Lymphocyte subpopulation changes by aging.

Peripheral blood lymphocyte suspensions of healthy young and aged subjects were tested for the percentage of (1) E-rosetting cells by three different modifications of the rosette technique; (2) alpha-naphthylacetate esterase positive cells of different staining patterns; (3) IgG-Fc receptor positive cells; (4) C3 receptor positive cells; (5) labile and stable bound surface immunoglobulin positive cells; and (6) cells bearing different classes of immunoglobulins on their surface or intracytoplasmically. Age dependent changes were registered both within the T-cell and the B-cell subpopulations, some of which may be due to in vivo activation of lymphocytes. Attention is called to some technical aspects of lymphocyte subpopulation determinations and to the significance of quantitative changes in the proportions of lymphocyte subpopulations in respect to the age dependent functional changes of lymphocytes.

Morphological studies of the pigment present in the seminal vesicles of orchidectomized hamsters, and the preventive action of vitamin E and methylene blue.

Authors studies the histological and electromicroscopical picture of the seminal vesicles of hamsters after orchidectomy, after orchidectomy and Vitamin E feeding and after orchidectomy and Methylene Blue treatment. They states that 120 days after orchidectomy large amounts of lipofuscin and lamellar structures, myelin-like forms developed in the cytoplasm of the epithelial cells and macrophages of the seminal vesicles. Vitamin E feeding inhibited the appearance of lipofuscin, while Methylene Blue significantly reduced the amount of lipofuscin.

[Age-related changes in the lymphocytes as a model of aging].

Lymphocytes of spleen and lymph nodes of Wistar rats and CBA/Ca mice, as well as human peripheric lymphocytes have shown similar age related alterations. These alterations appeared in form of the loss of the mitochondrial christae which were replaced by myelin-like figures or filamentous structure. In some cases lymphocytes contained electron lucid material and giant mitochondria. These mitochondrial changes were seen in human T-lymphocytes. Authors believe that lymphocytes represent a suitable model for the study of the process of aging and also for testing factors influencing it.

The influence of influenza vaccinations on human peripheral blood lymphocytes relating to aging.

Twenty healthy, aged more than 60 years were compared to 20 healthy young volunteers less than 40 years following influenza vaccinations. 0.5 ml, 10 U. of killed influenza vaccine Hong Kong B/873 was repeatedly given intramuscularly. Blood was collected before the vaccination, 6 days, respectively 6 weeks after vaccinations. The purified venous blood lymphocytes were examined by immunohistological and sheep erythrocyte rosette-forming methods. The absolute number of T and B lymphocyes was counted. It has been found that increase of IgG-bearing cells was higher in young persons, than in the elderly both in the first and in the revaccination. The absolute number of IgM-bearing lymphocytes increased in the aged after influenza vaccinations. The vaccine had no significant effect on the absolute number of IgA-bearing lymphocytes. The absolute number of T lymphocytes participating in the immune reaction showed a transitory increase solely in young persons. Experiments aiming to demonstrate the reduced activation of B lymphocytes in the elderly with the influenza vaccinations. It can be concluded, that the proliferation ability of the immunoglobulin-bearing B cells and T cells is reduced due to old age.

[Experimental investigation on possibilities of regeneration of the immunity in old animals (author's transl)].

The effect of Levamisol, potassium polyadenylate, complete Freund's adjuvant and "antigen premunition" on the primary and secondary immunological response to sheep erythrocytes as well as the reaction of "graft versus host" were tested on mice of the CBA strain belonging to various age groups. The greatest stimulation effect in both types of response, i.e. humoral and cellular, was produced by "premunition". Levasimole clearly increased the reaction of "graft versus host", while the humoral response was weakened. The application of potassium polyadenylate produced a weaker stimulus to the response to sheep erythrocytes, and it was not effective in the response of "graft versus host". The complete Freund's adjuvant had no influence whatsover on the humoral immune response in old mice.

Electronmicroscopical and immunoelectronmicroscopical examination of the lymphocytes of young and old people after influenza vaccination.

Authors examined the lymphocytes of young and old individuals before, 6 days and 6 weeks after influenza vaccination. Vaccination was carried out with killed Influenza B/Hong-Kong virus. Electronmicroscopic structure of the lymphocytes and membrane bound surface IgG and IgM were studied. Membrane bound surface IgG and IgM were seen on the small lymphocytes and on the membrane of the medium-sized lymphocytes in young and aged individuals before and after vaccination. In both age groups intracytoplasmic tubuloreticular structures were seen in the lymphocytes. These findings were not age dependent alterations. Age dependent changes could be detected on the mitochondria of the lymphoid cells. The mitochondrial crysts in aged individuals disappeared and were replaced by myelin-like forms or by faint density substance. The mitochondrial changes were present before vaccination and after vaccination in non labelled, presumably T lymphocytes. The injury of the mitochondrium can influence the function and the proliferative capacity of the cells.

Immuno-electronmicroscopic studies after influenza vaccination of young and old individuals.

Authors examined the lymphocytes of young and old individuals after influenza vaccination. They stated that membrane bound surface IgG and IgM were detectable on the lymphocytes and occasionally on lymphoblasts in both the old and young age groups. In both age-groups intracytoplasmic tubulo-reticular structures (TRS) were seen in the lymphocytes and lymphoblasts. In young individuals TRS was present in the peroxidase positive cells, in the contrary in old persons TRS was never found in cells with surface immunoglobulins. In the TRS themselves peroxidase positive IgG and IgM could be detected.

Autoantibodies in aged individuals.

Authors determined in 282 individuals autoantibodies in 132 T and B lymphozytes. Theirs results were as follows: 1. The frequency of the presence of autoantibodies increases with age. 2. In the presence of autoantibodies the absolute T and B lymphocyte counts of aged persons markedly decrease; the absolute T and B lymphozyte counts of young adult people do not change by existing autoantibodies. 3. There is a significant difference between the absolute T cell counts of aged and young adult age groups, both age-groups having autoantibodies. 4. Comparing the frequency of occurence of autoantibodies in aged healthy persons and in aged ones with cardiovascular changes and being in the habit of smoking and consuming alcohole, respectively, a significant difference could be demonstrated; in the latter group the frequency was much higher.

The incidence of immunoglobulin-A deposits in glomerular diseases and its morphological examination: nephropathy with mesangial IgA dense deposits.

The presence of IgA with IgG, C', IgM and fibrin in 365 renal biopsy specimens has been studied by the immunohistological method. Glomerular IgA deposits were found in 38 cases (10.4%). The patients were divided into two groups according to the localization of IgA: 13 cases (3.56%) belonged to the first group, in which IgA showed diffuse mesangial localization. Light microscopic examination showed various stages of proliferative glomerulonephritis. These cases corresponded to the nephropathy described by Berger. In 25 cases (6.85%), belonging to the second group, the glomerular localization of IgA was found to be segmental and peripheral, appeared always in patches and was present with various histological alterations. Differentiation of the two kinds of alteration is possible primarily on the basis of the immunohistological pattern, but light microscopic and electron microscopic examination might also be helpful.

Herpesnephropathy.

Two cases of acute renal insufficiency occurred in association with episodes of severe encephalitis due to herpes simplex type I. The possibility was considered that the renal failure was due to viral infection of the kidneys, and animal experiments were carried out in an attempt to confirm this. Young New Zealand albino rabbits were infected i. v. with HSV type I; the virus antigen was detected in the kidney of 8 of 10 animals, and IgG was found on the GBM in 9 of 19 animals. Viruria was observed in 12 of the 29 infected animals, and electronmicroscopic examination confirmed the presence of immune complexes in the glomeruli.
