[Kinetics and clinical application of 99mTc-technegas].

Biological characteristics and clinical utilization of 99mTc-technegas were evaluated in three normal volunteers and 10 patients with various pulmonary diseases. 99mTc-pertechnetate (296 MBq (8 mCi] was injected in the crusible of 99mTc-technegas generator, and about 37 MBq (1 mCi) of 99mTc-technegas was inhaled in the lung by three times of deep inspiration. Blood activity was appeared immediately after inhalation of 99mTc-technegas and it was 0.2% of the inhaled dose/liter of blood after 2 hours. Urine activity was also increased after inhalation and cumulative dose was 4.96% of the inhaled dose after 24 hours. The tracer dose to distribution to the lung was about 95% of the total dose in whole body. Biological half time of 99mTc-technegas in the lung was 135 hours and pulmonary image was stable even after 24 hours. Radiation dose to the lung was 0.004 Gy/37 MBq (1 mCi). In patients with pulmonary disease, distribution of 99mTc-technegas was similar to that of 81mKr gas, although defect on 99mTc-technegas image was larger in severe obstructive disease. 99mTc-technegas study was superior over 133Xe gas study to identify the areas of ventilation abnormality more clearly, although quantitative evaluation was difficult in 99mTc-technegas study. On comparative study with 99mTc-aerosol, 99mTc-technegas distributed in more peripheral sites and both methods were cooperative for evaluation of ventilatory disturbance. These results suggest that 99mTc-technegas inhalation study is safe and useful method for ventilation study.

[A comparative study of myocardial scintigraphy with 99mTc-SQ-30,217 and 201Tl in cases with myocardial infarction].

A comparative study of myocardial scintigraphy with 99mTc-SQ-30217 (SQ) and 201Tl was performed in 6 patients having ischemic heart diseases which were confirmed by the coronary angiography. The SQ study was done 1 week after the 201Tl study. Both studies were performed in exercise and resting state. Degree of perfusion defect was compared between SQ and 201Tl SPECT by means of scoring 0-2 (0: no defect, 1: equivocal, 2: definite defect) by five doctors (two physicians and three radiologists). Ability of this agent to detect the ischemic lesions was similar to that of 201Tl except for two regions. Effect of the liver image on the evaluation of the inferior wall of the left ventricle was small on this study, although the hepatic accumulation of the tracer was prominent. This agent is a promising tracer for the evaluation of myocardial perfusion in the cases with myocardial infarction.

The maximum and minimum water content and cell volume of human erythrocytes in vitro.

The maximum and minimum water contents of human erythrocytes were measured after exposure to various osmotic pressures. Within a range of osmolarities, at which no haemolysis occurred, the water content reached its maximum, 78.1%, at 180 mosM and its minimum, 54.8%, at 800 mosM. Simultaneously, the mean cell volume increased to 98.5 microns 3 at 180 mosM and decreased to 77.2 microns 3 at 800 mosM.

Cloning of the cellulase gene from Ruminococcus albus and its expression in Escherichia coli.

The gene for cellulase from Ruminococcus albus F-40 was cloned in Escherichia coli HB101 with pBR322. A 3.4-kilobase-pair HindIII fragment encoding cellulase hybridized with the chromosomal DNA of R. albus. The Ouchterlony double-fusion test gave a single precipitation line between the cloned enzyme and the cellulase from R. albus. The size of the cloned fragment was reduced by using HindIII and EcoRI. The resulting active fragment had a size of 1.9 kilobase pairs; and the restriction sites EcoRI, BamHI, PvuII, EcoRI, PvuII, and HindIII, in that order, were ligated into pUC19 at the EcoRI and HindIII sites (pURA1). Cellulase production by E. coli JM103(pURA1) in Luria-Bertani broth was remarkably enhanced, up to approximately 80 times, by controlling the pH at 6.5 and by reducing the concentration of NaCl in the broth to 80 mM.

Trimethoprim interference in methotrexate assay by an enzyme inhibition assay kit.

Spurious methotrexate (MTX) concentrations in sera and cerebrospinal fluids from leukemia patients who were given trimethoprim (TMP) were estimated using an MTX assay kit which is based on its inhibition of dihydrofolate reductase. The interference of TMP with MTX was confirmed in the assay system. A concentration of 0.17 microgram/ml of TMP gave a value for an apparent MTX of 10 microM.

Cross-reactivity between cowpox and vaccinia viruses with monoclonal antibodies. Brief report.

Hybridoma cell lines producing monoclonal antibodies against cowpox virus (CPV) and vaccinia virus (VV) were established to examine the specific and cross-reactive antigenic determinants of these viruses. Monoclonal antibodies against CPV (LB red strain) and VV (Lister strain and Ikeda strain) were classified into several groups on the basis of the results of immunofluorescence and haemagglutination inhibition tests. It was suggested that the groups defined above include the group of antibodies reacting with each of known major antigens of poxvirus, i.e. nucleoprotein (NP) antigen, heat labile and stable complex (LS) antigen, haemagglutinin (HA), cell surface antigen (CSA) and type A inclusion body (A).

Cation imbalance in erythrocytes, serum and 24-hour urine from patients with essential hypertension and adolescents with high blood pressure.

Electrolyte concentrations in the erythrocytes, serum and 24-hr urine were measured in 34 untreated essential hypertensives (EH), 32 normotensives with a family history of hypertension (N+), 112 normotensives without a family history of hypertension (N-), 76 senior high school students with high blood pressure (H) and 110 students with normal blood pressure (C). For the measurement of intra-erythrocyte electrolytes, a new method to decrease the trapped plasma was developed and adopted. A new simple method to collect 24-hr urine was devised, too. The results were as follows: 1) The sodium concentration [Na] and the ratio of [Na] to potassium concentration [Na/K] in the erythrocytes were significantly higher in the EH and N+ than in the group of N- (p less than 0.01). 2) There was a positive correlation (r = 0.43) between the [Na/K] in the serum and erythrocytes in the 3 groups. The [Na/K] value in the erythrocytes to a given [Na/K] value in the serum was higher in EH than in the group of N-. 3) When the group of H was compared with C, the Na excretion of the 24-hr urine in the former group was found to be slightly higher, and the [Na/K] of the 24-hr urine and the [Na] and [Na/K] values of the erythrocytes were significantly higher, too. It may therefore be said that an increase in the intracellular [Na] or [Na/K], which may be caused genetically, and an excess of Na intake and an increase of the ratio of Na/K intake are presumed to play an important role in the pathogenesis of essential hypertension.

[Clinical application of extracellular fluid measurement using 35S-sodium sulfate (author's transl)].

ECF (extracellular fluid volume) was measured by 35S-sodium sulfate and the same time value of PV (plasma volume) was determined by 131I-RISA in each subject. In normal ECF was an average of 181 ml/kg, 6.21 l/m2 and PV was 49.1 ml/kg, 1.69 l/m2. Both values of ECF and PV in essential hypertension were almost the same as those in normal. In congestive heart failure ECF was 232 ml/kg, 7.51 l/m2 and PV was 55.8 ml/kg, 1.80 l/m2. In the patients with ascites or edema, ECF was 256 ml/kg, 8.69 l/m2 and PV was 53.0 ml/kg, 1.79 l/m2. The results of the measurement by the two compounds also corresponded to the conditions of the patients who showed electrolytes disorders (ACTH deficiency, SIADH) and was improved by treatment.
