The Rieske FeS center from the gram-positive bacterium PS3 and its interaction with the menaquinone pool studied by EPR.

The Rieske 2Fe2S center from Bacillus PS3, a Gram-positive thermophilic eubacterium, has been studied by EPR spectroscopy. Its redox midpoint potential at pH 7.0 was determined to be +165 +/- 10 mV and was found to decrease with an apparent slope of -80 mV/pH unit above pH 7.9. The Qo-site inhibitor stigmatellin induced spectral changes analogous to those reported for Rieske centers from mitochondria and chloroplasts. The redox midpoint potential of the PS3 Rieske cluster was not affected by stigmatellin. The orientation of the g tensor was similar to other Rieske centers (gz and gy are oriented parallel, gx is oriented perpendicular to the membrane plane). The shape of the EPR spectrum of the Rieske cluster from PS3 changed as a function of the redox state of the menaquinone (MK) pool. This permitted the redox midpoint potential of the MK pool to be determined in the membrane. Values of -60 +/- 20 mV at pH 7.0 and of -130 +/- 20 mV at pH 8.0 were obtained. The results are compared with already published data from other Rieske centers. It is proposed that all Rieske centers that function in electron transport chains using MK as pool quinone show common features that distinguish them from Rieske centers operating in ubiquinone- or plastoquinone-based electron transfer chains.

[Ultrasonic diagnosis in infiltrative gastrointestinal diseases (author's transl)].

The cockade phenomenon in the ultrasound picture is a sign of an infiltration of the gastrointestinal wall caused by tumour or inflammation. In 27 out of 40 patients with known infiltrations of the wall the cockade phenomenon could be demonstrated. In 16 out of 21 patients in whom this ultrasound phenomenon had primarily led to a suspected wall infiltration further diagnostic investigations proved organic wall thickening. Even if the cockade phenomenon is thus neither a specific nor a sensitive sign of gastrointestinal wall infiltrations the observation of this phenomenon is particularly important in patients in whom wall infiltrations of the gastrointestinal tract are not suspected.

[Echographic diagnosis of the pancreas. Catamnesis of 481 cases (author's transl)].

In order to assess the efficiency of ultrasonic diagnosis of the pancreas, echographic findings in 481 patients were compared with the final diagnosis in the case sheets. Agreement between the echographic results and the final diagnosis was found in 94.6% with normal findings, in 77% of patients with pancreatic carcinoma and in 85% of patients with chronic pancreatitis. Altogether we have found agreement between the echographic results and the final diagnosis in 86.6% of all cases referred for diagnosis of the pancreas. The results correspond to the fiqures given by Rettenmaier. With adequate experience ultrasonic diagnosis should therefore be used as early as possible with a broad indication for all diseases of the pancreas.

The development of anorectal continence and its significance in the diagnosiso of Hirschsprung's disease.

The development of the anorectal continence reaction was studied in a series of 138 electromanometric recordings performed in 62 newborn and six premature infants. Forty-five babies were observed in a longitudinal study. In 11% an internal sphincter relaxation was observed at the time of birth. The same reflex was present in about 50% after the third day and in all infants after the 12th day of life. The development of a rectal continence reaction depends on an intact internal sphincter relaxation. The puborectalis muscle, is fully functional at birth. On the basis of these findings electromanometric recordings can be used to exclude aganglionic megacolon as early as the first day of life. An absent internal sphincter relaxation reflex, is diagnostic only after the 12th day of life.
