Properties of cell lines established from transitional cell cancers of the human urinary tract.

Characterization studies have been carried out on eight cell lines (253J, 192B, 639V, 647V, 486P, 575A,743E, and 751G) established from transitional cell cancers of the human urinary tract. Although subtle morphological differences exist among individual lines, each has an epithelial morphology and exhibits multilayering. The doubling times for the cells range from 20 to 56 hr, and at least a 1-to-3 split can be achieved when they are subcultured every 4th day. Karyotypic analysis revealed a hyperdiploid stemline for each cell line, and presence of a Y chromosome was confirmed by Q banding in five of the lines. The tumorigenic nature of the cell lines was demonstrated by their production of tumors in hamsters and confirmed by colony formation in agar. The transitional cell cancer lines were shown to be free of Mycoplasma, and their glucose-6-phosphate dehydrogenase mobility patterns and their Karyotypes prove that they are not HeLa cells.

In vitro cultivation of human renal cell cancer. I. Establishment of cells in culture.

A technique for initiating and propagating epithelial cell cultures of human renal cell cancer and adjacent nontumor kidney is described. Seventy-five percent of the tumors and 79% of the adjacent kidney specimens cultured with this method have shown initial outgrowth and have been subcultured at least once. Two renal cell cancer cultures initiated by this method have now been in continuous culture more than 6 months, have been subcultured 27 and 18 times, and now appear to be stable lines. The ability to establish long-term in vitro cultures of human renal cell cancers will facilitate studies concerning the immunoreactivity, cholesterol metabolism, the isolation of renal-cell-cancer-specific antigens, and in vitro chemotherapy testing and will further our understanding of the basic biology of human renal cell cancer.

Pelvic exenteration: complications of urinary diversion.

We report complications directly related to urinary diversion among 32 patients who underwent pelvic exenteration for gynecologic malignancy. Eight patients (25 per cent) sustained 10 major complications, including 8 urinary fistulas and 2 fistulas from the entero-enterostomy sites. The frequency of these complications was greater among patients who had had total exenteration than among those who had had anterior exenteration. We attempt to identify factors responsible for these complications and discuss methods for reducing the morbidity related to urinary diversion in these patients.

Suppression of antitumor lymphocyte mediated cytotoxicity by human chorionic gonadotropins.

The effect of human chorionic gonadotropin on the in vitro cytotoxicity of lymphocytes against human transitional cell carcinoma cells was studied in 9 patients using the microcytotoxicity assay. Lymphocytes from 6 patients were cytotoxic against transitional cell carcinoma cells. However, in the presence of human chorionic gonadotropin the cytotoxicity of these lymphocytes against transitional carcinoma cells was diminished. Human chorionic gonadotropin alone or in combination with non-cytotoxic lymphocytes did not change the growth of target cells and human chorionic gonadotropin did not have a direct toxic effect on lymphocytes alone. These results demonstrate that human chorionic gonadotropin can inhibit the cytotoxic action of lymphocytes against human tumor cells in vitro. Human chorionic gonadotropin may contribute to the poor prognosis associated with certain human chorionic gonadotropin-secreting human tumors.

Changes in cell-mediated cytotoxicity during the clinical course of patients with bladder carcinoma.

The conventional cell-mediated cytotoxicity assay was modified so that more useful estimates of cell-mediated immunity to transitional cell carcimoma of the urinary tract could be obtained. With this modified assay changes in the levels of cell-mediated immunity in transitional cell carcinoma patients to transitional cell carcinoma target cells were measured during the clinical course. Initial results from a small number of patients with invasive and non-invasive tumors showed that anti-transitional cell carcinoma cell-mediated immunity increased after the tumor was removed and during administration of Bacillus Calmette-Guerin.

Surgical repair of post-hysterectomy vesicovaginal fistulas.

Herein we describe in detail the preoperative evaluation, surgical technique and postoperative care used in repairing 13 post-hysterectomy vesicovaginal fistulas by the transabdominal transvesical approach. We were successful in all 13 cases, although 1 required 2 operations. Excellent results can be obtained in these patients if a complete urological diagnostic study is done, a careful technique is used during the operation and specialized postoperative care is given. The transabdominal transvesical approach to this problem is safer and gives better results than the transvaginal operation.

Human cell-mediated cytotoxicity estimated by lymphocyte titration.

Human lymphocyte immunity to tumor-derived target cells was estimated by titrating lymphocyte concentration to achieve a 50% reduction of target cell survival. This lymphocyte titration assay gave estimates of cytotoxicity that were different from those obtained with the conventional cell-mediated cytotoxicity assays but were more proportional to lymphocyte activity. Estimates of cytotoxicity obtained using the lymphocyte titration assay were reproducible upon repeated testin over the course of several months and were relatively unaffected by two- to fourfold variations in target cell concentration. Target cell-specific cytotoxicity was reproducible but often did not appear to be tumor specific.

In vitro cultivation of epithelial cells derived from tumors of the human urinary tract.

Finely minced explants from 54 TCC2 of the human urinary tract were cultured in vitro in an attempt to establish cell lines. Cells with epithelial morphology grew out from 48 tumor explants, and long-term cell cultures were established from 10. Six of the cell cultures have been maintained for over 18 months with 50 to 70 transfers and, therefore, are considered permanent cell lines. The epithelial cells in the established cultures are small, exhibit rapid doubling time, and show multilayering. The cells were examined both microscopically and by cultivation techniques, and they were found to be free from contaminating microorganisms, including Mycoplasma. The established cultures grow rapidly in roller bottles and, therefore, can be produced in large quantities. These cells also remain viable after being stored for 3 years in liquid nitrogen.

Lymphocyte antibody interaction in cytotoxicity against human transitional cell carcinoma.

The interaction of antibodies and lymphocytes in their immune reaction against human transitional cell carcinomas was studied using the in vitro microcytotoxicity assay. A non-complement dependent, IgG antibody was detected in the serum of occasional transitional cell carcinoma patients, which induced cytotoxicity against transitional cell carcinoma target cells by lymphocytes from donors with and without transitional cell carcinoma. The observation that lymphocytes from transitional cell carcinoma donors were more sensitive to activation by this anti-transitional cell carcinoma, lymphocyte dependent antibody is compatible with the hypothesis that the surface of lymphocytes from some transitional cell carcinoma donors is coated in vivo with an anti-transitional cell carcinoma lymphocyte dependent antibody and that this antibody may be a significant factor in immunity to transitional cell carcinomas of the urinary tract.

Sexual function following high retroperitoneal lymphadenectomy.

The mechanism of infertility after high retroperitoneal lymphadenectomy was studied in 36 patients. In 35 patients infertility occurred because of unavoidable seminal vesicular and ductus deferential sympathetic denervation, with consequent aspermia. Potency and orgasm were unchanged in all patients and there was no evidence of retrograde ejaculation. The findings suggest that sympatholytic drugs be studied for possible application as reversible male contraceptives.

Adult teratoma of the testis metastasizing as adult teratoma: case report and review of literature.

A case of adult teratoma of the testis in a 19-year-old white man is presented, The tumor metastasized to the retroperitoneal lymph nodes and the metastases were also well differentiated, benign-appearing tissues. This case suggests that even the benign-appearing components of these tumors have malignant potential. Because these tumors are malignant, they should be treated by radical orchiectomy and radical retroperitoneal dissection.
