[Effect of morphine, beta-endorphin, and met-enkephalin on electrical activity of Helix pomatia snail neurons].

Application of morphine and methionine-enkephaline using microionophoretic technique (20-100 nA, 1-15 sec) or perfusion (10(-7)--10(-6) M) caused inhibition of the impulsation, which in some of the cases was accompanied by hyperpolarization of the membrane and the decrease in its resistance, as well as by the reduction of EPSP's. The increase in impulsation with membrane depolarization was less frequent. beta-Endorphine (10(-7)--10(-6) M) caused a substantially longer inhibition of the impulse activity than morphine or met-enkephaline.

[Induction of a delayed cyclic AMP-sensitive sodium current in mollusk neurons by G-protein activators].

The experiments have been carried out with isolated Helix pomatia neurons by the method of two-microelectrode voltage clamping. cAMP and the activators of G-proteins, F-, GTP and Gpp (NH)p, were injected via ionophoresis into the neurons using a seven-channel microelectrode. It is found that the G-protein activators induce both input and output currents in the neurons where slow sodium current can be activated by cAMP. The input G-activated and cAMP-activated currents are similar in kinetics, voltage dependence and reverse voltage. The G- and cAMP-responses are not additive. The results obtained allow the conclusion that the G-protein activators can stimulate the slow cAMP-sensitive sodium channels.

[The role of cAMP in the electrophysiological effects of morphine and enkephalins].

The review considers the data on the morphine and enkephalins electrophysiological effects which were obtained in snail neurons and might be explained by a decrease in the level of cAMP in the cytoplasm. These effects are: a membrane hyperpolarization, a depression of the bursting activity, an inhibition of the excitatory serotonin and dopamine responses.

[The nootropic agent vinpocetine blocks the delayed rectifier potassium channel more strongly than the high-conductance calcium channel].

In isolated neurons of Helix snail high-threshold Ca(2+)- and K(+)-currents were recorded using two-microelectrode voltage clamp technique. Extracellular application of nootropic drug vinpocetine rapidly and reversibly blocked both types of current. Vinpocetine effects were dose-dependent and voltage-independent. The maximal effect of Ca(2+)-current blockade was 27% during application of vinpocetine in the concentration of 600 mcM. The maximal blockade of K(+)-current was 75% when the drug was applied in the concentration of 30 mcM. Results suggest that K(+)-channels are more probable targets for vinpocetine than Ca(2+)-channels.
