A rapid method of staining ultrathin sections for surgical pathology TEM with the use of the microwave oven.

A rapid microwave method is described for staining ultrathin sections for surgical pathology transmission electron microscopy (TEM). Three sets of Epon sections of human heart biopsy were mounted on unsupported 200-mesh Rhodium-coated copper grids and were stained with uranyl acetate (UA) and lead citrate. The first set of grids was stained for 15 seconds in each solution with the aid of a microwave oven, and the second set was stained routinely for 30 minutes in UA and 10 minutes in lead citrate at room temperature. The third control set was stained for 15 seconds in each solution without microwave bombardment. The overall image quality of the TEM micrographs generated by the "quick-stained" microwave enhanced sections was better than routine stained and control sections. The microwave-treated sections have more contrast, less artifacts in the form of precipitate, and a more uniform overall staining.

Comparative stereology of the lizard and frog myocardium.

The atria and ventricles of the frog and lizard were quantitated using stereologic techniques. The volume fraction (Vv) and surface density (Sv) of the free, junctional and total sarcoplasmic reticulum and mitochondria of the lizard atrium and ventricle were greater than in the corresponding chambers in the frog. Myofibrillar volume fraction and plasmalemmal surface density did not differ between the two species. The volume fraction and surface density of the free and total SR, and myocardial granules were greater in the lizard atrium than ventricle but the myofibrillar Vv and mitochondrial Vv and Sv were less. The Sv of the free SR, total SR, and the Vv and Sv of myocardial granules of the frog atrium were greater than in the frog ventricle. There were no differences between myofibrils and mitochondria in the frog atrium and ventricle.

Carcinoma of the larynx and hypopharynx: computed tomographic-histopathologic correlations.

Twenty consecutive patients underwent computed tomography (CT) prior to total laryngectomy in order to assess the accuracy of CT scanning in the evaluation of carcinoma of the larynx and hypopharynx. CT scans were prospectively interpreted and the extent of tumor was recorded graphically at five major levels of the larynx. The extent of tumor was similarly evaluated on whole-mount histologic sections of the laryngeal specimens prepared in the horizontal plane similar to the CT scans. The results were compared to assess the ability of CT to identify tumor extent accurately. Despite good correlation of gross tumor extent between histologic specimens and CT scanning, specific pitfalls in CT diagnosis were identified. Overestimation of tumor extent was caused by edematous changes in six patients and tumor-associated inflammatory changes in three patients. In seven patients, mass effect from adjacent bulky tumor significantly distorted normal structures, mimicking tumor involvement. Small foci of mucosal tumor in three patients and microscopic cartilage involvement in two patients were not identified on CT scanning, causing underestimation of tumor extent.

A fixative for use in muscle histochemistry.

A fixative solution that preserves the activity of some relevant enzymes in muscle histochemistry is described. Portions of human muscle biopsy specimens and selected murine muscles were fresh frozen or placed in the fixative at room temperature for up to 1 month before freezing. Cryostat sections of fresh frozen and fixed frozen tissue were assayed for nicotinamide adenine dinucleotide phosphate (NADH)-tetrazolium reductase (NADH), several adenosine triphosphatases (ATPases), myoadenylate deaminase (MD), and phosphorylase. NADH, ATPase, and MD activity were preserved following fixation but phosphorylase was not preserved. Murine spleen and kidney were similarly tested for acid phosphatase (acid phos), alkaline phosphatase (alk phos), and nonspecific esterase (NSE). Alk phos activity was preserved but acid phos and NSE activity were significantly reduced following fixation. This fixative is useful in some circumstances for processing or shipping human muscle biopsy specimens and experimental tissues.

Antigens of human pancreatic adenocarcinoma cells defined by murine monoclonal antibodies.

We have elicited and characterized the serological specificity of five murine monoclonal antibodies (DU-PAN-1, 2, 3, 4, and 5) to a human pancreatic tumor cell line, HPAF. The antibodies are not detecting HLA-associated antigens since all of the monoclonals failed to react with human lymphoid and myeloid cell lines and uncultured cells. All of the monoclonals except DU-PAN-5 reacted with four of five pancreatic tumor cell lines and two of two uncultured pancreatic tumors. An immunoperoxidase technique was used to determine the presence of the antigens detected by the monoclonal antibodies in frozen sections of tumor and adult and fetal normal tissues. DU-PAN-1 antigen was detected on pancreatic tumors, and a transitional cell carcinoma of the bladder, but was detected on no other adult or fetal normal tissues including pancreas. One of the antigens (DU-PAN-2) defined by the monoclonals was present on pancreatic ductal epithelial cells and showed a restricted distribution on tumor cells from some other carcinoma patients and on cells from certain fetal tissues. DU-PAN-3 antigen was present on adult and fetal pancreatic cells and certain tumor cells but could not be detected on cells of other fetal or adult normal tissues. DU-PAN-4 and DU-PAN-5 antigens have a more widespread distribution on normal or tumor cell types.

Comparative stereology of mouse atria.

