Three lycopodium alkaloid N-oxides from Huperzia serrata.

Three lycopodium alkaloid N-oxides, huperzine J (1), huperzine K (2) and huperzine L (3) were obtained from Huperzia serrata (Thunb.) Trev. Their structures were elucidated by spectroscopic methods and the relative configurations of 1 and 3 were established via NOESY NMR observations; optical rotation values and CD data are presented.

Preliminary study on the cleavage of fusion protein GST-CMIV with palladium(II) complex.

A novel method for post-treatment of gene-engineered proteins is reported. A coden of Cys-His unit is introduced into the N-terminal of cecropin CMIV by using PCR. The gene is expressed in E. coli fused with GST. After purification, the fusion protein is cleaved by [Pd(en)(H2O)2]2+ at the His-Arg bond and the cecropin CMIV with antibacterial activity is obtained. The preliminary results held some promise of success for application of the palladium(II) complex as cleavage agent for the production of peptide drugs from gene-engineering fusion proteins.

A three-dimensional, two-way, parabolic equation model for acoustic backscattering in a cylindrical coordinate system

A new PE model for solving three-dimensional, forward and backward sound propagation in a cylindrical coordinate system is presented. The model marches a wave field in the radial direction including the azimuthal diffraction effects, and solves for a backscattered field based on a three-dimensional, single scattering approach. A periodic sidewall boundary condition is applied for computations in a 360-degree sector, while an approximate sidewall boundary condition is used for calculation in a sector less than 360 degrees. These two sidewall boundary conditions are verified by the numerical results. The major drawback of using the cylindrical coordinate system, when the backscattering solution is valid within a limited area, is analyzed using a geometrical-optical interpretation. The model may be useful for studying three-dimensional backscattering phenomena comprising azimuthal diffraction effects.

Interleukin-12 as an adjuvant promotes immunoglobulin G and type 1 cytokine recall responses to major surface protein 2 of the ehrlichial pathogen Anaplasma marginale.

Anaplasma marginale is a tick-transmitted pathogen of cattle closely related to the human ehrlichiae, Ehrlichia chaffeensis and the agent of human granulocytic ehrlichiosis (HGE). These pathogens have in common a structurally conserved outer membrane protein (OMP) designated the major surface protein 2 (MSP-2) in A. marginale and HGE and OMP-1 in E. chaffeensis. Protective immunity against ehrlichial pathogens is believed to require induction of gamma interferon (IFN-gamma) and opsonizing immunoglobulin (Ig) subclasses directed against OMP epitopes that, in concert, activate macrophages for phagocytosis and killing. Because interleukin-12 (IL-12) acts as an adjuvant for protein immunization to induce IFN-gamma and protective immunity against intracellular pathogens, we hypothesized that as an adjuvant with MSP-2, IL-12 would augment type 1 recall responses to A. marginale. IL-12 was coadsorbed with MSP-2 to alum and shown to significantly enhance IFN-gamma production by lymph node cells (LNC) and LNC-derived CD4(+) T-cell lines from immunized calves following recall stimulation with A. marginale. LNC proliferation and IL-2 production were also enhanced in IL-12-treated calves. Elevated recall proliferative responses by peripheral blood mononuclear cells were still evident 9 months after immunization. Serum IgG levels were consistently increased in IL-12 immunized calves, predominantly due to higher IgG1 responses. The results support the use of IL-12 coadsorbed with OMP of ehrlichial pathogens in alum to amplify both antibody and type-1 cytokine responses important for protective immunity.

Hypoxic pulmonary vasoconstriction is modified by P-450 metabolites.

