Immunogenetics of childhood celiac disease: the association with HDA DR3 and DR7 in unrelated patients with multiply affected families.

The association between childhood celiac disease and the histocompatibility complex was studied in five multiply affected families and 20 unrelated patients. The diagnosis of celiac disease (CD) was established by three consecutive jejunal biopsies: 1) at the initiation of the diagnosis; 2) following a gluten-free diet; and 3) after gluten provocation. The results of this study indicate that a significant association exists between celiac disease, DR3 (P less than 0.001) and DR7 (P less than 0.001). The relative risks for these antigens were 8.56 and 5.05, respectively. Segregation analysis in the families suggested that the susceptibility genes associated with the histocompatibility antigens are not inherited in either a dominant or a recessive pattern. While an intermediate type fits better into the genetic framework of CD, more than one susceptibility and protecting gene may be involved in the pathogenesis of this disease.

Alloantigens of the human T (HT) genetic region of the HLA linkage group.

This article describes an approach that characterizes alloantisera reactive with new determinants on mitogen-activated human lymphocytes. These markers, which are tentatively called HT-antigens, are different from the classical HLA-A,C,C, and DR antigens but are associated with beta 2-microglobulin. Several alloantisera were tested extensively on lymphocytes that were treated initially with an antigen-blocking technique using turkey anti-beta 2-microglobulin and turkey anti-p29,34 sera or with an antigen-modulation technique using xeno- and alloantisera. It was concluded that these tested antisera recognized three different determinants (HT1, HT3, and HT (1 + 3) that appeared to segregate with the HLA-A locus of the HLA haplotype. HT antigens may be analogous to the murine TL-Qa antigens since HT antigens are associated with beta 2-microglobulin, are generally related to the HLA region, and are expressed on mitogen activated T cells but not on resting T cells.

Occlusal and orthodontic considerations in the periodontally involved dentition.

In the formulation of a treatment plan for patients with advanced periodontal disease, we must consider the benefits that orthodontics can supply. By improving the position of malposed teeth, we create a better environment for the health of the gingivae and attachment apparatus, as well as the possibility of improving osseous defects as the result of orthodontically stimulated osteogenic activity. The ultimate goal of treatment must be, in addition to control of inflammation, plaque and trauma, the creation of optimal morphologic and functional conditions which will allow these objectives to be obtained. This will provide the prosthodontist and periodontist a better chance to prolong the life of the dentition.

Platelet HLA typing and cross-matching by a direct microcytotoxic assay. A new method employing carboxyfluoresceindiacetate-stained platelets.

Carboxyfluoresceindiacetate-labelled platelets can be used in direct microcytotoxic assays for the purpose of typing platelets for a number of antigenic determinants, such as HLA and beta-2 microglobulin. The assay when used in direct platelet cross-matching, where thrombocytopenic patient sera were screened, resulted in the uncovering of many extrapositive and weakly positive cross-matches not identified by the lymphocyte cross-match. The simplicity and rapidity of this new method makes it an attractive alternative for future use in research and clinical studies.

Vitiligo and the HLA system.

77 Vitiligo patients were investigated for their HLA phenotype. Analysis of this material according to age of onset and ethnic origin revealed some significant characteristics. Young Jewish Moroccan patients showed a high frequency of B13 when compared to matching controls. Young patients of Yemenite origin had a strikingly high frequency of BW35. Jewish Yemenites showed a lack of B13 both in patients and in controls. It is suggested that studies by undertaken to clarify this problem further.

HLA antigens in ulcerative colitis and Crohn's disease in Israel.

The frequency of HLA antigens was studied in 60 Israeli Jewish patients with ulcerative colitis (UC) and in 18 with Crohn's disease (CD), and it was compared to that in healthy, ethnically-matched controls. From this study it would appear that HLA-BW 35 is highly associated with UC in Israeli Jewish patients and that HLA-AW 24 is present at a high frequency in the "early onset" group and also in the group with "severe" disease, which was composed mainly of female patients. In the CD group, the frequency of the HLA antigens did not differ significantly from that in the control group.

A newly designed whole microplate automatic harvester for lymphocyte stimulation assays.

A unique automated sampling manifold designed to recover cells grown in standard 96 well microplates from their culture medium is described. Cells are recovered and washed on fiber glass filter discs. Incorporation of radioisotopes into cells, can then be measured by appropriate counting of the filter discs. Typical applications include termination of mixed lymphocyte cultures, assays of mitogen stimulation of lymphocytes and antigen-specific lymphocyte transformation and assays of interferon activity. The harvester can also be used in other biological systems where collection and washing of precipitates is desired.

HLA-A and HLA-B antigens in transitional cell carcinoma of the bladder.

A possible association between HLA-A and HLA-B loci alleles and bladder carcinoma has been investigated in 44 patients with biopsy proved transitional cell carcinoma of the bladder. The patients were compared to 400 healthy controls and matched for ethnic origin. Typing was done by the standard National Institutes of Health microlymphocytotoxicity technique. Comparison between the HLA-A and HLA-B series of the patients and the controls, and the different stages of disease did not yield any significant difference.

HLA system in relation to carbohydrate metabolism in lichen planus.

HLA antigens were determined in a group of fifty-two patients with lichen planus. In the group as a whole the frequency of A28 was increased but not significantly so. When the patients were subdivided according to the results of an oral glucose tolerance test, however, a highly significant increase in the frequency of A28 was found among the non-diabetic patients.

HLA, DRW (IA-like) and additional surface antigens expressed on adenoid T and B lymphocytes.

Pregnancy sera were found to react differently when screened simultaneously with adenoid and peripheral blood lymphocytes (PBL) obtained from the same individual. Some of the reactions were attributed to the higher sensitivity of adenoid lymphocytes to a low titer HLA or Ia antibody, while others were shown to be due to the expression of DRw (Ia-like) antigens on both T and B lymphocytes separated from adenoids. Positive reactions specific to adenoid lymphocytes, or manifested only following anti-beta2-microglobulin (a-beta2-m) treatment, suggest the presence of antigens other than HLA or Ia on the cell membrane. Unmasking of antigenic sites by the a-beta2-m capping process and rearrangement of the membrane structure may also contribute to the observed phenomena.

HLA antigens in genetic neutropenia of Yemenite Jews.

The HLA antigenic system was studied in Yemenite Jews with genetic neutropenia. No deviation in antigen frequency was observed when compared with matched controls from the same ethnic group. HLA antigen frequency and common haplotypes of the Yemenite Jews were found entirely different from those of the African blacks, known to have the same anomaly.

Study of the HLA antigen in Arab children with paralytic poliomyelitis.

In 1974 and 1976, two outbreaks of paralytic poliomyelitis occurred in a vaccine-protected infant population in the Gaza Strip. To test the possibility of an increased susceptibility to the disease, the frequency of HLA antigens was studied in 58 of the affected children and compared with 113 control subjects. HLA-AW19 and -B7 were found more frequently in the affected children. These differences were not statistically significant but are consistent with the possibility that patients with the paralytic disease may have an HLA genetic makeup different from that of the rest of the population.