The left and right atria of the mouse were compared to each other and to the mouse left ventricle using stereologic techniques. The volume fraction (Vv) and surface area per unit cell volume (Sv) of the interior junctional sarcoplasmic reticulum (IJSR), total JSR and extended JSR were greater in the left atrium than in right. The Vv and Sv of the free SR, transverse tubules, and mitochondria were similar in the two atria. It is suggested that the differences in junctional sarcoplasmic reticulum between the atria can be accounted for by a difference in distribution of two types of cells whose anatomy is analogous to working and conducting fibers in the ventricle. The Sv and Vv of the transverse tubules, mitochondria, and all the components of the sarcoplasmic reticulum except for the free SR were greater in the left ventricle than in either atrium. The greater calcium content and sensitivity to extracellular calcium of the atria may explain the greater volume of free SR in the atria as compared to the left ventricle. The Sv of the plasmalemma of the atria and of the Sv of the plasmalemma of the transverse tubules of the left ventricles supports the suggestion of others that there is a constant ratio of surface area to cell volume in cardiac cells.

Facilitated light microscopic cytochemical diagnosis of acute myelogenous leukemia.

Hydroperoxidase-positive Phi bodies and rods are much more prominent and prevalent than rods visualized with a Romanovsky-type stain (Auer rods) in immature leukocytes of patients with active acute myelogenous leukemia (AML). They are readily observed with the light microscope in peripheral blood or marrow films of AML patients stained to show their peroxidatic activity. In many of these patients, Auer rods, which apparently constitute only a small subpopulation of the hydroperoxidase-positive Phi bodies and rods, were detected with difficulty, if at all. The hydroperoxidase-positive Phi bodies and rods were observed in 92% of 36 patients with active disease. They were never observed in leukocytes of patients with other hematopoietic disorders or of normal individuals. Thus, they facilitated the distinction of AML from acute lymphocytic leukemia and chronic granulocytic leukemia in blast crisis. They were absent in full clinical remission after chemotherapy and were greatly diminished in partial remission. They were present in disease relapse and reappeared in five patients who had been in full remission. These results suggest that these hydroperoxidase-positive enlarged particles are pathognomonic of AML and that monitoring them with the light microscope may aid in guiding its clinical management.

Ceruminous gland adenocarcinoma: a light and electron microscopic study.

Tumors of ceruminous gland origin in the external auditory canal are rare in man. A case is described in which such a tumor presented as an invasive vascular temporal bone neoplasm, mimicking a glomus jugulare tumor. Light and electron microscopic study of this tumor confirmed a diagnosis of ceruminous gland adenocarcinoma. Tumors of ceruminous gland origin appear to have a distinctive clinical behavior by virtue of their unique anatomical location in the external auditory canal. Our experience with this case establishes another clinical picture characterizing the histologic type of ceruminoma designated as a ceruminous gland adenocarcinoma. We feel that the generic term "ceruminoma," with its implied histologic subgroups, is useful to the clinician when he encounters a tumor arising from the modified sweat glands of the external auditory canal.

Comparative stereology of the mouse and finch left ventricle.

The volume fractions and surface per unit cell volume of some subcellular components of the left ventricles of the finch and mouse were quantitated by stereologic techniques. These species were chosen for study because they have similar heart rates but differ morphologically in some respects: fiber diameter is larger in the mouse; the mouse has transverse tubules while the finch does not; and the finch has a form of junctional sarcoplasmic reticulum (JSR), extended JSR (EJSR), located in the cell interior with no direct plasmalemmal contact, while the mouse interior JSR (IJSR) abuts on transverse tubules. Our data show that the volume fraction (Vv) and surface area per unit cell volume (Sv) of total SR, and free SR (FSR) are similar. The volume fractions of mitochondria, myofibrils, and total junctional SR were also similar. The Sv of the cell surface of the finch was similar to the Sv of the cell surface of the mouse (Sv-plasmalemma plus Sv of the transverse tubules). The principal difference was in the distribution of JSR; the mouse peripheral JSR (PJSR) represents only 9% of the total JSR, while the finch PJSR accounts for 24% of the bird's JSR. The similar volume fractions of total junctional SR (PJSR + EJSR in the finch; PJSR + IJSR in the mouse) suggest that the EJSR is not an embryologic remnant, and raises the possibility that some function of JSR is independent of plasmalemmal contact.

The steroid profile of a virilizing ovarian tumor.

A case report of a 25-year-old female with a sex cord stromal virilizing ovarian tumor is presented. The pathway of ovarian steroid secretion in this tumor is elucidated with the dominant elements being pregnenolone, 17-hydroxypregnenolone, 17-hydroxyprogesterone, androstenedione, and testosterone. The tumor primarily made testosterone (T) with lesser elevations of androstenedione (A), dehydroepiandrosterone (DHEA), and dihydrotestosterone (DHT). Expert pathologic opinions differed whether this neoplasm was a Sertoli-Leydig tumor or a virilizing granulosa tumor; therefore, it was probably a gynandroblastoma. A unilateral salpingo-oophorectomy was performed and the patient promptly resumed normal ovarian function with ovulation.

Peripheral couplings in human skeletal muscle.

The importance of peripheral couplings in muscle function has been uncertain because of the failure to demonstrate them in human skeletal muscle. This study has utilized ruthenium red staining to demonstrate peripheral couplings in human skeletal muscle biopsies. This finding supports the belief that the peripheral coupling is a basic component of striated muscle.

Benign vaginal rhabdomyoma: a light and electron microscopic study.

A benign vaginal rhabdomyoma from a 49-year-old woman was examined by light and electron microscopy. While many of the ultrastructural features conformed to those noted in previously described rhabdomyomas, certain features are described for the first time. These include attachment plaques between cells enclosed within a common basement membrane, cytoplasmic bodies, and peripheral couplings. Finally, the literature is reviewed and the reported cases of rhabdomyoma are divided into adult and fetal types based upon light microscopic criteria.
