Specific cleavage of Mcl-1 by caspase-3 in tumor necrosis factor-related apoptosis-inducing ligand (TRAIL)-induced apoptosis in Jurkat leukemia T cells.

Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) induces programmed cell death through the caspase activation cascade and translocation of cleaved Bid (tBid) by the apical caspase-8 to mitochondria to induce oligomerization of multidomain Bax and Bak. However, the roles of prosurvival Bcl-2 family proteins in TRAIL apoptosis remain elusive. Here we showed that, besides the specific cleavage and activation of Bid by caspase-8 and caspase-3, TRAIL-induced apoptosis in Jurkat T cells required the specific cleavage of Mcl-1 at Asp-127 and Asp-157 by caspase-3, while other prototypic antiapoptotic factors such as Bcl-2 or Bcl-X(L) seemed not to be affected. Mutation at Asp-127 and Asp-157 of Mcl-1 led to cellular resistance to TRAIL-induced apoptosis. In sharp contrast to cycloheximide-induced Mcl-1 dilapidation, TRAIL did not activate proteasomal degradation of Mcl-1 in Jurkat cells. We further established for the first time that the C-terminal domain of Mcl-1 became proapoptotic as a result of caspase-3 cleavage, and its physical interaction and cooperation with tBid, Bak, and voltage-dependent anion-selective channel 1 promoted mitochondrial apoptosis. These results suggested that removal of N-terminal domains of Bid by caspase-8 and Mcl-1 by caspase-3 enabled the maximal mitochondrial perturbation that potentiated TRAIL-induced apoptosis.

Effect of impaired glucose uptake on postexercise glycogen repletion in skeletal muscles of insulin-treated streptozotocin-diabetic fasted rats.

During recovery from intense exercise performed while fasting, the replenishment of muscle glycogen stores from glucose requires the activation of glucose transport. This study examines if insulin-treated streptozotocin (STZ) diabetes in rats impairs the rate of muscle glucose utilization and glycogen repletion when no food is ingested during recovery from high-intensity exercise. Rats fasted for 24 hours were injected with high doses of STZ (150 mg/kg) to cause severe diabetes, and their glycemia was normalized for 10 days with twice-daily insulin injections. High-intensity exercise in these rats resulted in a marked increase in plasma glucose, which remained higher than preexercise levels thereafter, whereas in control animals, the rise in glycemia was only of a short duration. During recovery, the rates of 2-deoxy-[(3)H]glucose utilization in muscles rich in fast twitch red fibers (red and mixed gastrocnemius muscles) were much lower in STZ-diabetic than in control rats, but were not affected by diabetes in muscles comprised mainly of fast twitch white fibers (white gastrocnemius muscle). Despite these effects on glucose utilization, STZ diabetes had no inhibitory effect on the rate and extent of glycogen deposition and fractional velocities of glycogen synthase across all muscles. In conclusion, although insulin-treated STZ diabetes in fasted rats inhibits glucose transport rates in fast twitch red muscle fibers post-intense exercise, this has no effect on muscle glycogen repletion either because glucose transport does not control the rate of glycogen synthesis or because of a compensatory increase in the activity of lactate glyconeogenesis in these muscles.

[The clinical significance of interphase fluorescence in situ hybridization monitoring chimeric status after sex-mismatched allogeneic hematopoietic stem cell transplantation for leukemia].

