxyl group at C15 may facilitate this reaction. The above formation of C21-bile acids shows that mammalian liver is able to degrade the side chain of cholesterol beyond the C24 stage, even in the absence of a blocking group at C24. C21-bile acids, or one of their precursors, are hydroxylated in the liver by a hitherto unknown 11 beta-hydroxylase. The possible physiological importance of the C21-bile acids is discussed.

[P pills and risk of cancer].

The article summarizes present knowledge on the relationship between use of oral contraceptives use and the risk of hormone dependent cancers. The authors conclude that, at present, this knowledge does not warrant a change in current prescription practice.

Education and breast cancer mortality: experience from a large Norwegian cohort study.

Socioeconomic status has been regarded as an established risk factor for breast cancer. We have studied the association between socioeconomic status, operationalized as years of completed schooling, and breast cancer mortality in a Norwegian cohort. A total of 425,884 currently married women, aged 35-54 years at the Norwegian Census in 1970, was followed through 1985. No relation was found between level of education and breast cancer mortality.

Reduction in sella turcica volume. An effect of long-term treatment with the somatostatin analogue, SMS 201-995, in acromegalic patients.

Ten patients with acromegaly were treated with the long-acting somatostatin analogue, Sandostatin (SMS 201-995, octreotide) for more than one year. Computerized tomography was performed before and on 4 different occasions during the treatment. Sella turcica volumes were calculated in nine patients and showed a gradual decrease in all, averaging 32% +/- 14.6%, P less than 0.001 at the end of the study, which is probably indicative of a simultaneous reduction in adenoma size.

Psychosocial well-being in testicular cancer patients.

149 testicular cancer patients with no evidence of disease for 3 or more years filled in a questionnaire which covered the following subjects: psychosocial well-being, working ability and use of analgesics/tranquilisers. The questions were chosen to compare cancer patients' morbidity with that of age-matched controls. The patients had been treated with surgery (32 patients), radiotherapy (39 patients), cisplatin-based chemotherapy plus surgery (46 patients) or chemotherapy plus radiotherapy with or without surgery (32 patients). Since no systematic differences between the treatment groups were found, the analyses were undertaken with all patients combined. The patients felt significantly less exhausted after a working day, were more satisfied with life and felt stronger and more fit than the controls. On the other hand, the patients reported a significantly higher incidence of anxiety and depression than the normal population. The results indicate that patients treated for a malignant disease may have greater fluctuations in mood and affect than the general population.

Breast cancer mortality and the change in fertility risk factors at menopause: a prospective study of 800,000 married Norwegian women.

I examined breast cancer mortality in relation to fertility factors in a cohort of 800,814 married Norwegian women aged 25-74 years at the start of follow-up. Women aged 25-44 years with a first birth after the age of 35 years had a RR = 2.58 compared with women with a first birth before the age of 20 years. For women aged 45-74 years, the corresponding RR was 1.35. On the other hand, the rate differences between women with a late first birth (35+ years) and an early birth (less than 20 years) were similar for women of different ages. Small rate differences were found for premenopausal women between uniparous women and women with 6-7 children, but postmenopausal women with many children had lower rates of breast cancer mortality than uniparous women. For postmenopausal women the rate differences were stable over age categories. This study points toward a change in fertility risk factors at menopause.

Central nervous system calcifications following tap water rinsing in autopsy material from children. A pitfall.

CNS from 80 children collected consecutively at the Institute of Forensic Medicine, Aarhus, were investigated. Owing to peculiar calcifications of the first 41 CNS examined, we gradually came to suspect that some external factors had influenced the results. Different fixation methods used on the CNS, rinsed or not rinsed in tap water, led us to suggest that the relatively high calcium content in the tap water in our part of the country was the cause. The non-pathologic calcification could presumably mimic metabolic diseases.