20-Hydroxyeicosatetraenoic acid (20-HETE) is a cytochrome P-450 4A (CYP4A) metabolite of arachidonic acid (AA) in human and rabbit lung microsomes and is a dilator of isolated human pulmonary arteries (PA). However, little is known regarding the contribution of P-450 metabolites to pulmonary vascular tone. We examined 1) the effect of two mechanistically distinct omega- and omega1-hydroxylase inhibitors on perfusion pressures in isolated rabbit lungs ventilated with normoxic or hypoxic gases, 2) changes in rabbit PA ring tone elicited by 20-HETE or omega- and omega1-hydroxylase inhibitors, and 3) expression of CYP4A protein in lung tissue. A modest increase in perfusion pressure (55 +/- 11% above normoxic conditions) was observed in isolated perfused lungs during ventilation with hypoxic gas (FI(O(2)) = 0.05). Inhibitors of 20-HETE synthesis, 17-oxydecanoic acid (17-ODYA) or N-methylsulfonyl-12,12-dibromododec-11-enamide (DDMS), increased baseline perfusion pressure above that of vehicle and amplified hypoxia-induced increases in perfusion pressures by 92 +/- 11% and 105 +/- 11% over baseline pressures, respectively. 20-HETE relaxed phenylephrine (PE)-constricted PA rings. Treatment with 17-ODYA enhanced PE-induced contraction of PA rings, consistent with inhibition of a product that promotes arterial relaxation, whereas 6-(20-propargyloxyphenyl)hexanoic acid (PPOH), an epoxygenase inhibitor, blunted contraction to PE. Conversion of AA into 20-HETE was blocked by 17-ODYA, DDMS, and hypoxia. CYP4A immunospecific protein confirms expression of CYP4A in male rabbit lung tissue. Our data suggest that endogenously produced 20-HETE could modify rabbit pulmonary vascular tone, particularly under hypoxic conditions.

Epoxyeicosatrienoic acids constrict isolated pressurized rabbit pulmonary arteries.

Little information is available regarding the vasoactive effects of epoxyeicosatrienoic acids (EETs) in the lung. We demonstrate that 5, 6-, 8,9-, 11,12-, and 14,15-EETs contract pressurized rabbit pulmonary arteries in a concentration-dependent manner. Constriction to 5,6-EET methyl ester or 14,15-EET is blocked by indomethacin or ibuprofen (10(-5) M), SQ-29548, endothelial denuding, or submaximal preconstriction with the thromboxane mimetic U-46619. Constriction of pulmonary artery rings to phenylephrine is blunted by treatment with the epoxygenase inhibitor N-methylsulfonyl-6-(2-propargyloxyphenyl)hexanamide. Pulmonary arteries and peripheral lung microsomes metabolize arachidonate to products that comigrate on reverse-phrase HPLC with authentic regioisomers of 5,6-, 8,9-, 11,12-, and 14,15-EETs, but no cyclooxygenase products of EETs could be demonstrated. Proteins of the CYP2B, CYP2E, CYP2J, CYP1A, and CYP2C subfamilies are present in pulmonary artery and peripheral lung microsomes. Constriction of isolated rabbit pulmonary arteries to EETs is nonregioselective and depends on intact endothelium and cyclooxygenase, consistent with the formation of a pressor prostanoid compound. These data raise the possibility that EETs may contribute to regulation of pulmonary vascular tone.

N-[2-(4-methylphenyl)ethyl]-N'-[2-(5-bromopyridyl)]-thiourea as a potent inhibitor of NNRTI-resistant and multidrug-resistant human immunodeficiency virus type 1.

The composite non-nucleoside reverse transcriptase inhibitor (NNRTI) binding pocket model was used to study a number of thiourea analogues with different substitutions at the 4-phenyl position including N-[2-(4-methylphenyl)ethyl]-N'-[2-(5-bromopyridyl)]-thiourea (compound HI-244), which inhibited recombinant RT better than trovirdine or compound HI-275 with an unsubstituted phenyl ring. HI-244 effectively inhibited the replication of HIV-1 strain HTLV(IIIB) in human peripheral blood mononuclear cells with an IC50 value of 0.007 microM, which is equal to the IC50 value of trovirdine. Notably, HI-244 was 20 times more effective than trovirdine against the multidrug-resistant HIV-1 strain RT-MDR with a V106A mutation (as well as additional mutations involving the RT residues 74V, 41L and 215Y) and seven times more potent than trovirdine against the NNRTI-resistant HIV-1 strain A17 with a Y181C mutation.

[The application of polymerase chain reaction-single strain conformation polymorphism in the pedigree analysis of familial hypercholerolemia patients].