OBJECTIVE: To explore the association between chimerism, minimal residual disease (MRD) and relapse after sex-mismatched allogeneic hematopoietic stem cell transplantation (allo-HSCT) for leukemia. METHODS: Fifty-seven patients with leukemia received allogeneic hematopoietic stem cell grafts from HLA-matched or partially matched, but sex-mismatched donors. Chimeric status and MRD were detected by dual-color interphase fluorescence in situ hybridization (I-FISH) using X/Y sex chromosome centromere DNA probe and bcr/abl dual fusion DNA probe, respectively, at different time points after transplantation. SPSS software was used to analyse the correlation between chimeric status, MRD and relapse. RESULTS: In comparison with karyotype analysis, I-FISH was of higher sensitivity in detecting sex chromosome and bcr/abl fusion gene. Chimeric status was negatively correlated with MRD (r=-0.9690, P<0.01). In the early times of transplantation (within 3 months), mixed chimerism had higher relapse rate than did complete chimerism. Chimeric status and MRD were correlated with leukemic relapse (r=-8240, P<0.01; r=-0.9040, P<0.01). The decrease in chimeric status occurred before leukemic relapse in hematology. CONCLUSION: I-FISH is a more specific and sensitive test for monitoring MRD after transplantation. The clinical value of sex chromosome is identical to that of the special tumor gene for monitoring MRD after transplantation. Chimeric status is negatively correlated with MRD. Chimeric status and MRD are associated with leukemic relapse. The decrease in chimeric status is considered a mark of leukemic relapse after transplantation.

[Change of free hemoglobin content after mixing type B whole blood with different amounts of type O whole blood].

To investigate the changes of free hemoglobin (FHb) content after mixing type B whole blood with different amounts of type O whole blood at room temperature and at 37 degrees C, two lots of type B whole blood stored at 4 degrees C for 24 hours were randomly taken as recipient blood, and were packed as 60 ml respectively. Type O blood was taken as donor blood. 60 ml type B whole bloods were mixed with different amounts of type O whole blood, i.e. with 9, 12, 15 and 18 ml. The mixed blood was packed into 100 ml plastic blood bags and stored at 37 degrees C or room temperature, shaken once every 15 minutes. Free hemoglobin content was determined for the harvested samples at 1, 2, 4, 8 and 12 hours after store. The results showed that there was no significant elevation of FHb within 12 hours after mixing B whole blood with different amounts of type O whole blood. In another lot, there was obvious difference in FHb after 1 hour store along with the prolongation of store at either room temperature or 37 degrees C. In one lot, there was no difference of FHb (P > 0.05) during 1 - 8 hours of store at room temperature or 37 degrees C, but significant difference at 12 hours of store (P < 0.001). In another lot, there was no difference of FHb (P > 0.05) within 1 hour of store at room temperature and at 37 degrees C, but significant difference during 2 approximately 8 hours of store (P < 0.001). It is concluded that the FHb would not change significantly within 12 hours after type B blood was mixed with 1 200 ml of type O whole blood, but when the mixed blood was placed at room temperature or at 37 degrees C for 8 hours, the FHb content approaches, even exceeds 170.4 mg/L which was observed in the blood stored for 2 days. It suggests that freshly collected blood must be put into refrigerator of 2 approximately 4 degrees C for storing as soon as possible, so as to decrease the catabolism of erythrocyte and the releasing of FHb and other metabolites which are deleterious to the recipients.

[Analysis of gene expression information in the fat and muscle tissues of pig].

An in silico study was developed to detect the gene expression profile of pig fat and muscle tissues by using the porcine EST resources and human gene sequences,and thus provided candidate information for basic genetic analysis in meat quality improvement of pig. In this study,a BLAST search was performed to identify homologies between the cDNA sequences of human genes and the ESTs sequences of pig,and the high homologous records were screened out. Four Java programs were developed to retrieve and collect sequences, and analyze the BLAST alignment results. By statistical analysis, it was found that there were at least 2002 genes expressed in the fat and muscle tissues of pig, and 1087 in the fat tissue, 1205 in the muscle respectively (290 genes co-expressed in the two tissues). The top-ranking records were screened out,and meantime, 114 basic active genes (BAGs) were found to express in the two tissues,80 in the fat tissue and 34 in the muscle tissue respectively. The top 10 records were described in the paper. This study was summarized in relation to current meat quality improvement of pig at molecular level.

[Preparation and identification of monoclonal antibody against heterogeneous nuclear ribonucleoprotein B1].

