Route-dependent effects of the non-NMDA receptor antagonist CNQX on plasma corticosterone levels in mice.

A non-N-methyl-D-aspartate (non-NMDA) receptor antagonist, 6-cyano-7-nitro-quinoxaline-2,3-dione (CNQX), administered intracerebroventricularly (0.1-0.5 microg), significantly inhibited the immobilization stress-induced plasma corticosterone levels in a dose-dependent manner. However, CNQX administered intraperitoneally (1-10 mg/kg) showed a dose-dependent increase of basal plasma corticosterone levels in nonstressed mice and an additive increase of plasma corticosterone levels at the dose of 10 mg/kg in 1 h immobilization-stressed mice. The results suggest that the central and peripheral non-NMDA receptors may be differently involved in the regulation of plasma corticosterone levels in non- and immobilization-stressed mice.

Inhibition of intracerebroventricular injection stress-induced plasma corticosterone levels by intracerebroventricularly administered compound K, a ginseng saponin metabolite, in mice.

Effects of major intestinal metabolites of ginsenosides, including compound K (IH-901, 20-O-beta-D-glucopyranosyl-20(S)-protopanaxadiol), compound Y (IH-902, 20-O-[alpha-L-arabinopyranosyl (1-->6)-beta-D-glucopyranosyl]-20(S)-protopanaxadiol), and ginsenoside Mc (IH-903, 20-O-[alpha-L-arabinofuranosyl (1-->6)-beta-D-glucopyranosyl]-20(S)-protopanaxadiol), on acute stress-induced plasma corticosterone levels were studied in mice. Intracerebroventricularly (i.c.v.) administered compound K (1 microg) attenuated the i.c.v. injection stress-induced increase in plasma corticosterone level, and this inhibitory effect was not affected by co-administered N(G)-nitro-L-arginine methyl ester, a nitric oxide synthase inhibitor. Compound K administered intraperitoneally affected neither the i.c.v. injection stress- nor the immobilization stress-induced increase in plasma corticosterone levels. Compound K and ginsenoside Mc did not affect plasma corticosterone levels induced by the two stress modalities used in this study.

Serotonin depletion enhances the intracerebroventricularly administered MK-801-induced plasma interleukin-6 levels in mice.

To investigate the effect of serotonin depletion on the intracerebroventricularly (i.c.v.) administered MK-801-induced plasma interleukin-6 (IL-6) levels, we pretreated mice with parachlorophenylalanine (PCPA, 300 mg/kg, i.p.) 3 d before an i.c.v. injection of MK-801 (1 microg). The i.c.v. MK-801-induced rise of plasma IL-6 level was markedly enhanced in the PCPA-pretreated mice. However, serotonin depletion by PCPA pretreatment did not affect the i.c.v. MK-801-induced increase in plasma corticosterone level. These results suggest that serotonergic system is involved in the suppressive regulation of MK-801-induced plasma IL-6 level.

Marker-assisted introgression of quantitative trait loci associated with plant regeneration ability in anther culture of Rice (Oryza sativa L.).

A marker-assisted selection (MAS) breeding program was used to improve the plant regenerability of indica rice. A significant quantitative trait loci (QTL) that is associated with the capacity for green plant regeneration in the anther culture of rice was mapped on chromosome 10 using recombinant inbred (RI) population from Milyang 23/Gihobyeo. The marker that was chosen to follow the QTL region was used in MAS. This marker co-segregated with the regeneration ability in F2 individuals that were derived from MGRI 079/IR 36. In order to clarify the relationship between this marker and plant regenerability, the backcross population was screened with a RFLP marker. The capacity of plant regeneration of the backcross population was clearly distinguished by the marker genotype. The development of near isogenic line (NILs) with high regenerability through MAS will save time, labor, and cost in indica rice breeding.

Sesquiterpenes with hepatoprotective activity from Cnidium monnieri on tacrine-induced cytotoxicity in Hep G2 cells.

Bioassay-guided fractionation of the EtOH extract of Cnidium monnieri (Apiaceae) furnished two hepatoprotective sesquiterpenes, torilin (1) and torilolone (2), together with a new derivative, 1-hydroxytorilin (3). Compounds 1 and 2 showed hepatoprotective effects on tacrine-induced cytotoxicity in human liver-derived Hep G2 cells. The EC50 values of compounds 1 and 2 were 20.6 +/- 1.86 (P < 0.01) and 3.6 +/- 0.1 (P < 0.01) microM, respectively. Silybin as a positive control showed an EC50 value of 69.0 +/- 3.4 microM.
