Effect of pentoxifylline on platelet aggregation.

The effect of various concentrations of pentoxifylline, a methylxanthine, on platelet aggregation was evaluated in vitro by Born's method. The study was performed on platelet-rich plasma obtained from two series of clinically healthy subjects: in the first, pentoxifylline was added to platelet-rich plasma just before ADP or epinephrine; in the second, pentoxifylline was incubated with platelet-rich plasma for 150 minutes at 37 degrees C before testing. Statistical analysis of the results demonstrates that pentoxifylline exerts a dose-dependent inhibiting action on platelet aggregation, particularly when epinephrine is chosen as the aggregating agent, and that the pre-incubation of pentoxifylline with blood platelets greatly enhances its inhibitory effect.

Polyacrilonytrile versus cuprophan membranes for hemodialysis: evaluation of efficacy and biocompatibility by platelet aggregation studies.

The short- and long-term effect of hemodialysis with two different membranes -- cuprophan and polyacrilonytrile -- on platelet aggregation has been investigated in 12 uremic patients undergoing extracorporeal dialysis, passing from one treatment to the other. Cuprophan membranes failed to correct the defective platelet aggregation of uremia, and their thrombogenicity was documented by a fall in platelet count and further impairment of platelet aggregation during dialysis. On the contrary, polyacrilonitrile membranes showed the capacity to correct completely but transiently the platelet aggregation, without changes in platelet count. The results indicate that polyacrilonytrile membranes show a better biocompatibility toward platelets than cuprophan membranes.

Effect of different concentrations of citrate on the sensitivity of an "in vitro" spontaneous platelet aggregation test.

Our data show that variations of the concentration of citrate in blood used for the "in vitro" spontaneous platelet aggregation tests, at the limits of those generally held to be without effect, have a definite influence on the results obtained. In addition, out findings suggest that a correct selection of one of the two variants of the test allows a flexibility of the sensitivity and, reciprocally, the specificity without interfering with the quality of the information which they are capable of providing. Thus, a differentiated application of Test A and Test B is possible according to different experimental or diagnostic necessities for the prevailing need of, respectively, a maximum sensitivity or of a maximum specificity.

Effect of peritoneal dialysis, haemodialysis and kidney transplantation on blood platelet function. I. Platelet aggregation by ADP and epinephrine.

Quantitation of the thrombocytopathy of uraemics may be one useful way of evaluating forms of therapy. 24 patients treated by haemodialysis and peritoneal dialysis at two different times had platelet aggregation studies whose parameters were compared with those of 24 normal persons, 5 successful transplants or 13 untreated uraemics. Renal transplantation and peritoneal dialysis improved platelet function. The haemodialysis procedure itself impaired platelet function: this was not due to heparin.

Epidemiological survey on multiple sclerosis in Italy.

An epidemiological survey on Multiple Sclerosis was carried out in 1975 with similar methods in 9 italian provinces: 4 in the North, 2 in central Italy and 3 in the south and islands. It was found that the incidence of the disease was partically uniform in the northern provinces, in the central Italy and in Bari province. It is important however to complete in greater detail such survey in the south of Italy where the data are still incomplete.