OBJECTIVE: To establish anti-heterogeneous nuclear ribonucleoprotein B1 monoclonal antibody cell line and to identify immunological activity. METHODS: BALB/c mice were immunized with specific hnRNPB1 polypeptide by multiple-place hypodermic injection. The splenic cells of the mice were fused with SP2/0 mouse myeloma cells. The hybridoma culture supernants were screened by indirect ELISA with artificial synthetic peptide. RESULTS: Two hybridoma cell lines against hnRNPB1 were successfully obtained; the titers of antibody in ascites were 1: 1.8 x 10(5). The McAb showed the character of no cross reactions. Immunohistochemical analysis demonstrated that the antibody could bind with hnRNPB1 specifically. CONCLUSION: The McAb prepared so by this laboratory is of good specificity and sensitivity.

[Growth inhibition and demethylation by a component of natural drug, CDP, in cancer cell lines].

OBJECTIVE: To investigate the inhibition of tumor cell lines' growth and the demethylation of p16 gene by a component of natural drug, CDP. METHODS: Human liver cancer cell line Huh-7 and breast cancer cell line T47D were treated with the CDP and DNA methyltransferase inhibitor, 5-azacytidine (5-aza-C). The inhibition of growth was assayed by MTT; the methylation status of p16 gene promoter was analyzed by MSP. RESULTS: 1. The CpG islands in promoter of p16 gene were identified as completely methylated in Huh-7 and T47D. 2. CDP and 5-aza-C inhibited the proliferation of Huh-7 and T47D cell lines in a dose, time-dependent mode. 3. Methylation-specific bands of CpG islands in p16 gene' promoter were still existed in Huh-7 and T47D, while unmethylation-specific bands appeared in Huh-7 after the cells being treated with 25, 50, 75 and 100 micromol/ L of CDP for 6 days as well as in T47D after the cells being treated with 50, 75 and 100 micromol/L of CDP for 6 days. 4. After the cells being treated with 50 micromol/L of CDP, the methylation-specific bands of CpG island in p16 gene' promoter were still existed in Huh-7 and T47D; unmethylation-specific bands appeared at 12 h and lasted to 144 h in Huh-7 while at 72 h and lasted to 144 h in T47D. CONCLUSION: CDP has inhibition effect on the cancer cell lines and has the ability to reverse the hypermethylated p16 gene promoter. These results suggest that CDP has great potential in the development of effective anticancer drugs.

A facile chemical route to semiconductor metal sulfide nanocrystal superlattices.

We report a facile chemical route for the synthesis of monodisperse nanocrystals of various metal sulfides (PbS, Cu(2)S, and Ag(2)S) and their assemblies into nanocrystal superlattices (NCSs); the sulfides NCSs were precipitated by adding ethanol to nanocrystal colloids, which were obtained directly by a reaction between metal thiolate and thioacetamide in a pure dodecanethiol solvent.

[Hematopoietic stem cell transplantation for patients with chronic myelogenous leukemia].

