Prolyl hydroxylase activity as an index of liver damage induced by ethanol.
It was found that chronic intoxication of rats with ethanol results in an increase of prolyl hydroxylase activity in liver and serum of the experimental animals. The increase of enzyme activity precedes the morphological symptoms of liver damage. The possibility arises that the assay of prolyl hydroxylase in serum or in liver biopsy samples could be useful for the diagnosis of the tendency of some individuals to develop liver cirrhosis induced by ethanol.
Prolyl hydroxylase activity in liver and serum of rats and human subjects intoxicated by chronic administration of ethanol.
It was found that chronic intoxication of rats with ethanol results in an increase in prolyl hydroxylase activity in liver and serum of the investigated animals. Similar effects have been observed in some alcoholic patients. We hope that the assay of prolyl hydroxylase in serum or in liver biopsy samples might be useful for the diagnosis and prognosis of the tendency of some individuals to develop liver cirrhosis.
Heterogeneity of collagen isolated from the Guérin epithelioma.
Molecular polymorphism of collagen isolated from the Guérin epithelioma was investigated. Tumour homogenate was digested with pepsin in acidic solution. Above 70% of total tumour collagen was solubilized by this procedure and then it was submitted to fractionation by stepwise increase of NaCl concentration. It was found that collagen of this tumour is not a homogeneous protein. At least two different collagen types were found. As can be concluded from the chromatographic analyses, electrophoretic properties and amino acid composition, the tumour contains type I and type III collagen.

Stimulatory effect of platelet-breakdown products on muscle regeneration.
The effect of platelet-breakdown products on the process of regeneration of mechanically injured anterior tibial muscles was investigated in rabbits. It was found that repeated injection of the platelet homogenate into the site of the damage accelerated muscle regeneration.

Stimulatory effect of platelet homogenate on amino acid transport into L-929 cells.
It was found that the addition of platelet homogenate to confluent cultures of L-929 cells causes distinct increase of leucine and proline transport into the cells. A significant part of these amino acids is incorporated into high molecular, nondialysable material. The addition of cycloheximide to culture medium completely inhibits incorporation of proline and leucine into proteins but it does not affect the transport of these amino acids into the cells.
The stimulatory effect of platelet homogenate on prolyl hydroxylase activity in L-929 cells.
We have found that the addition of platelet homogenate to confluent cultures of L-929 cells increases 2-3 times the activity of prolyl hydroxylase in these cells. Furthermore, it was found that the platelet homogenate potentiates the effect of ferrous ions and ascorbic acid, which are known activators of prolyl hydroxylase. The effect of the platelet homogenate is diminished by cycloheximide. It seems probable that some products present in the platelet homogenate may promote biosynthesis of the enzyme or they stimulate glycolysis and accumulation of lactic acid, an activator of the hydroxylase.
Heterogeneity of collagen isolated from methylcholanthrene-induced sarcoma.
Rat fibrosarcoma induced by subcutaneous injection of methylcholanthrene was found to contain at least three different types of collagen. Two of them were identified as type I and type III collagens, the third (fraction B) seems to be specific for this tumour. The ratio of type I to type III collagen is lower in fibrosarcoma than in normal rat skin. The number of hydroxyproline residues in alpha 1 (I), alpha 2 (I) and alpha 1 (III) chains of tumour collagen appears to be higher than in the corresponding chains of rat skin collagen. Fraction B is composed of three identical alpha chains connected with disulphide bonds. It contains a relatively low amount of glycine: 234 molecules per 1000 residues. The amount of hydroxyproline and cysteine is similar to that found in the type III collagen.
Cytoplasmic 3H-arginine polypeptides of Ehrlich ascites tumour cells.
Low molecular weight ninhydrin positive peptide fractions of the Ehrlich tumour cell cytoplasm were isolated and characterized. After preliminary gel filtration of the cytoplasm on Sephadex G-25 column, the peptide mixture was fractionated on cationic exchanger SP-Sephadex C-25 column and eluted with increasing pH gradient. Five peaks were obtained. Only the first peak contained sugar component. All five peptides were studied with respect to molecular weight, isoelectric point and electrophoretic homogeneity. The cytoplasm of Ehrlich tumour cells contains one peptide of acidic (pI-5.0), two slightly basic (pI-7.7 and pI-7.7) and two strongly basic nature (pI-8.7 and pI-8.9). Molecular weights varied from 8 500 to 18 500 daltons. The origin of these peptides is briefly discussed.

Content and distribution of collagen in venous and polyester aortal grafts.
The studies were carried out on autogenous venous and polyester grafts implanted in the abdominal aorta of dogs. These grafts were excised 1,4 and 36 months after implantation and together with the normal arteries and veins, after dissection into intimal, medial, and adventitial layers, tested for their hydroxyproline and nitrogen content. It was found that the collagen content and the hydroxyproline/nitrogen ratio in the various layers of the venous and polyester grafts increased as time elapsed after the operation. The increase in collagen content was, however much higher in the polyester grafts than in the venous. The relative index of the collagen content expressed by the above ratio was found to be almost equal in the intimal and medial layers of the venous and polyester grafts 3 years after the operation and to be slightly higher than the content in the corresponding layers of the normal arteries and veins.

Stimulatory effect of blood platelet factor(s) on glycolysis in cell cultures.
The addition of bovine blood platelet homogenate to confluent cultures of three established mouse cell lines (L-929, Balb c/3T3 and SC) brings about a rapid increase of lactic acid formation. Homogenates prepared from quiescent cells previously treated with platelet breakdown products show an increased rate of glycolysis as compared with homogenates of nonstimulated cell cultures. Activities of hexokinase, pyruvate kinase and lactate dehydrogenase remained unaltered but the activity of phosphofructokinase increased by about 60 per cent. Transport of 2-deoxy-D-glucose into the cells was slightly stimulated. It is concluded that the transport of glucose was not a limiting factor in confluent 3T3 cells. The findings suggest that the increase of phosphofructokinase activity may have accounted for the changes in glycolysis seen in intact cells and cell-free homogenates prepared from cultures exposed to some unknown factor(s) present in platelet homogenate.

Stimulatory effect of platelet homogenate on collagen biosynthesis in L-929 cells.
Confluent cultures of L-929 cells were incubated at 37 degrees C for 16 h in serum-free culture medium supplemented with ascorbic acid, beta-aminopropionitrile and 5 3H-proline and the amount of collagen synthesized in such conditions was evaluated. It was found that addition of platelet homogenate to incubation medium causes significant increase of collagen biosynthesis by these cells.
