Structural importance of the amino-terminal residue of pancreatic phospholipase A2.

To study the structural importance of the NH2-terminal Ala1 residue of pancreatic phospholipase A2, several mutants were prepared by a stepwise semisynthetic approach. 13C NMR spectroscopy of 90%-enriched [[3-13C]Ala1] phospholipases A2 shows the pK values of the alpha-NH3+ groups of porcine enzyme, porcine isoenzyme, bovine enzyme, and equine enzyme to be 8.4, 8.8, 8.9, and 8.8, respectively. A group titrating with a pK of approximately 6.3, present only in the porcine and equine phospholipases A2, presumably originating from Glu71, disappears in the presence of 0.2 M Ca2+, while the pK values of their alpha-NH3+ groups shift to 9.3 and 9.0, respectively. No such effects were observed for the porcine isoenzyme and bovine enzyme, which lack an acidic side chain in position 71. It can thus be concluded that the equine phospholipase A2, like the porcine enzyme, possesses in addition to the catalytic Ca2+ binding site also a second, low-affinity Ca2+ binding site, which is not present in the porcine isophospholipase A2 and bovine phospholipase A2. From the titration behavior of the alpha-NH3+ group of equine and porcine [[3-13C]-Ala1]phospholipases A2 in the presence of micelles of n-hexadecylphosphocholine, it seems very likely that at alkaline pH the equine phospholipase A2, like the porcine enzyme, requires the second Ca2+ ion for optimal binding to neutral lipid-water interfaces. Semisynthetic porcine phospholipase A2 analogues in which the position of the alpha-NH3+ group is varied have lost their affinity toward neutral lipid-water interfaces and consequently their catalytic activity on micellar substrates. Most of these phospholipase A2 analogues retain, however, some of their enzymatic activity on monomeric substrate. Substitution of the side chain of Ala1 in porcine phospholipase A2 by hydrophobic side chains abolishes almost all activity due to the loss of affinity for neutral lipid-water interfaces. In contrast, NH2-terminal residues having more polar side chains affect only slightly phospholipase A2 activity. Compared to the native enzyme, these latter phospholipase A2 analogues show an increased penetration capacity for monolayers of 1,2-didecanoyl-sn-glycero-3-phosphocholine. Probably, the interaction of hydrophobic amino acid residues at the 1-position with other hydrophobic side chains present in their vicinity prevents the correct positioning of the alpha-NH3+ group, thereby leading to loss of catalytic activity.(ABSTRACT TRUNCATED AT 400 WORDS)

[Diethylstilbestrol in urine of slaughtered cattle].

The results of an investigation during 1981-1984 for the presence of diethylstilbestrol (DES) in 12576 samples of bovine urine in the Netherlands are presented. The screening was performed by radioimmunochemical detection after chromatographic purification on celite. Subsequent final confirmation was performed by combined high pressure liquid chromatography-gas chromatography mass spectrometry. On the average in 1.7% of the 12576 samples the presence of stilbenes , primarily DES, was proven. Probably due to the reintroduction of the meat inspection act regarding the control for DES, a substantial decrease to 0.2% (4 samples) was observed in the first three months of 1984. In these 4 samples out of 2333 samples the presence of dienestrol (DE) was confirmed. No DES or hexestrol (HEX) could be detected.

Identification of several stilbene derivatives in bovine urine by means of high performance liquid chromatographic fractionation and immunochemical detection.

Radioimmunochemical detection (RIA) following fractionation of urine extracts via high performance liquid chromatography (HPLC) turned out to be a very specific method for the identification of stilbene derivatives in bovine urine. Combination of the high resolution of the HPLC with a specific RIA is a suitable method to discriminate between the presence of different stilbene derivatives like diethylstilboestrol (DES), dienoestrol (DE) and hexoestrol (HEX) or other unknown compounds interfering in the celite-RIA used in the screening. Using this screening method 8200 samples of bovine urine were investigated on the presence of stilbene derivatives of which 133 were classified as 'positive'. In 106 'positive' urines the presence of DES was shown and in 19 'positive' urines the presence of DE or HEX, using the method described in this report whereas in 8 'positive' urines an unknown immunochemical active compound was detected. During 1.5 year of comparative investigation no qualitative discrepancies occurred between the results of the HPLC-immunogram procedure and the final confirmation by high resolution gas chromatography-mass spectometry (GCMS).

Monitoring and identification of residues of anabolic preparations in slaughtered cattle by HPLC with diode array detection.

The new combination of isocratic high performance liquid chromatography (HPLC) with on line UV spectrum detection via a diode array configuration has been applied to the detection and identification of anabolics present in application sites of cattle. Combination of the characteristic retention time in the HPLC chromatogram and a comparison of the full spectrum between 190-400 nm of the anabolic components with that of a standard resulted in a very reliable identification. By means of this method 117 samples of application sites were investigated for the presence of anabolic residues. Of the xenobiotic anabolics , 19-nortestosterone (NT) was found most frequently (in 96 cases), whereas diethylstilbestrol (DES) was found in only 11 cases. In all samples the identification of NT and DES was confirmed by high resolution gas chromatography-mass spectrometry (GCMS).