BACKGROUND & OBJECTIVE: Chronic myelogenous leukemia (CML) is a malignant hematological disease.CML patients are commonly treated with hematopoietic stem cells transplantation (HSCT). This study was designed to evaluate the effect of HSCT on the patients with CML. METHODS: Forty-four patients with CML were treated by HSCT, including 8 cases treated with purging autologous transplantation, 30 cases with related donor allogeneic HSCT (allo-HSCT), and 6 cases with unrelated donor allo-HSCT. The conditioning regimen was TBI (total-body irradiation)+CY (CTX) protocol in 31 patients and modified BuCY (hydroxyurea, busulfan, Ara-C, CTX) protocal in 12 patients, and MACC (Melphalan, Ara-C, CTX and CCNU) protocol in one patient. CsA (cyclosporine) and MTX were used in the patients with related donor allo-HSCT, and CsA and MTX added to mycophenolate mofetil (MMF) and antithymocyte globulin (ATG) were used in the patients with unrelated donor all-HSCT for graft versus host disease (GVHD) prophylaxis. Otherwise, CsA was only used in the patients with accelerated phase(AP) and blast crisis(BC) for GVHD prophylaxis. Kaplan-Meier survival analysis model was used to estimate the overall survival (OS) and the disease-free survival (DSF) at 5 years after transplantation. RESULTS: Eight patients with autologous transplantation, except one case died of transplantation-related-complication, obtained part or complete cytogenetic remission within 3 months after transplantation. One patient, who was BC and obtained complete remission (CR) in hematology before transplantation,obtained complete molecular remission for 81 months after autologous transplantation. All patients obtained CR, except one patient died of hepatic veno-occlusive disease (VOD) and one case did not obtained CR, in 36 patients with allo-HSCT after transplantation. The incidence of infection and VOD during transplantation were 38.6% and 9.1%, respectively. The incidences of hemorrhagic cystitis (HC) and cytomegalovirus (CMV) pneumonia after transplantation were 15.9% and 11.4%, respectively. VOD, HC, and CMV pneumonia did not occur in the patients with autologous transplantation. The incidence of acute GVHD in the patients with related and unrelated donor transplantation were 40.0% and 33.3%, respectively. The incidence of chronic GVHD was 43.4% in the patients with related donor transplantation. The rates of transplant-related mortality (TRM) in the patients with autologous and allogeneic transplantation were 12.5% and 16.7%, respectively. The rates of relapse in patients with autologous and related donor transplantation were 37.5% and 13.3%, respectively. The DFSs at 5 years in patients with autologous and related donor transplantation after transplantation were 18.7% and 53.7%, respectively. The DFS at 5 years in patients with CP (chronic phase) or AP and BC before transplantation were 66.4% and 26.7%, respectively. CONCLUSION: all-HSCT shows higher clinical cure rate to CML patients with CP. CsA+MTX+MMF+ATG protocol is more effective for acute GVHD prophylaxis and can decrease the incidence and severity of acute GVHD in patients with unrelated donor transplantation. Autologous transplantation with bone marrow purged can prolong the survival time and a few patients may be cured with autologous transplantation in CML.

[Morphological and quantatitive capillary changes in aging human brain].

OBJECTIVE: To investigate morphological changes of capillary in aging brain and explore the role of vascular factor in brain aging. METHODS: Twenty-eight brains of individuals (mean age 65 years) who died without clinical or pathological involvement of nervous system and 6 brains of Alzheimer's disease (AD) patients (mean age 83 years) were obtained at autopsy. Sections from frontal lobe, occipital lobe, striatum and hippocampus of normal subjects and sections from hippocampus of AD patients were used for hematoxylin eosin (HE), lox fast blue (LFB), toluidine blue stains and ulex europaeus agglutinin (UEA) immunostaining. After observations of morphological changes of neuron and capillary, computer-aid image analysis was performed to quantify numerical density and area density of neuron and capillary in frontal lobe, occipital lobe, putamen, CA3 sector of normal subjects and CA3 sector of AD patients. Numerical ratio and area ratio of neuron and capillary were then calculated. Correlations between neuron/capillary ratio and age were estimated using Pearson's correlation test. Difference of neuron/capillary ratio in CA3 sectors between AD patients and advanced aged normal subjects (> 75 years) was analyzed with Student's t-test. RESULTS: Several pathological microvascular changes, including increased tortuosity, looping, bundling, stringing, and effacement of endothelia were seen in aged subjects and more prevalent in AD patients. Numerical ratio and area ratio of neuron and capillary of frontal lobe, occipital lobe and putamen significantly increased with age in normal aging subjects. CONCLUSIONS: Morphological changes and relative decrease in number and capacity of capillary in aging brain may reduce cerebral blood flow and metabolism, and consequently result in functional impairment of aging brain. Vascular factors may play an important role in the development of brain aging.

[Effects of allogeneic hematopoietic stem cell transplantation with very-high-dose conditioning regimen for refractory leukemia].

