Ancient DNA unveils distinctive ancestries in the Bronze and Iron Ages of East Tianshan.

The East Tianshan Mountains occupy a key corridor between Central and East Asia, but their population history remains poorly understood. Here we report genome-wide data from 135 ancient individuals from 11 archaeological sites. We identify a previously unrecognized Bronze Age admixture between populations related to Yellow River millet farmers and steppe pastoralists associated with the Chemurchek culture. In contrast, we find little genetic contribution from contemporaneous middle-to-late Bronze Age steppe pastoralists, despite their eastward expansion across the Eurasian Steppe. By the Iron Age, regional populations had become more heterogeneous, incorporating additional eastern and steppe-related sources while retaining variable contributions from Early Bronze Age groups. These results reveal sustained demographic interactions in eastern Central Asia nearly 1800 years preceding the establishment of the historic Silk Road.

The expression of retinoic acid receptors in lymph nodes of young children and the effect of all-trans-retinoic acid on the B cells from lymph nodes.

To elucidate the mechanism of vitamin A enhancing the production of antibody, we examined the expression and distribution of retinoic acid receptors mRNA in lymph nodes of young children ( < or = 5 years of age), and investigated the affection of all-trans-retinoic acid on the maturation and activation of the B cells isolated from the lymph nodes and cultured in vitro. Twenty-four normal lymph nodes were collected. By in situ hybridization, we found that all six retinoic acid receptor genes expressed and distributed widely in the lymph nodes, and the levels of mRNA for retinoic acid receptor genes (detected by RT-fluorescent quantitative PCR) were increased with the development of children. In vitro culture, all-trans retinoic acid promoted the maturation and activation of the B cells from the lymph nodes. Thus, promotion on B cells in lymph node may be the important mechanism of vitamin A reinforcing the humoral immunity in young children.

Coherence phenomena in the phase-sensitive optical parametric amplification inside a cavity.

We theoretically and experimentally demonstrate coherence phenomena in optical parametric amplification inside a cavity. The mode splitting in the transmission spectra of a phase-sensitive optical parametric amplifier is observed. Especially, we show that a very narrow dip and peak, which are the shape of a delta function, appear in the transmission profile. The origin of the coherence phenomenon in this system is the interference between the harmonic pump field and the subharmonic seed field in cooperation with dissipation of the cavity.

Molecular diagnosis of human cancer type by gene expression profiles and independent component analysis.

The precise diagnosis of cancer type based on microarray data is of particular importance and is also a challenging task. We have devised a novel pattern recognition procedure based on independent component analysis (ICA). Different from the conventional cancer classification methods, which are limited in their clinical applicability of cancer diagnosis, our method extracts explicitly, by ICA algorithm, a set of specific diagnostic patterns of normal and tumor tissues corresponding to a set of biomarkers for clinical use. We validated our procedure with the colon and prostate cancer data sets and achieved good diagnosis (>90%) on the data sets studied here. This technique is also suitable for the identification of diagnostic expression patterns for other human cancers and demonstrates the feasibility of simple and accurate molecular cancer diagnostics for clinical implementation.

[Effect of retinoic acid on the development of B cells from lymph nodes of young children and the pathway of the effect].

OBJECTIVE: To investigate the effect of retinoic acid on the differentiation and maturation of B cells from lymph node of children, and relational changes of the expression levels of retinoic acid receptor genes. METHODS: Twenty-four patients with digestive tract malformation underwent surgical operation in the surgical ward of our hospital. They were divided into 3 groups according to age: < 1 yr, 1 - 3 yr, -5 yr, 8 cases in each age group. The lymph nodes in the margin of excised tissues were obtained. The cells separated from lymph nodes were cultured in vitro. The cells were divided into 5 groups: Retinoic acid (RA), RA plus Ro41-5253, RA receptor antagonist (RA+Ro), lipopolysaccharide (LPS), lipopolysaccharide plus RA (LPS+RA) and control, and were subjected to the corresponding treatments. After 24 h and 48 h of cell culture, the surface markers on the B cells were detected by flow cytometer to observe the maturation and activation of B cells. The expression levels of retinoic acid receptor genes were quantitatively analyzed by RT- fluorescent quantitative PCR. RESULTS: After culture, in < 1 yr group, the number of the mature B cells (IgM(+)IgD(+)CD25(-)) in the RA group was significantly higher than that in the control (24 h: 23% +/- 5% vs. 17% +/- 3%; 48 h: 28% +/- 6% vs. 22% +/- 4%) (P < 0.05), and that in the RA + RO group was not significantly different from that in the control (24 h: 16% +/- 4%; 48 h: 20% +/- 9%) (P > 0.05). The number of activated B cells (IgM(+)CD25(+)) in the LPS + RA group was obviously higher than that in the LPS group (24 h: 82% +/- 10% vs. 76% +/- 8%; 48 h: 83% +/- 8% vs. 78% +/- 10%)(P < 0.05). The level of RARalpha gene expression of B cells (lg copies/50 ng RNA) in the RA group was significantly higher than that in the control (24 h: 7.03 +/- 1.36 vs. 5.79 +/- 2.05; 48 h: 7.91 +/- 1.60 vs. 6.21 +/- 1.88) (P < 0.05), and that in the LPS + RA group was significantly higher than that in the LPS group (24 h: 7.29 +/- 1.53 vs. 5.98 +/- 1.48; 48 h: 7.83 +/- 1.66 vs. 5.79 +/- 2.36)(P < 0.05). In 1 - 3 yr group the changes of maturation and activation of B cells in the lymph nodes were the same as the < 1 yr group. In -5 yr group, such changes were not significant. CONCLUSION: RA can promote the development of B cell from lymph node in vitro culture in <or= 3 yr children, which may be an important mechanism of vitamin A reinforcing the anti-infective immunity in young children. The expression and regulation of retinoic acid receptor genes may take part in the ontogenesis of B cell, and play a key role in the regulation of retinoic acid.