OBJECTIVE: To discuss the value of polymerase chain reaction-single strand conformation polymorphism(PCR-SSCP) in the pedigree analysis of familial hypercholesterolemia(FH) patients. METHODS: For 4 patients with FH detected by PCR-SSCP and DNA sequence analysis (1 homozygote has point mutation in exon 7, 1 heterozygote in exon 14, and 2 heterozygotes in 3' part of exon 4 ), members of each pedigree, altogether 23 individuals, were analysed using PCR-SSCP. RESULTS: Every member of the 4 pedigrees was examined genetically. Besides the 4 probands, another 1 homozygote and 8 heterozygotes were found. CONCLUSION: The PCR-SSCP method can be used in the pedigree analysis of FH probands. The early diagnosis of siblings and relatives can help to provide genetic consultation and direction so as to pay attention to development of hypercholesterolemia.

[Application of bio-electrical impedance analysis in the compensation period of intracranial pressure increasing].

In the compensation period of intracranial pressure increasing, brain impedance rises continuously while cerebrospinal fluid volume changes. Along with the illness progressing, intracranial pressure would rise obviously, but the intracranial component relative volume would go near to constant; at the same time, brain impedance fluctuates around a baseline and there appears an inflection point on the impedance track record curve after the compensation period.

Effect of stimulation of shenshu point on the aging process of genital system in aged female rats and the role of monoamine neurotransmitters.

In this experiment, among some aged female rats aged over 18 months, and young female rats aged 3 months whose central noradrenergic nerve endings were injured by ventricular injection of 6-hydroxy-dopamine (6-OHDA), it was observed that catgut embedding at bilateral Shenshu (UB 23) points could obviously shorten sexual cycles, increase the frequency of sexual cycle, and slow down the aging process of the genital system in both the aged rats and in the rats with injured noradrenergic endings. After electroacupuncture (EA) at Shenshu (UB 23) points in the aged rats, the frequency of neuronal discharges in locus coeruleus (LC) was elevated and the activating rate of LC to neurons in the medial preoptic area (MPOA) of the hypothalamus was increased, while obvious effect on nucleus raphes magnus (NRM) and the effect of NRM on MPOA were not marked. It is suggested that stimulation of Shenshu (UB 23) point can strengthen the excitability of noradrenergic neurons, activate the ascending pathway of the brain stem--hypothalamus, raise the catecholamine (CA)/5-hydroxytryptamine (5-HT) ratio in the hypothalamus of the aged rats, so as to delay the aging process of the genital system.

Effects of carbon tetrachloride-injured hepatocytes on hepatic stellate cell activation and salvianolic acid A preventive action in vitro.

OBJECTIVE: To investigate the effects of carbon tetrachloride (CCl(4))-injured hepatocyte on hepatic stellate cell (HSC) activation, the relation between lipid peroxidation and liver fibrosis, and the action mechanisms of salvianolic acid A (SA-A), one of water soluble components extracted from radix salvia miltiorrhizae, against HSC activation and liver fibrosis. METHODS: The hepatocytes were isolated from the normal rats, incubated with SA-A at different concentrations, respectively, and vitamin E severed as control. At the same time the cells were fumigated with CCl(4) for 24h, and ALT activities in the medium were measured. After the cell medium was changed and cultured for another 24h, the medium was collected as "hepatocyte conditioned medium" (HCM) and measured for malondiadehyde (MDA) content. Then HCM was incubated with primary cultured HSC for 48h, and HSC proliferation, type I collagen gene expression and protein production were observed. RESULTS: ALT activity and MDA content in the hepatocyte medium were increased remarkably after the cell was injured. HCM could obviously promote HSC proliferation, type I collagen mRNA expression and protein production. SA-A could markedly inhibit ALT activity and MDA content in hepatocytes, and decreased collagen gene expression and protein production. CONCLUSION: The secretion from the peroxidative injured hepatocyte could stimulate HSC activation, SA-A could decrease activation by alleviation of hepatocyte peroxidation.

[Bacterial resistance in streptococcus pneumoniae].