OBJECTIVE: To explore the regimen-related toxicity (RRT) and therapeutic effects of very-high-dose conditioning regimen combined with induction of graft-versus-leukemia (GVL) effects in allogeneic hematopoietic stem cell transplantation (allo-HSCT) for refractory leukemia with unattainable complete remission (CR) before transplantation. METHODS: Eighteen patients who failed to obtain CR before transplantation received very-high-dose conditioning regimen protocol (experimental group), and 62 patients with acute leukemia with CR or with chronic myeloid leukemia in the chronic phase before transplantation received total body irradiation plus cyclophosphamide (CTX) or modified BuCY (hydroxyurea, busulfan, Ara-C, CTX) protocol (control group). In patients with refractory leukemia who did not develop graft-versus-host disease (GVHD) 30 d after the transplantation, GVL was induced by rapid reduction of the dosage of cyclosporin A or by donor lymphocytic infusion. The incidence and mortality of RRT and the rates of CR, GVHD and leukemia relapse after transplantation were investigated. Kaplan-Meier survival analysis model was used to estimate the disease-free survival (DFS) rate at 3 years post-transplantation. RESULTS: Except for one patient in the experimental group and two in the control group who died of transplantation- related complications, all the other patients obtained hematopoietic reconstitution. The total incidence of RRT was 100% in both groups, involving most frequently the stomach and intestines at the rate as high as 83.3% in the experimental group and 85.5% in the control group. RRT involving the oral cavity occurred in 44.4% and 62.9%, and that involving the bladder in 16.7% and 33.9% of the cases in the experimental group and control group, respectively, all similar between the groups (P=0.823, 0.172 and 0.244, respectively). The RRT mortality was 0 and 5% in the experimental and control groups, respectively (P=0.341). With the exception of one patient who died of infection, all the other patients treated with very-high-dose conditioning regimen obtained CR. The incidences of acute/chronic GVHD were 58.8%/92.6% and 40.0%/55.8%, respectively, in the experimental and control groups. The incidence of leukemia relapse was 11.8% and 18.3%, and DFS at 3 years after transplantation was (61.2+/-12.3)% and (65.0+/-7.4)% (P=0.6311) in the two groups, respectively. CONCLUSION: Consecutive very-high-dose conditioning regimen combined with GVL induction after transplantation can increase the rate of CR and DFS, without increasing RRT incidence and mortality in allo-HSCT for the refractory leukemia with unattainable CR pre-transplantation.

[A study on difference of ocular motor parameters in myopia].

OBJECTIVE: This study sought to assess the difference of these ocular motor parameters in myopia onset and development were explored. METHODS: 31 emmetropia and 46 myopia adult subjects were included in this study. The myopia subjects were divided into three groups as mild myopia, moderate myopia and high myopia. Phoria at distance and near, positive and negative relative accommodation (NRA/PRA) were measured. Accommodative convergence and accommodation stimulus ratio (AC/A) was determined through phoria values' calculation. RESULTS: Calculated AC/A of myopia was higher than that of emmetropia, and there were trends of elevated AC/A with myopia power increasing. There was significant difference in phoria in near between the myopia group and emmetropia group, but none in that of distance. There was no significant difference in NRA. However, PRA of myopia was lower than that of the emmetropia (P <0.05). CONCLUSIONS: Ocular motor parameters are different in myopia eyes. AC/A is one of key parameters among them, and AC/A is increased in myopia, which indicates out of focus of retinal image in the developing myopia.

[The effects of anti-inflammatory on activation of nuclear factor-kappaB and expression of cell adhesion molecules in patients with ulcerative colitis].

