Changes in cell surface and cortical cytoplasmic organization during early embryogenesis in the preimplantation mouse embryo.

Membrane topography and organization of cortical cytoskeletal elements and organelles during early embryogenesis of the mouse have been studied by transmission and scanning electron microscopy with improved cellular preservation. At the four- and early eight-cell stages, blastomeres are round, and scanning electron microscopy shows a uniform distribution of microvilli over the cell surface. At the onset of morphogenesis, a reorganization of the blastomere surface is observed in which microvilli becomes restricted to an apical region and the basal zone of intercellular contact. As the blastomeres spread on each other during compaction, many microvilli remain in the basal region of imminent cell-cell contacts, but few are present where the cells have completed spreading on each other. Microvilli on the surface of these embryos contain linear arrays of microfilaments with lateral cross bridges. Microtubules and mitochondria become localized beneath the apposed cell membranes during compaction. Arrays of cortical microtubules are aligned parallel to regions of apposed membranes. During cytokinesis, microtubules become redistributed in the region of the mitotic spindle, and fewer microvilli are present on most of the cell surface. The cell surface and cortical changes initiated during compaction are the first manifestations of cell polarity in embryogenesis. These and previous findings are interpreted as evidence that cell surface changes associated with trophoblast development appear as early as the eight-cell stage. Our observations suggest that morphogenesis involves the activation of a developmental program which coordinately controls cortical cytoplasmic and cell surface organization.

Rationale developed by the Environmental Protection Agency for the assessment of carcinogenic risks.

The intent of the Environmental Protection Agency's "Interim Guideline for Carcinogen Risk Assessment" is to provide an evaluation of the evidence regarding suspect carcinogens that encapsulates judgments on the quality and adequacy of data, the likelihood that the agent is a human carcinogen, and an estimate of the magnitude of the cancer burden that could be ascribed to the agent if no regulatory action were taken. Every effort shoult be made to reduce environmental contamination by carcinogens to the lowest possible level; this view stems from uncertainties about qualitative and quantitative extrapolations from animals to man, the lack of evidence for a dose threshold, the great range of possible interactions among environmental carcinogens and cofactors, and the broad spectrum of human susceptibility. New knowledge in the field of carcinogenesis is developing, and modifications of the Environmental Protection Agency's "Interim Guideline for Carcinogen Risk Assessment" must be made periodically.

Stroke rehabilitation: maintenance of achieved gains.

Gains made during rehabilitation following stroke were maintained or improved by the majority of rehabilitants when assessed 2 to 12 years later. Five measures of rehabilitative activity studied were self care, mobility, amount of time spent at the rehabilitant's major daily activity, vocational status, and over-all rehabilitative status. Independent variables that correlated positively with these rehabilitative functions were living at home, rather than in a nursing home, and the rehabilitant's having an accepting, rather than non-accepting, attitude about his present status. The functional levels of rehabilitation are maintained as well through education of the rehabilitant and his family as through the use of public health nursing and community resources. The interviewed rehabilitants in this study maintained the gains in function achieved during stroke rehabilitation throughout the long survival time, averaging seven to eight years; and functional loss, when it did occur, was usually secondary to a superimposed health problem. This study shows that the gains in the level of functioning achieved during stroke rehabilitation presist in long-surviving stroke patients and that the quality of life is enhanced from the perspectives of the patient and his family.

Specific deuteromethylation by the Eschweiler-Clarke reaction. Synthesis of differently labelled variants of trimethylamine and their use for the preparation of labelled choline and acetylcholine.

Reductive methylation (the Eschweiler-Clarke reaction) was used as a route for the synthesis of differently deuterium labelled variants of trimethylamine with the ultimate aim of preparing labelled variants of choline and acetylcholine. Combinations of unlabelled and labelled formaldehyde and formic acid yielded symmetrically labelled trimethylamines of high isotopic purity. The labelled congeners to acetylcholine and choline that were prepared subsequently provide suitable internal standards and tracers to be used in mass spectral analysis and in the evaluation of the pharmacokinetics of the parent compounds.

Cytological observations of the ovarian epithelium in mammals during the reproductive cycle.

We have studied the ovarian epithelijm at various stages of the reproductive cycle in a number of mammalian species utilizing light microscopy, scanning microscopy, the freeze-fracture technique, transmission microscopy and by employing specialized tracers that use lanthanum and horseradish peroxidase. We found that the epithelial cells are joined by incomplete tight junctions, gap junctions, and desmosomes. The cytoplasmic matrix contains a large irregularly shaped nucleus, few microtubules, microfilaments, mitochondria, endoplasmic reticulum and a host of coated and non-coated vesicles of varying diameters. The saccules comprising the large Golgi complex and its companion vesicles are associated with a basal body-centriole complex: some of these saccules and affiliated vesicles are acid phosphatase positive. Surface modifications of ovarian epithelial cells include numerous microvilli, some of which have a bulbous tip, and plications of the lateral plasma membrane which are thought to accomodate volume changes of the ovary during follicular development. Many coated and non-coated endocytotic caveolae were found on these cells, particularly in the basal area. These caveolae internalized exogeneously administered horseradish peroxidase. We view the marked endocytotic activity as an efficient transport mechanism for partially removing substances from the interstitium of the ovary and the peritoneum.