A genetic variation map for chicken with 2.8 million single-nucleotide polymorphisms.

We describe a genetic variation map for the chicken genome containing 2.8 million single-nucleotide polymorphisms (SNPs). This map is based on a comparison of the sequences of three domestic chicken breeds (a broiler, a layer and a Chinese silkie) with that of their wild ancestor, red jungle fowl. Subsequent experiments indicate that at least 90% of the variant sites are true SNPs, and at least 70% are common SNPs that segregate in many domestic breeds. Mean nucleotide diversity is about five SNPs per kilobase for almost every possible comparison between red jungle fowl and domestic lines, between two different domestic lines, and within domestic lines--in contrast to the notion that domestic animals are highly inbred relative to their wild ancestors. In fact, most of the SNPs originated before domestication, and there is little evidence of selective sweeps for adaptive alleles on length scales greater than 100 kilobases.

[Combined open prostatectomy and preperitoneal inguinal herniorrhaphy: a 21-case report].

OBJECTIVE: To evaluate the effect of simultaneous open prostatectomy and preperitoneal inguinal herniorrhaphy. METHODS: Twenty-one patients with benign prostatic hyperplasia(BPH) and inguinal hernia, underwent simultaneous suprapubic or retropubic prostatectomy and preperitoneal inguinal herniorrhaphy through Pfannenstiel's incision. In 4 cases with bigger inguinal mass, hernia repair was performed with polypropylene meshwork (PPM). RESULTS: The 21 patients were followed up for 6 months to 7 years. There were 2 cases of recurrence (9.5%), with no morbidity and mortality in the other 19 cases. CONCLUSIONS: Open prostatectomy and preperitoneal inguinal herniorrhaphy can be performed during the same session without increasing infection and recurrence. The operation method is simple and the effect is positive, especially for older men.

[Investigation of the bacteria in the seawater of Xisha in the South China Sea and their antibiotic sensitivity profile].

OBJECTIVE: To investigate the distribution and antibiotic sensitivity of the bacteria in the seawater of Xisha in the South China Sea. METHODS: The samples of the seawater in Xisha were collected for bacterial counting, identification and antibiotic sensitivity tests. RESULT: Of the 84 bacterial strains isolated from the 80 seawater samples, 38 (45.24%) belonged to Vibrio alginolyticus, 11 (13.10%) to Sphingomonas, 9 (10.72%) to Comamonas terrigena, 7 (8.33%) to Werepseudomonas, 6 (7.14%) to Moraxella, 5 (5.95%) to Cedeceadavisae, and 8 to other bacteria including 2 Aeromona, 2 Acinetobacter, 1 Pantoea agglomerans, 1 Kingella kingae, 1 Shewanella putrefaciens and 1 Suttonella indologenes. Tests of the antibiotic sensitivity showed that all the bacteria were high sensitive to 16 kinds of the antibiotics with the exception of Vibrio alginolyticus, Sphingomonas and comamonas terrigena that were less sensitive to aztreonam or piperacillin and Sphingomonas to cefotaxime or cefepime. CONCLUSION: The investigation will be of great significance for early-stage prevention and cure of bacterial infection acquired from the seawater.

[Rehabilitation therapy in early stage following spinal cord injuries].