The purpose of this study is to assess the effects of anti-inflammatory on activation of nuclear factor-kappaB and mRNA and protein expression of intercellular adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecule-1 (VCAM-1) in intestinal mucosal biopsy specimens from patients with ulcerative colitis (UC). A total of 27 cases with UC were investigated. 15 cases received sulfasalazine (SASP) treatment or SASP and glucocorticoid treatment, 12 cases did not receive any medication related with UC. Normal mucosa from 9 colon cancer cases served as control. Ten pieces of intestinal mucosal biopsy specimens were obtained from each patient. The mRNA expression of ICAM-1 and VCAM-1 were determined by reversal transcription-polymerase chain reaction (RT-PCR). The protein levels of ICAM-1 and VCAM-1 were measured by enzyme linked immunosorbent assay (ELISA). NF-kappaB DNA binding activity was evaluated by electrophoretic mobility shift assay (EMSA). The results showed that NF-kappaB DNA binding activity, mRNA and protein expression of ICAM-1 and VCAM-1 were increased significantly in patients with UC, compared with normal control (P<0.05). Glucocorticoids and SASP markedly inhibited NF-kappaB activation and significantly decreased mRNA and protein expression of ICAM-1 and VCAM-1 (P<0.05). Adhesion molecules (ICAM-1 and VCAM-1) gene activation had significant positive correlation with the NF-kappaB DNA binding activity (r=0.8652 P<0.05, r=0.7902, P<0.05, respectively). We concluded that NF-kappaB is a major and essential factor in regulating the expression of adhesion molecules, it plays an important role in the pathogenesis of UC. SASP and glucocorticoids ameliorate UC via inhibition of NF-kappaB activation and reduction of adhesion molecules expression.

[Therapeutic efficacy of hematopoietic stem cell transplantation in patients with chronic myelogenous leukemia].

OBJECTIVE: To evaluate the clineffects of autologous and allogeneic hematopoietic stem cell transplantation (HSCT) for chronic myelogenous leukemia (CML). METHODS: Fifty-seven patients with CML were treated by HSCT, including 8 patients treated with autologous transplantation in vivo and vitro purging minimal residual disease (MRD), 39 with related donor allogeneic HSCT (allo-HSCT), and 10 with unrelated donor allo-HSCT. For the conditioning regimen, total-body irradiation with cyclophosphamide (CTX) was given in 32 patients, modified BuCY protocol (hydroxyurea, busulfan, Ara-C, CTX) in 24 patients, and MACC protocol (melphalan, Ara-C, CTX and lomustine) in one patient. Cyclosporine (CsA) and methotrexate (MTX) were used in patients with related donor allo-HSCT, and the combination of CsA, MTX , mycophenolate mofetil (MMF), and antithymocyte globulin (ATG) in unrelated donor all-HSCT to prevent graft versus host disease (GVHD). Kaplan-Meier survival analysis model was used to estimate the overall survival and the disease-free survival (DFS) at 5 years after transplantation. RESULTS: In 8 patients with autologous transplantation, 7 obtained partial or complete cytogenetic remission (CR) within 3 months after transplantation and one died of transplantation-related complication. In 49 patients with allo-HSCT transplantation, all patients obtained CR except for two patients, one of whom failed to obtain CR and the other died of hepatic veno-occlusive disease. The incidence of infection and veno-occlusive disease during transplantation was 33.3% and 7.0%, respectively. The incidence of hemorrhagic cystitis and cytomegalovirus interstitial pneumonia after transplantation was 22.8% and 8.8%, respectively. Veno-occlusive disease, hemorrhagic cystitis or cytomegalovirus interstitial pneumonia did not occur in patients with autologous transplantation. The incidence of acute GVHD was 41.0% and 48.6%, and that of chronic GVHD 40.0% and 42.9% in patients with related and unrelated transplantation, respectively. The rate of relapse was 57.1% and 12.8%, with DFS at 5 years of 25.0% and 61.7%, respectively, in patients with autologous and related donor transplantation. The DFS at 5 years was 70.7% and 34.1%, respectively, in patients with chronic/accelerated phases and blast crisis be fore trans plantation. CONCLUSION: allo-HSCT can produce a higher clinical cure rate in CML patients in chronic phase CsA+MTX+MMF+ ATG protocol is more effective for prevention and alleviation of acute GVHD in patients with unrelated donor transplantation. Autologous transplantation with bone marrow purging helps prolong the patients' survival and even obtain clinical cure of CML.

