Cell-mediated immune responses between HLA-identical siblings: recognition of antigenic changes associated with acute myelogenous leukaemia.

Cell-mediated immune reactions between a patient suffering from acute myelogenous leukaemia (AML) and an HLA-identical sibling were studied in order to characterize the in vitro reactions in MLC and CML prior to bone marrow transplantation. Our results indicated that antigenic differences were detectable between the blasts and the remission lymphocytes. While the normal sibling did not respond in MLC to her HLA-identical sister's remission lymphocytes, there was an anti-blast response. This proliferative response, however, did not lead to the development of detectable cytotoxic cells capable of destroying blast cells. Unrelated individuals, on the other hand, responded strongly both in MLC and CML to the allogeneic tumour blasts and remission lymphocytes of the patient and the lymphocytes of the healthy sibling. The kinetics and magnitude of the MLC response to blast cells was different from that to remission lymphocytes. This response indicated that the blast cells expressed antigenic differences which were recognized in MLC by both the HLA-identical sibling and unrelated individuals. Furthermore, these tumour cells were capable of sensitizing allogeneic, but not syngeneic lymphocytes to become cytotoxic, though they seemed to be more resistant to destruction in CML than normal cells.

[Follow up of early childhood schizophrenics. Progress of 6 cases over 25 years].

Around the year 1950, we observed five cases of infantile schizophrenia; these patients were personally re-examined in 1956 and 1977. Another patient, under the care of the psychiatrists of the Public Health Department was included at a later date. One case of periodic catatonia is distinguished from other cases by a normal development up to puberty. The other five cases suffer from a "systematic" catatonia with characteristic syndromes and a slowly progressing course. As early as at the age of two or five years, they were noticeable for autism and psychomotor syndromes. The diagnosis of the catatonic subform was confirmed after 25 years. Although originally able children their intellectual development came to a standstill during the progressing process as did the maturing of the personality. A causative factor of the early development can be seen in the lack of stimulation to which four only children and one child brought up in a home were exposed.

[Cathepsin, phosphoprotein-phosphatase and acid phosphatase in the soluble fraction of the cattle brain cortex: purification and properties (author's transl)].

Cattle brain cortex was homogenised in 0, 29 mol/1 sucrose and centrifuged at 101 000 X g. The supernatant contains the majority of 3 enzymes participating in protein turnover: cathepsin (EC 3.4.4.23), phosphoprotein phosphatase (EC 3.1.3.16) and acid phosphatase (EC 3.1.3.2). They were separated by chromatography on Sephadex G 200 in neutral buffer. The cathepsin was purified up to 380 fold by gel filtration on Sephadex and column electrophoresis. The pH optimum of cathepsin was 5.7. At 37 degrees C no decrease of activity was measurable during 30 min. The Km was found to be 2.75 mg/ml Casein Hammarsten. The molecular weight by gel filtration and exclusion-gel electrophoresis was about 45 000, corresponding to the cathepsin from human liver (Barrett, A.J. (1970) Biochem. J. 117, 601-607). The sedimentation constant 3.0 S20,W is comparable with the values of proteinase of different origin, and the composition is similar with respect to the high proportion of acidic amino acids. The phosphoprotein phosphatase can be further purified by chromatography on hydroxyapatite and by column electrophoresis. The pH optimum of phosphoprotein phosphatase was about pH 5.5. At 45 degrees C no decrease of activity was measurable during 20 min; the Km was 1.43 mg/ml casein isoelectric. The pH optimum of acid phosphatase was about 5.6. At 54 degrees C NO DECREASE OF ACTIVITY WAs measurable during 30 min; the Km was 2 mumol/1 for Sodium phenolphthalein diphosphate. All three enzymes slowly lost their activity during several weeks at - 4 degrees C, apparently by self digestion in the cold.

Preliminary evidence for a fourth phosphoglucomutase (PGM) locus in the dog.

The PGM polymorphisms were investigated in starch gel electrophoresis in a total of 34 dogs. After prolonged incubation, a zone located 5-6 cm anodal to PGM3 was found in which two clear bands were identified. Two dogs had only the faster band, and two dogs had only the slower band. This provides preliminary evidence for the existence of a fourth PGM locus in dogs. A series of exclusion experiments indicates that the newly found bands contain PGM activity.
