[Use of hemolysis in gel in the serodiagnosis of rubella. Comparison with hemagglutination inhibition].

The hemolysis in gel test (HIG) was compared with the hemagglutination inhibition test (HI). This new test is simple and time-sparing since it does not require dilution or serum pre-treatment, and can be measured directly in mm. It is not more expensive than the HI method. It has proved sensitive and is not affected by non specific serum hemagglutination inhibitors and is conclusive whenever HI titers are looked upon with suspicion (titers of less than 1/20). It is therefore well adapted to mass screening of immunity against rubella but in the field of recent infections HI method still has its role to play. Using the HIG test as a reference, we measured the loss of Ig to rubella following the three most commonly employed methods for removal of inhibitors: the mean values of 16 sera were 25% of antibodies lost after kaolin pre-treatment, 62% lost after heparine-Mn-Cl2 procedure 75% lost after dextran-CaCl2 treatment.

[Infantile diarrhea and rotavirus (author's transl)].

The acute and convalescent sera of 328 children with acute enteritis were checked with the bovine diarrhea virus as antigen in complement fixation test. 13% had a seroconversion. Some cases were associated with skin rashes or signs of respiratory infection.

Spermatozoa as potential carriers of HIV.

In order to investigate the role of germ cells in the sexual transmission of immunodeficiency virus (HIV), spermatozoa from healthy HIV-seronegative men were incubated in vitro with HIV1. After washing, they were cocultured with peripheral blood leukocytes from seronegative blood donors. Reverse transcriptase assays and p24 antigen tests were performed in culture supernatants. Electron microscopy examination of these HIV-incubated spermatozoa was carried out, as well as the search for CD4 molecules on their surface. Although virus bound to and seemed to enter spermatozoa despite the absence of detectable CD4 epitopes on their surface, no replication of HIV was apparent. However, HIV particles on the surface of spermatozoa were capable of infecting CD4 T lymphocytes. Present results would seem to preclude artificial insemination between an HIV-seropositive man and an HIV-seronegative woman.

High concentrations of 2-5A, the interferon intracellular mediator, in the blood of children with acute viral infections.

We measured the concentration of 2-5A (2',5'-oligoadenylate), an intracellular mediator of the antiviral action of interferon, in the blood of children with acute viral and bacterial infectious diseases. 2-5A concentration was found to be elevated in several children with viral diseases. This elevation seemed transient and was not specific for viral infections. We provide arguments for the use of 2-5A as a marker of the evolution of diagnosed viral diseases.

[Hepatitis delta virus RNA detection by one-step RT-PCR].

We have developed a method of detection HDV RNA where the reverse transcription and amplification are carried out in the same tube, thus reducing the handling time and the contamination risk. RNA extracted from serum or plasma on a microcolumn technique (kit QIAamp viral RNA, QIAgen) is submitted to reverse transcription and amplification by using the One-Step RT-PCR kit (QIAgen). Primers and probe were placed in the conserved ribozymic regions of the HDV genome. The sensitivity of the technique was estimated to 420 copies per reaction by using serial dilutions of a tittered sample. Its specificity was shown by the negativity of 24 samples from anti-HDV negative subjects, either healthy or coinfected by HBV, HCV or HGV. A low tittered sample was reproductively detected in different series. This One-step RT-PCR technique is specific, reproducible and allows the fast detection of active HDV RNA replication. Its adaptation to the routine diagnosis in a laboratory of medical virology is possible and will allow its evaluation on a greater number samples.

[Neonatal myocardial infarct associated with maternal-fetal infection due to Coxsackie B4 virus].

Heart failure with myocardial infarction occurred in the course of a neonatal infection with aseptic meningitis. Coxsackie B4 infection was found in the child and its mother. The unusual myocardial action of the virus, coronary embolus, intermediate role of a pancreatitis. By age 8 months, under digitalis glycosides treatment, growth and psychomotor development were normal.

[Post-infectious Epstein-Barr virus encephalitis].

A case of meningoencephalitis is reported in a 14 year old boy. After an acute onset with fever and coma, the outcome was rapidly favorable. Antibody titers in acute and convalescent sera demonstrated recent infection with EBV. EBV encephalitis appears to be a post-infectious encephalitis with no direct viral invasion of the CNS and no acute viral replication.

[Value of the fractionation of serum immunoglobulins for the diagnosis of rubella infection in pregnant women. Critical study].

The authors present the results of fractionation by ultracentrifugation in the identification of rubella antibodies in 20 vaccinated children and 389 pregnant women suspected of having rubella. Testing for specific IgM was constantly positive when an exanthematous disease was seen in association with a significant increase an antibody level as measured by standard techniques, making questionable the reed of fractionation under such circumstances. By contrast, however, the shortest delay in collection of the first sample of serum renders fractionation essential. Alone, it made possible the diagnostic of rubella in patients in whom the first serum, although collected within 3 to 5 days after the rash, had the same antibody level as a second serum collected later. Whilst fractionisation requested for contact only without rash did not reveal specific IgM, positive results were obtained rarely in sera with a high antibody level on routine examination. The very uncertain significance of these last results, the frequency of cases of recent rubella with very modest antibody levels and the occasional finding of duodtful results, render the authors highly sceptical as to the value of fractionisation in the presence of a high serum level on routine examination in a pregnant woman.