OBJECTIVE: To investigate the prevalence of Streptococcus pneumoniae in patients with respiratory tract infection and healthy children and to determine the antibiotic susceptibility in S. pneumoniae. METHODS: Sputum and throat swabs were collected from the adult and children patients. Nasopharyngeal specimens were collected from healthy children with aseptic swab and cultured for S. pneumoniae. Antibiotic susceptibility was determined by agar dilution method. RESULTS: The isolation rates of S. pneumoniae were 10% in both children and adult out-patients, being 30.2% in 2-6 year-old healthy children. Among 82 clinical isolates, only one was low-level resistant to penicillin (MIC 0.125 mg/L). In 228 S. pneumoniae strains from healthy children, 32 were low-level resistant to penicillin (MIC 0.125-0.5 mg/L), the resistant rate being 14.0%. S. pneumoniae was usually resistant to erythromycin, clindamycin and SMZ-TMP. CONCLUSION: The penicillin-resistant strains are far more resistant to all the agents tested than susceptible strains except fluoroquinolones and vancomycin; in addition, several strains resistant to the third generation cephalosporins are found.

[Introduction to management of environmental sanitation in public concession in Shanghai in 1930s].

The environmental sanitation in Shanghai public concession is one of the regions with better administration. A complete set of administration rules was proposed by the Hygienic Department and Logistic Department of its public Security Bureau. It also spent a wealth of money to organize personnel and material supply to treat its environmental sanitation. This experience is worthy of summarization.

[Neurofibromatosis type 2].

OBJECTIVE: To investigate the clinical characteristics and management strategies of bilateral acoustic neuromas. METHODS: The data of 7 patients with bilateral acoustic neuromas collected between 1990 to 1998 were retrospectively analyzed. RESULTS: Altogether 122 patients with acoustic tumors were treated from 1990 to 1998, in which 7 cases (5.8%, 6 male and 1 female) had bilateral acoustic neuromas. The age at onset of symptoms ranged from 13 to 60 years (average 29.1 years). Progressive hearing loss and tinnitus were the initial symptoms in 4 cases. Either strabismus, ptosis, headache or dysequilibrium was presented in 4 cases. Six cases complicated tumors in the central nervous system and/or other sites. Five cases had cafe au lait spots. One case had posterior subcapsular lenticular opacity. Four cases fell into severe (Wishart) type and 2 into mild (Gardner) type. The tumors were unilaterally removed in 4 patients through the retrolabyrinthine approach (1 case) or the retrosigmoid approach (3 cases). In these 4 patients, one died of central respiratory failure after the operation; two had contralateral tumor removal through retrosigmoid approach 3 weeks after the first surgery. One of the patients died of encephaledema after the surgery. No hearing impairment and facial nerve paralysis occurred in one case operated on through the retrolabyrinthine approach, whereas in those through retrosigmoid approach, 4 ears had hearing loss and 3 sides had facial nerve paralysis. CONCLUSION: The clinical characteristics and treatment strategies for bilateral acoustic neuromas are different from those of unilateral acoustic neuroma. Individualization of management is a prerequisite for the success of the treatment. To avoid injury to the VII and VIII cranial nerves, monitoring the nerve functions during the surgery is important.

N'-[2-(2-thiophene)ethyl]-N'-[2-(5-bromopyridyl)] thiourea as a potent inhibitor of NNI-resistant and multidrug-resistant human immunodeficiency virus-1.

The thiophene-ethyl thiourea (TET) compound N'-[2-(2-thiophene)ethyl]-N'-[2-(5-bromopyridyl)]-thiourea (compound HI-443) was five times more potent than trovirdine, 1250 times more potent than nevirapine, 100 times more potent than delavirdine, 75 times more potent than MKC-442, and 50 times more potent than AZT against the multidrug resistant HIV-1 strain RT-MDR with a V106A mutation. HI-443 was almost as potent against the NNI-resistant HIV-1 strain A17 with a Y181C mutation as it was against HTLV(IIIB). The activity of HI-443 against A17 was ten times more potent than that of trovirdine, 2083 times more potent than that of nevirapine, and 1042 times more potent than that of delavirdine. HI-443 inhibited the replication of the NNI-resistant HIV-1 strain A17 variant with Y181C plus K103N mutations in RT with an IC50 value of 3.3 microM, whereas the IC50 values of trovirdine, nevirapine, and delavirdine were all >100 microM. These findings establish the novel thiophene containing thiourea compound HI-443 as a novel NNI with potent antiviral activity against NNI-sensitive, NNI-resistant and multidrug-resistant strains of HIV-1.
