Oxiracetam pharmacokinetics following single and multiple dose administration in the elderly.

The kinetics of the nootropic drug oxiracetam was studied in 6 elderly female patients (age range 69-96 years) in good physical condition. After a single oral dose (800 mg), serum oxiracetam levels reached a peak (25 +/- 6 micrograms/ml) within 1-3 h and declined thereafter with a half-life of 3 to 6 h. Eighty-four percent of the administered dose was recovered in urine as unchanged drug within 24 h. During a maintenance regimen (800 b.i.d. at 8 a.m. and 2 p.m.) for 7 days oxiracetam did not show any accumulation in serum, trough serum levels before the morning and afternoon doses being 4.6 +/- 2.3 and 17.0 +/- 8.2 micrograms/ml respectively on day 4 and 5.7 +/- 3.3 and 17.9 +/- 7.7 micrograms/ml respectively on day 7. Comparison with previous pharmacokinetic data obtained in young subjects suggests that the clearance of the drug is lower in the elderly, probably as a result of the physiological decrease in renal function in old age.

Blood kinetics of sulfamonomethoxine and oxytetracycline following intrauterine spray injection in dairy cows.

Intrauterine administration was performed in six Friesan cows with a disposable spray preparation containing 3 g of sulfamonomethoxine and 3 g of oxytetracycline, in order to investigate their serum kinetics. Sulfamonomethoxine levels were determined by a reversed-phase HPLC method, whilst oxytetracycline quantities were detected by a microbiological method. The sulphonamide had a peak 1.17 h after the administration, the tetracycline reached its highest concentration after 8 h. The bioavailability of both drugs was low and detectable drug amounts were no longer recovered after 24 h.

Productive infection of HUVEC by HHV-8 is associated with changes compatible with angiogenic transformations.

Kaposi's Sarcoma (KS) is an angioproliferative disease associated with human herpesvirus 8 (HHV-8) infection. We have characterized the morphologic and phenotypic modifications of HUVEC in a model of productive HHV-8 infection. HHV-8 replication was associated with ultra-structural changes, flattened soma and a loss of marginal folds and intercellular contacts, and morphologic features, spindle cell conversion and cordon-like structures formation. Phenotypic changes observed on cordon-like structures included partial loss and redistribution of CD31/PECAM-1 and VE-cadherin, uPAR up-regulation and de novo expression of CD13/APN. Such changes demonstrate the induction, in HUVEC, of an angiogenic profile. Most of these findings are directly linked to HHV-8-encoded proteins expression, suggesting that HHV-8 itself may participate to the initial steps of the angiogenic transformation in KS.

Rapid and sensitive detection of recombinant soluble proteins in the supernatant of transfected mammalian cells.

Cloning and expression of recombinant soluble proteins could be quite a difficult task, especially when it comes to reliably detect minute amounts of the soluble protein in the supernatant of transfected mammalian cells. Timing and sensitivity are of the essence in order to optimise the benefits/costs balance and to decide which clones to grow further and which ones to discard. Here we propose a modified inhibition assay. The key feature of this approach is the development of a sensitive and quantitative test to detect the presence of the recombinant soluble protein by exploiting its ability to compete with the binding of a specific monoclonal antibody to a target cell. The described procedure is a sensitive, efficient, dependable and low cost method.

Age-related changes in interleukin 2 responsiveness of resting and activated human mononuclear cells.

BACKGROUND: Although the "T cell model" on the immunological decline associated with aging is gaining increasing support, the relationship between response to IL2 and aging has not yet been investigated. METHODS: Human peripheral blood mononuclear cells (PBMC) from healthy elderly donors were cultured with increasing concentrations (1-1000 U/ml) of recombinant interleukin 2 (rIL2). RESULTS: PBMC from older donors proliferated as much as those from younger donors until the fifth day of culture, but showed a reduced capability to proliferate in the following days (8th and 10th). Culturing PBMC with PHA we obtained the opposite result. In fact, the known depressed proliferative response of elderly people to the mitogen PHA is detectable only in the early days of culture. Furthermore, the response to rIL2 of 1% or 0.125% PHA-stimulated PBMC showed an important impairment of the proliferative rate in the elderly population with respect to younger controls, although no differences in the number of IL2 receptor-positive cells were detected. This phenomenon was demonstrable mainly in the first several days of culture. CONCLUSIONS: These data suggest a delay in the activation rather than a real functional impairment of PBMC from old donors.

Interleukin 2 and mitogen driven proliferation in human peripheral mononuclear cells: a kinetic analysis.

The role of the T lymphocyte growth factor, interleukin 2 (IL2), on human lymphocyte proliferation was investigated. The results define the extent and the chronological sequence of IL2 effects on unstimulated mononuclear cells and on an activated population enriched in IL2 receptor positive cells. Important differences were observed in the proliferative response to the mitogen PHA when the IL2 concentration in the culture medium was varied. The data suggest that IL2 concentration is critical in the responsiveness to a mitogenic stimulus.

[Gastric emphysema associated with gastric volvulus].

The gastric volvulus is an infrequent entity requiring surgical treatment in both the acute and chronic cases. The case of an 81-year old female patient attended for an episode of gastric obstruction caused by a volvulus and whose simple stomach radiography showed an image of gastric emphysema is reported. The possible causes of gastric emphysema and its differential diagnosis with emphysematous gastritis are discussed.