The use of affinity matrices in the purification of inhibin from bovine follicular fluid.

The protein associated with inhibin-like activity in bovine follicular fluid was purified 80- to 120-fold after successive adsorptions on different affinity matrices, i.e. Matrex gel red A, phenyl sepharose, omega-aminohexyl agarose and concanavalin-A sepharose. Partial characterization of the active protein resulted in the conclusion that inhibin from bovine follicular fluid is a hydrophobic glycoprotein with an apparent molecular weight between 60 000 and 70 000 daltons. An antiserum, raised against an 80-fold purified preparation, prevented the inhibin-like action of bovine follicular fluid on pituitary cells in vitro.

Methylation of histidine-48 in pancreatic phospholipase A2. Role of histidine and calcium ion in the catalytic mechanism.

It is known that His-48 is part of the active center in pancreatic phospholipase. To further elucidate the role of histidine-48 in the active center of pancreatic phospholipase A2, we have modified the enzyme with a number of bromo ketones and methyl benzenesulfonates. Rapid methylation occurred with methyl p-nitrobenzenesulfonate. Methylated phospholipase shows total loss of enzymatic activity whereas binding of substrate and the cofactor Ca2+ remains intact. Amino acid analysis of methylated equine phospholipase showed the loss of the single molecule of histidine and the formation of one molecule of 2-amino-3-(1-methyl-5-imidazolyl)propanoic acid (1-methylhistidine). Equine phospholipase was also modified by [13C]methyl p-nitrobenzenesulfonate and the methylated enzyme was studied by 13C NMR. The results indicate that the proton on the nitrogen in position 3 of the imidazole ring is involved in a strong interaction with a buried carboxylate group, thereby hindering rotation of the imidazole ring, and that the nitrogen in position 1 is involved in catalysis. These data are in full agreement with the three-dimensional structure at 1.7-A resolution of bovine pancreatic phospholipase. A catalytic mechanism is proposed in which a water molecule which is close to the nitrogen at position 1 of the imidazole ring of the Asp-99-His-48 couple acts as the nucleophile. A comparison is made between phospholipase A2 and the serine esterases.

Proton-nuclear-magnetic-resonance/pH-titration studies of the histidines of pancreatic phospholipase A2.

The study by means of 1H nuclear magnetic resonance (NMR) of the histidines of phospholipase A2 isolated from porcine, bovine and equine pancreas is reported. Assignment of the histidine resonances was achieved by comparison of different enzymes and the use of paramagnetic probes. pH titration curves for various histidyl resonances were obtained and compared in the presence and absence of calcium. Calcium is shown to lower the pKa of the active site histidine. The NMR results are compared with the known X-ray three-dimensional structure for the bovine enzyme.

A photochemically induced dynamic nuclear polarization study of pancreatic phospholipase A2. NMR assignment of some aromatic residues.

Application of the photochemically induced dynamic nuclear polarization technique to porcine pancreatic phospholipase A2 and its precursor resulted in the observation of nuclear spin polarization for the single tryptophan and 2 tyrosine residues. Using selectively nitrated enzymes, the tyrosine resonances could be assigned to residues 69 and 123. Both tryptophan-3 and tyrosine-69 are of particular interest because they are part of the lipid binding site.

A sensitive CCD image system for detection of chemiluminescent reactions.

A new ultra-sensitive light detection system is described for the detection of chemiluminescence reactions. The system consists of a CCD camera equipped with a two-stage image intensifier coupled to an image analysis system with real-time filtering capabilities. The sensitivity was determined with the enhanced chemiluminescence system using horseradish peroxidase as label. Additional possibilities for further increase in sensitivity and the potential use in immunoassays and other applications are discussed.

A chemiluminescent immunoassay for 17 alpha-methyltestosterone.

An immunoassay for the xenobiotic anabolic compound methyltestosterone (MT) is presented. The detection is based on chemiluminescence of a MT-isoluminol conjugate. Applications are presented for detection of methyltestosterone and cross-reacting compounds in bovine urine and tissues of application sites isolated from slaughtered cattle, illegally treated with hormonal anabolics.

A highly specific detection method for diethylstilbestrol in bovine urine by radioimmunoassay following high performance liquid chromatography.

A specific radioimmunoassay (RIA) for diethylstilbestrol (DES) is presented following purification of sample extracts by isocratic reversed phase high performance liquid chromatography. Applications are given of a forensic investigation for the control of DES in bovine urine. Specificity of HPLC-RIA is compared with that of RIA's with other (chromatographic) purification procedures and with gas chromatography-mass spectrometry. Implications of the use of these techniques in practice and the use of various DES-antisera are discussed.

Rheumatoid vasculitis presenting as appendicitis.

Appendicitis was diagnosed in a 38-year-old patient with seropositive rheumatoid arthritis. Despite appendectomy the patient's clinical condition deteriorated. Thorough microscopical evaluation of the pathological anatomical specimens from the appendix made possible a diagnosis of necrotizing vasculitis. The systemic nature of the vasculitis was confirmed with a muscle biopsy. A complete remission was induced with immunosuppressive treatment. The case exemplifies a rare but serious manifestation of rheumatoid vasculitis.
