Role of the two-component signal transduction and the phosphoenolpyruvate: carbohydrate phosphotransferase systems in competence development of Haemophilus influenzae Rd.

Haemophilus influenzae Rd becomes competent for transformation by nutritional downshift or transient anaerobic growth through a process that requires cyclic AMP receptor protein and adenylate cyclase. Insertion mutations in crr or ptsI of the phosphoenolpyruvate:carbohydrate phosphotransferase system lowered transformation frequencies, and the effect was reversed by the addition of cyclic AMP. However, insertions into H. influenzae homologs of two-component signal transduction genes had no effect on competence.

[Calcification mechanism and anticaicification on cardial bioprostheses].

The high incidence of calcification in cardiac bioprostheses greatly shortens their service life. In this study, we found that the calcification of cardiac bioprostheses was related the content of free carboxyl groups in collagen molecule of cardiac bioprostheses. The method of chemical modification of glutaraldehyde (GA) treated cardiac bioprostheses with epoxy chloropropane (EC) was selected. The content of free carboryl groups in collagen molecule was reduced greatly in EC-modified valve tissues, and the calcified content of EC-modified porcine aortic valve, bovine pericardial and porcine pericardial valves was reduced greatly at 2, 4, 6 weeks after subcutaneous implantation in young New Zealand rabbits. There were good physical stability, mechanical tensile strength and no immunologic reactions against the valve in EC modified valves. These findings showed that the anti-calcified valve can be used in clinic.

Undetected HIV infection among patients admitted to an alcohol rehabilitation unit.

Of 300 patients admitted to an alcohol rehabilitation unit, serological testing of discarded admission blood samples revealed that 31 (10.3%) had HIV infection. Chart records indicated that four patients were known to be HIV infected on admission, and HIV infection was detected in another three patients during hospitalization; however, 24 (77.4%) of the HIV-positive patients were discharged with their HIV infection still undetected.

Comparison of HLA class II genes in Caucasoid, Chinese, and Japanese patients with primary Sjögren's syndrome.

To better define the genetic factors that predispose to primary Sjögren's syndrome (SS), we have used polymerase chain reaction in combination with oligonucleotide probe hybridization and DNA sequencing to analyze HLA-DRB1, -DQA1, -DQB1, and -DPB1 alleles in Caucasoid (California), Japanese (Tokyo), and Chinese (Shanghai and Beijing) SS patients. In comparison to local controls in each region, we found: 1) increased frequency of the predicted haplotype HLA-DRB1*0301-DRB3*0101-DQA1*0501-DQB1*0201 in Caucasoid patients (p < 0.001); 2) increased frequency of the predicted haplotype HLA-DRB1*0405-DRB4*0101-DQA1*0301-DQB1*0401 in Japanese patients (p < 0.05); 3) increased frequency of the predicted haplotype DRB1*0803-DQA1*0103-DQB1*0601 in Chinese patients (p < 0.05); and 4) no statistically significant association with DPB1 alleles in any group, although an increased number of Caucasoid and Japanese SS patients possessed DPB1*0301. Comparison of DNA sequences for the three disease-associated haplotypes in these ethnic groups revealed a shared region of predicted amino acids from positions 58 to 69 in the first domain of HLA-DQB1. These results extend previous studies by demonstrating that no single class II allele was associated with 1 degree SS in the different ethnic groups. However, a shared amino acid motif in the DQB1 first domain was present in each disease-associated haplotype.

Analysis of multi-parameter video measurements of human neutrophil movement and its relation to cell shape and cytosolic calcium.

We have developed a software system for the analysis of multiparameter video image data derived from cell tracking experiments. The software is part of an integrated system for the study of human neutrophil motility. Input is taken from a file produced by a separate tracking program. The present software calculates several analytic parameters related to movement as well as novel relations between the distribution of fluorescent probes within the cell and vectors which represent cell movement. The software is designed to facilitate data analysis on several platforms by taking input and producing output as ASCII files.