OBJECTIVE: To investigate the effects of rehabilitation therapy in early stage following spinal cord injury on the patients' activities of daily living (ADL) and functional independence measure (FIM). METHODS: Ninety-two patients with spinal cord injuries were divided into early rehabilitation group (n=48) and control group (n=44) according to the timing of the rehabilitation therapy. Rehabilitation assessment of the patients with Barthel index and FIM was carried out before and 6 months after the rehabilitation therapy respectively for comparison. Different rehabilitation regimens were adopted for specific clinic stage of the patients, lasting for a total of 6 months. RESULTS: Barthel index and FIM score were all improved after the rehabilitation therapy in both of the two groups, but the improvements were more significant in early rehabilitation group (P<0.01). CONCLUSION: Early rehabilitation treatment is crucial for patients with spinal cord injuries in that it improves the patients' motor function, ADL and functional independence after the injury.

[Prognostic value of changes of peripheral blood T cell subsets after thermoradiotherapy for nasopharyngeal carcinoma].

OBJECTIVE: To assess the prognostic value of changes of peripheral blood T cell subsets after thermoradiotherapy for nasopharyngeal carcinoma (NPC). METHODS: Peripheral blood T cell subsets in 20 normal subjects (control group), 30 NPC patients undergoing thermoradiotherapy, and 20 NPC patients undergoing radiotherapy were detected by flow cytometry. RESULTS: The percentages of CD3+CD4+ and CD8+CD28+ cells were decreased and the percentages of CD3+CD8+ and CD8+CD28- cells increased as compared with the measurements in normal persons. One month after thermoradiotherapy, the percentages of CD3+CD4+ and CD8+CD28+ cells further decreased and the percentages of CD3+CD8+ and CD8+CD28- cells further increased, which continued to worsen 3 months after the treatment and appeared to be related to the survival of the patients. CONCLUSION: T cell subsets of NPC patients are abnormal and their immune functions depressed in NPC patients within a long period after thermoradiotherapy. CD8+CD28+ and CD8+CD28- T cell subsets can be significant for prognostic assessment in these patients after thermoradiotherapy.

[Effect of power-frequency electromagnetic fields on stroke during rehabilitation].

OBJECTIVE: To explore the effects and mechanism of power-frequency electromagnetic fields on lipoprotein metabolism and homodynamic during stroke rehabilitation. METHODS: One hundred fifteen patients with stroke were divided into 2 groups, 55 cases of them were treated by exposure to power-frequency electromagnetic fields, 60 cases were treated as control group. Barthel index and Functional Independence Measure (FIM) were used to evaluate rehabilitation outcome. The lipoprotein and its subclasses, homodynamic parameters were compared at pre- and post-rehabilitation. RESULTS: The treatment group showed a statistically significant better prognosis compared with the control group(P<0.01). The score of Barthel index also increased after treatment(P<0.001). Total cholesterol(Tc), triglycerides(TG), low-density lipoprotein cholesterol(LDL-c) levels dropped and high-density lipoprotein cholesterol(HDL-c) increased significantly. The Tc/HDL-c, LDL-c/HDL-c ratio along with the blood and plasma viscosity decreased significantly compared with the controls (P<0.01). The ratio of stroke recurrence decreased significantly after the treatment compared with the control group (P<0.05). CONCLUSION: The power-frequency electromagnetic fields can improve lipoprotein metabolism and homodynamic parameters. It can improve the ADL and FIM of stroke patients, which may have significant implications for stroke patients.

[Soil seed banks of different grassland types of Alashan arid desert region, Inner Mongolia].

Investigations on the soil seed banks of different grassland types of Alashan arid desert region in Inner Mongolia showed that when the altitudes of sampling plots increased from 1,370 m to 1,750 m, the soil seed densities of natural grasslands decreased from 326 grain.m-2 to 76 grain.m-2. At the altitude of 1,100 m, the soil seed density was 56 grain.m-2 in typical desert plot, but only 8 grain.m-2 in the sand dune plot. After the vegetation was restored with Artemisa sphaerocephala on sand dune, its soil seed density increased obviously, almost as 17-29 times as sand dune's. In this study, 31 seeds m-2 were sampled, which belonged to 11 families. Among them, annual plants accounted for 77%, 100% and 70%, perennial plants accounted for 21%, 0 and 16%, and semi-shrubs accounted for 2%, 0 and 14%, respectively for natural grassland, sand dune and sown grassland. Shrub seeds were not found in all the soil seed bank plots. When the altitudes of sampling plots increased from 1,100 m to 1,750 m, the species richness (Margalet index) of soil seed banks of natural grasslands increased from 1.49 to 2.77, and the species diversity (Shannon-Winner index) increased from 0.697 to 0.917.