Gap junctions between the oocyte and companion follicle cells in the mammalian ovary.

Tracer and freeze-fracture electron microscopy of the ovaries of neonatal rat and adult mouse, rat, rabbit, and primate have revealed the presence of gap junctions between follicle cells and oocytes. The junctional connections are found at the ends of follicle cell projections which traverse the zona pellucida and terminate upon microvilli and evenly contoured nonmicrovillar regions of the oolemma. Gap junctions are often seen associated with a macula adherens type of junction. The gap junctions occasionally consist of minute ovoid plaques, but nore frequently appear as rectilinear single- or multiple-row aggregates of particles on the P-face or pits on the E-face. The functional significance of follicle cell-oocyte gap junctions is discussed with respect to the regulation of meiosis and luteinization.

Ultrastructural studies of frozen-thawed 8-cell mouse embryos.

Survival of frozen 8-cell mouse embryos transferred directly upon thawing to the uteri of Day 3 pseudopregnant foster mothers was significantly lower (26%) than the survival of unfrozen 8-cell embryos transferred immediately after collection (73%). When frozen-thawed 8-cell embryos were cultured for 20-24 hr before transfer survival was similar to that of unfrozen 8-cell embryos transferred after 20-24 hr inculture (65% and 73%, respectively). Ultrastructural examination of the frozen-thawed 8-cell embryos revealed no obvious damage to protoplasmic components.

Trace analysis of organic volatiles in water by gas chromatography-mass spectrometry with glass capillary columns.

Traces of volatile organic materials in water have been concentrated by gas phase stripping and adsorption onto a porous polymer. A simple all-glass sampling device is proposed which allows efficient concentration at elevated temperatures. Sample transfer from absorbent into a gas chromatographic column is effected by a simple one-step procedure involving heat desorption. The capacity of the absorbent has been determined for a number of model substances which are found in water. Under the sampling conditions used, compounds being less volatile than benzene are usually quantitatively retained, with some exceptions. Separations were effected with highly efficient glass capillary columns. Water samples, collected from a small number of locations, included both tap water and untreated waters. A number of volatiles have been determined by gas chromatography-mass spectrometry in drinking water and in a river which flows through an industrialized area. The drinking water examined contains a large number of chlorinated and brominated compounds whereas the river water is largely free from this class of substances. Derivatives of camphor and terpenes have been identified in this particular river.

Structural modifications of lutein cell gap junctions during pregnancy in the rat and the mouse.

By use of lanthanum tracer and freeze-fracture procedures it was found that granulosa-lutein cells of the pregnant mouse and rat ovaries are connected by gap junctions and septate-like zones of contact. Lutein cell gap junctions enlarge and become partially internalized by the end of the first week of gestation. Expansion of the gap junction domain appears to be due initially to intercalation of particles along borders of small gap junctions devoid of smaller non-junctional particles. The number of gap junction lined processes appearing at the cell border increases concomitantly with hypertrophy of the lutein cell during the second week of pregnancy. Strands of particulate or grooved membrane emanate from the margin of larger gap junctions undergoing interiorization. Most large gap junctions are intimately associated with elements of the smooth endoplasmic reticulum. Spherical gap junctional profiles assume a deeper location in the lutein cell and may form concentric arrays by term while true surface gap junctions appear to fragment in the post-partum corpus luteum. The modifications observed are interpreted with respect to biogenesis of the gap junction and the hormonal control of lutein cell function.

The formation and cytochemical characterization of cortical granules in ovarian oocytes of the golden hamster (Mesocricetus auratus).

The formation and cytochemical characterization of cortical granules in the ovarian oocytes of the golden hamster have been investigated by use of light and electron microscopical techniques. Particular emphasis is given to the changing population of organelles associated with cortical granule formation. Our observations indicate that cortical granules are produced by the participation of both the Golgi complex and the rough endoplasmic reticulum. Ultrastructural cytochemistry reveals that the cortical granules are composed of glycoprotein. The cortical granules are released at fertilization by a merocrine-type of secretory process.

Photo-incuced cross-linkage of gene-5 protein and bacteriophage fd DNA+.

The gene 5 protein, coded for by the bacteriophage fd, forms a complex with single stranded fd-DNA such that one gene 5 protein monomer interacts with four bases. Exposure of this complex to ultraviolet light results in the formation of covalent bonds between 25-30% of the gene 5 protein monomers which are bound to the DNA. In contrast, when the intact fd virion, which is a complex of coat protein and DNA, was exposed to ultraviolet irradiation, no detectable protein DNA cross-links were found.
