Neuropeptides with immunoregulatory functions: current status of investigations.

Neuropeptides (NP) from the central nervous system and the peripheral nervous system play a modulating role in the immune system. In principle, all immunological functions are influenced by NP either in terms of nonspecific immunity or cellular immunity. In the present review, some aspects of this interaction are summarized, namely the existence of receptors for NP on immune cells and the production of NP by leukocytes. Finally, the pathogenetic role of NP in diseases and the future perspectives of neuroimmunology are outlined.

[Effects of anesthesia and surgical trauma on the immune response of pediatric patients].

The AA. have evaluated the effects of anaesthesia and surgical trauma on the ability of lymphocytes from 10 patients to release Leukocyte Inhibiting Factor (LIF) after stimulation with phytohaemoagglutinin (PHA). In addition, they have studied the electrophoretic mobility of Polymorphonuclear-cells (PMN) and lymphocytes of 8 patients after incubation in autologus plasma. The experimental results show decrease of LIF production after operation in 6 individuals and reduction of eletrophoretic mobility of PMN and lymphocytes. The last finding is not significant on statistical point of view.

[Endotoxemia and phagocytic function in premature infants. II. Role of the blood platelets].

The AA. have studied the relationship between endotoxaemia, detected in premature children and platelets function, according to the most recent finding, supporting the presence of specific receptors for endotoxin at the surface of thrombocytes. The finding of 6 positive Limulus test on platelets over 7 cases support the ability of clearance from platelets towards endotoxin. Furthermore, the AA. suggest that the presence of endotoxaemia in the plasma is due to an impairment of more complex mechanism, as liver's detoxification, which seems the most important.

Percoll density gradient centrifugation. (An improved method for the enrichment of lymphocyte subsets mediating different activities).

Lymphocyte fractions obtained by Percoll density gradient centrifugation were assayed for their cytotoxic activities both in NK and ADCC cytotoxic systems. Furthermore, the release of a mediator, platelet slowing factor from the same subsets, was evaluated. Different mitogens (phytohemagglutinin, concanavalin A and lipid A) have been used for this purpose. The maximum of cytotoxic activities was recovered in E+ cell-enriched fractions. On the other hand, lipid A was able to stimulate the PSF production from enriched B cells (fraction I); phytohemagglutinin induced the release of PSF from E+ fractions (II and III) and concanavalin A from fraction II only. Our results suggest that Percoll density gradient is a very reproducible method in obtaining lymphocyte subsets enriched in cells mediating either cytotoxic activities or lymphokine release.
