The estimation of pairwise relationships.

Relationships between the individuals of a population have been previously studied from the point of view of prediction. Edwards (1967) suggested that the problem of detailed population structure could also be studied from the point of view of inference. Even where inferences of practical applicability cannot be made, such an approach can increase understanding of the relation between genealogical and genetic structure. In this paper we consider a specific problem which provides an introduction to the ideas and methods of genealogical inference. This is the problem of estimating the pairwise relationship between two individuals on the basis of their phenotypes at several loci. There is no theoretical problem in the extension from pairwise to joint relationship.

Genetic epidemiology: a review of the statistical basis.

Genetic epidemiology is the analysis of the familial distributions of traits, with a view to understanding any possible genetic basis; the fundamentals of this area are reviewed. The terminology of genetic models is fully described, since it is this wealth of terminology which can make it difficult for a statistician to enter the area. The statistical approaches to the analysis of data on sets of interrelated individuals are then discussed, and resulting problems of sampling design and analysis are considered.

Glucocorticoid regulation of rRNA synthesis.

Glucocorticoids inhibit transcription of the genes encoding rRNA (rDNA) in P1798 lymphoma cells. This is due to a decrease in the amount or activity of a transcription factor, called TFIC. TFIC has been purified to apparent homogeneity and the properties of this protein have been investigated in detail. TFIC is tightly associated with RNA polymerase I. The data indicate that TFIC is a bona fide initiation factor that is required for formation of the first phosphodiester bond of nascent pre-rRNA. Extracts from dexamethasone-treated cells are devoid of this factor and cannot form initiated complexes in vitro.

The inheritance of height in a Finnish population.

Although height is widely cited as the classic polygenic trait, there have been few large-scale studies of its inheritance since that of Pearson and Lee 80 years ago (Pearson and Lee 1903). Values of heritability derived in many standard genetics texts are based on those data. The data of Rissanen and Nikkila (1977) on 2869 individuals in 392 three-generation families provide an opportunity to make comparisons in a contemporary European population. We examine here height differences between regions, sexes and generations, and consider the form of the population distribution. Analysing the data within the classic polygenic framework we obtain a much higher spouse correlation in height than shown by earlier studies, a smaller heritability, and only a small effect of common-sib environment or genetic dominance deviations.

Gene survival in isolated populations: the number of distinct genes on Tristan da Cunha.

Gene extinction probabilities over the ancestral genealogy of Tristan da Cunha are analysed, to determine the probability distributions of numbers of surviving distinct founder genes. Autosomal, X-linked and Y-linked genes are considered, and also the bivariate distributions of genes originating in males and in females. One founder group shows a greater variety of surviving genes than another, although there is little difference in their total genetic contributions. This excess variety is concentrated in male lines of descent. The paper demonstrates the types of analysis that can be used to illuminate and summarize lists of gene extinction probabilities.