[Research on the growth inhibition of T47D cell and reversion of p16 hypermethylation by CDP].

OBJECTIVE: To inquire into the mechanism of CDP (a purified component of Chinese crude drug) in inhibiting the breast cancer T47D cell growth. METHODS: T47D cells were treated with different concentration of 5-azacytidine (5-aza-CR) and CDP for several days. The growth rate was assessed by cell proliferation experiment (MTT colorimetric assay). The changes in apoptic peak and cell cycle distribution were detected by flow cytometry (FCM). The levels of methylation and unmethylation status of p16 were detected by methylation specific PCR (MSP). RESULTS: After treatment with the two drugs, the cell growth rate decreased in a dose and time dependent manner (P<0.05). The cell cycle was influenced by the well-chosen concentration of 5-aza-CR (2 micromol/L) and CDP (50 micromol/L): the cell number increased from 65.1% to 71.3%, 84.3% in G0/G1 phase and decreased from 19.4% to 14.3%, 7.2% in S phase. Demethylation on p16 gene occurred after treatment with any of the two drugs for 6 days. CONCLUSION: CDP can reverse p16 hypermethylation and may hence inhibit the proliferation of tumor cells.

[Autologous transplantation of recombinant interleukine-2 activated bone marrow for acute promyelocytic leukemia].

OBJECTIVE: To explore the benefit of autologous transplantation of interleukine-2 activated bone marrow (ABM) in acute promyelocytic leukemia (APL) as consolidation therapy. METHODS: 31 patients with APL, 27 in first complete remission (CR1), 3 in CR2, and 1 in partial remission (PR) after the second relapse, were treated with autologous transplantation of ABM. The conditioning regimens included MACC protocol (Melphalan, Ara-C, CTX, and CCNU) in 26 patients and TBI + CY protocol in 5 patients. The PML/RARa fusion gene was measured by fluorescence in situ hybridization. Kaplan-Meier survival analysis model was used to estimate the disease-free survival (DFS) rate at 5 years post-transplantation and COX regression model was used to analyze the DFS-influencing factors, including sex, PML/RARa positive state, pre-transplantation state (CR1, CR2, or PR), white blood cell (WBC) count, and platelet (PBC) count. RESULTS: The 27 patients in CR1 had a DFS time of 3 to 113 months (with a mean DFS time of 46 months), and a 5-year DFS rate of 100%; none of them suffered a relapse after transplantation. One of the 3 patients in CR2 relapsed in 19 months after the transplantation and the other two patients had been in DFS state for 7 and 33 months respectively. The patient in PR obtained CR and relapsed 8 months after the transplantation. All the patients showed reconstitution of hematopoiesis. None of the patients died of transplantation-related complication. Multivariate analysis showed that long-term survival was correlated with the pre-transplantation statues and not with sex, positivity of PML/RARa, and WBC and PBC counts. CONCLUSION: Autologous transplantation of ABM reduces the relapse and increases the long-term survival in the patients with APL in CR, especially in CR1.

Cerebral amyloid angiopathy in aged Chinese: a clinico-neuropathological study.

To investigate the prevalence and clinico-neuropathological characteristics of cerebral amyloid angiopathy (CAA) in aged Chinese and its relationship to dementia and cerebrovascular lesions, we examined 362 archived brains of elderly with immunohistochemical staining for beta-amyloid peptide and Congo red, Bodian and Luxol fast blue stains. We found that: (1) CAA appeared in 31.7% examined brains without sexual preponderance, and the incidence increased with age; (2) the frontal lobe was most frequently involved in CAA, followed by occipital and parietal lobe; (3) subcortical white matter and cerebellum dentate nucleus areas may also be affected by CAA; (4) CAA has a close relationship to Alzheimer's disease and multiple cerebrovascular lesions; (5) CAA alone may result in dementia.
