Model testing in radioligand/receptor interaction by Monte Carlo simulation.

Monte Carlo simulations have been applied for evaluating the reliability of parameter estimates as well as for testing models in radioligand saturation binding experiments. Scatchard analysis was compared to the nonlinear least-square curve fitting method for one-site saturation binding curves. It was found that linear regression analysis from the transformed data in the Scatchard plot yielded generally less accurate parameter estimates than nonlinear regression analysis of untransformed data. The advantage of the nonlinear least-squares curve fitting method was especially pronounced in cases where the scatter and number of data points, as well as the radioligand concentration range, were chosen similar to less optimal experimental conditions. Under such circumstances, several KD and Bmax values derived by Scatchard analysis led to physically impossible negative values whereas the same data analyzed by nonlinear regression yielded reasonable parameter estimates. Furthermore, it was found that for both means of analysis, KD and Bmax correlated positively. In another set of Monte Carlo experiments, saturation binding curves involving two receptor sites were generated and subsequently analyzed according to both a one-site and a two-site model. The confidence with which one is able to distinguish the two-site model from nonlinear least-squares curve fitting was then estimated for optimal, as well as for, less ideal experimental conditions.

Elevated adrenaline and increased alpha-adrenoceptor-mediated vasoconstriction in essential hypertension.

In patients with essential hypertension, plasma adrenaline, regardless of age, was consistently higher than in normotensive controls; adrenaline correlated with heart rate and the vasodilator response in the forearm circulation produced by postjunctional alpha 1-adrenoceptor blockade with prazosin. This dilator response to prazosin was greater in hypertensive patients. Together, this suggests elevated sympathetic activity and enforced vasoconstriction via postjunctional alpha 1-adrenoceptors in essential hypertension. beta-Adrenoceptor-mediated cardiovascular responses decrease with age and even more with high blood pressure, which contributes to unopposed alpha-adrenoceptor-mediated vasoconstriction. This could explain the transition from an early high cardiac output into a later high peripheral resistance form of hypertension.

High-affinity binding sites for [3H] verapamil in cardiac membranes.

The calcium channel blocker [3H]verapamil showed specific binding of high affinity (equilibrium dissociation constant, KD, of 4.25 nM) and low density (maximal number of binding sites, Bmax, 50 fmol/mg protein) in frog heart membranes. Nitrendipine, a new calcium channel blocker, had a potency comparable to verapamil in the inhibition of [3H]verapamil binding. Concentrations of Ca2+ in the medium exceeding 3 mM decreased [3H]verapamil binding. These findings are consistent with the concept that high-affinity binding sites for calcium channel blockers are associated with sarcolemmal calcium channels.

'Proopiocortin fragments' in normal human adult pituitary. Distribution and ultrastructural characterization of immunoreactive cells.

The distribution and ultrastructure of cells immunoreactive towards antisera against synthetic fragments of proopiocortin were studied in human pituitaries by immunohistochemical methods. Anterior lobe cells exhibiting the ultrastructural characteristics of corticolipotropes, as well as all epithelial cells 'invading' the posterior lobe, display beta-endorphin/beta-lipotropin-ACTH immunoreactivities. At low dilution a beta-lipotropin antiserum, raised with highly purified antigen, stains also somatotropes. alpha-MSH antibodies bind to certain corticolipotropic cells in the pars distalis and to only a few of the cells 'invading' the pars nervosa. Met-enkephalin antisera react only with isolated cells in the anterior lobe. These results indicate a striking difference in the processing of the proopiocortin precursor molecule in different corticolipotropic cells of the human hypophysis. Furthermore, the hypothesis of a homology between cells 'invading' the human neurohypophysis and those of the intermediate lobe of lower vertebrates, is questioned.

Free calcium concentration in platelets closely relates to blood pressure in normal and essentially hypertensive subjects.

Calcium has been implicated in smooth muscle contraction, arterial resistance, and the pathophysiology of essential hypertension. Using the intracellularly trapped fluorescent dye quin2 , the free calcium concentration in platelets was found to be elevated in patients with borderline (n = 8, p less than 0.01) and established essential hypertension (n = 23, p less than 0.001) when compared with normotensive subjects (n = 30). There was a close correlation between intracellular free calcium and systolic blood pressure (n = 61, r = 0.882, p less than 0.001) as well as diastolic blood pressure (n = 61, r = 0.950, p less than 0.001). The slopes of the regression lines did not differ between the groups.
