A transmembrane electrical potential generated by respiration is not equivalent to a diffusion potential of the same magnitude for ATP synthesis by Bacillus firmus RAB.

ATP synthesis by starved whole cells of alkalophilic Bacillus firmus RAB was energized by addition of DL-malate or by imposition of a valinomycin-mediated K+ diffusion potential. At pH 9.0, the transmembrane electrical potentials produced by these two means were similar in magnitude, at close to -170 mV. While N,N'-dicyclohexylcarbodiimide-sensitive ATP synthesis occurred upon the addition of DL-malate, no ATP was synthesized in response to a diffusion potential. In contrast, Na+-dependent accumulation of alpha-aminoisobutyric acid was energized equally well by DL-malate or a diffusion potential at pH 9.0. Even at pH 7.0, DL-malate was more efficacious than a diffusion potential in energizing ATP synthesis as assessed by determining the phosphorylation potentials generated at transmembrane electrical potential values of different magnitudes. Both modes of energization did, however, result in N,N'-dicyclohexylcarbodiimide-sensitive ATP synthesis at pH 7.0. The results of these studies are consistent with a model of energy coupling in which one pathway of protons between the respiratory proton pumps and the H+-translocating ATPase is direct or localized. Whereas an artificially imposed electrochemical gradient of protons can energize ATP synthesis under certain experimental conditions, in the proton-poor milieu that is optimal for growth of alkalophilic bacilli, the direct proton pathway may be crucial.

Priority in the pharmacological management of chronic schizophrenia: the quality of life issues.

The progress of pharmacological management of schizophrenia is briefly reviewed, highlighting some of the major limitations especially in regard to side-effects which have been identified. The importance of the quality of life issues in chronic patients undergoing pharmacological treatment is stressed. A possible new approach is the use of beta-adrenergic blocking drugs which may have symptomatic effects as well as quality of life effects.

Chromosome studies of patients on long-term lithium therapy for psychiatric disorders.

Chromosome studies, including sister chromatid exchange techniques, were performed on blood samples from 23 patients receiving long-term, continuous lithium therapy for psychiatric disorders. Nineteen healthy, age-matched individuals were used as controls. No increase in chromosome damage was detected, when samples from the lithium group were compared with the control group samples.

Huntington's disease: the establishment of a national register.

The historical background of Huntington's disease in Australia is briefly described, together with the development of professional and self-help groups. An existing Huntington's disease register in Victoria is described in detail. A case is made for the need for the establishment of an Australia-wide register of Huntington's disease. Certain ethical issues and safeguards necessary to such a register are discussed.

A linkage study of the loci for Huntington's disease and some common polymorphic markers.

The phenotypic frequencies of 24 polymorphic blood markers in a sample of patients with Huntington's disease (HD) have been compared with those in a sample of healthy Australian volunteers in an effort to detect any associations between HD and the markers concerned. The Rh factor, c, has a significantly lower frequency in the HD sample while ACP1c and Gm1,2 have a significantly higher frequency. The linkage relations of the HD locus have been analysed with respect to the various marker loci concerned. This analysis involved the development of methods to overcome the general lack of genetic data concerning the affected parent and the possibility that presently unaffected offspring may be asymptomatic carriers of the HD gene. The results suggest that close linkage between the HD locus and Fy, ADA, ACP1, Gc or Bg is highly unlikely. They also suggest a low probability of close linkage to ABO, Rh, Jk, Lu, AK1, PGM1 or C3. Positive linkage scores were obtained for P, Hp and Gm. The results are inconclusive for MNSs, K, Le, Se, GPT and Inv. The available data were uninformative for linkage between the HD locus and Co, 6-PGD or E1.

Platelet monoamine oxidase activity in Huntington's chorea.

The possibility that genetically determined abnormalities in the monoamine oxidase of certain central nervous system aminergic neurones may play a part in the pathology of Huntington's chorea was investigated using human platelet monoamine oxidase. Significantly elevated monoamine oxidase activity was found in male patients compared to control subjects suggesting this may be a screening test for this disorder. Low monoamine oxidase activity was associated with a better clinical response to drugs.

Comparison of low doses of haloperidol and diazepam in anxiety states.

A clinical comparison was made between a butyrophenone drug (haloperidol), and an established antianxiety agent of the benzodiazepine group (diazepam). Both drugs were highly effective in reducing anxiety symptoms, with few and mild side effects of diminishing intensity throughout the trial period.

A probable case of mutation in Huntington's disease.

A patient is described in whom Huntington's disease was diagnosed at the age of 34 years. No evidence of the disorder was found in either parent. Their parentage of the alleged mutant could not be excluded from a study of the inheritance of 25 genetic markers.

Double-blind comparison of clozapine with chlorpromazine in acute schizophrenic illness.

Double Blind comparative trial of a new dibenzodiazepine derivative Clozapine (Leponex) with Chlorpromazine was conducted in the treatment of acute schizophrenic illness over a 6 week period. Factor Analysis of ratings in 9 matched pairs indicates that Clozapine, at 300 mg. per day, is comparable in efficacy to Chlorpromazine in all factors except "Irritability" for which Clozapine appears to be superior. Illness severity and Global Change ratings in all patients showed that Clozapine is more effective in producting a shift towards improvement at the end of 6 weeks. Major side effects reported in Clozapine confirmed sedation and hypersalivation as consistent problems and presence of rigidity and tremor (extra-pyramidal) being at variance with other studies.
