[Chemical analysis of lipopolysac-charides derived from oral anacerobes]

Lipopolysaccharides were isolated from six oral anaerobes(Va.Bg.Bf.Fn.Aa.Co.)by the hot phenol-water procedure and purified by PCP method. The six species can be all extracted their LPS with different yields (from 0.70 to 2.83% of cell dried weight).Comparable of LPS with the enterobacteriaceae(Sm.),they were the constructed with hexosamine, fatty acids and phosphorus etc.On the other hand, they showed much distinct differences from the enterobacterial LPS structure as follow:(1) 2-keto-3-deoxy-D-mannooctonate(KDO) was not detected by routine procedure.(2)The amount of heptose is reltively lower.(3) Methylated and branched fatty acids were detected. These chemical characteristics may relate to biological activities and pathogenic potential of LPS and bacteria themselves.

Intracellular pH and sodium-proton exchange activity of lymphocytes in stroke-prone spontaneously hypertensive rats.

1. At the age of 20 weeks, intracellular pH (pHi) of circulating lymphocytes suspended in HCO(3-)-free NaCl media was not significantly different between stroke-prone spontaneously hypertensive rats (SHRSP) and normotensive Wistar-Kyoto rats (WKY). 2. The initial recovery rate of pHi in lymphocytes tended to be greater in SHRSP than in WKY after the addition of 60 mmol/L or 120 mmol/L of NaCl, but there was no statistically significant difference. 3. The H+ equivalent efflux rate, which was a true reflection of Na(+)-H(+) activity, was significantly greater in SHRSP than in WKY (P less than 0.05). The difference in H+ equivalent efflux rate was not due to the difference in cellular buffering power between the two groups (P greater than 0.05). An increased Na(+)-H(+) exchange activity may play a partial role in the pathogenesis of hypertension.

Degradation of 2,4,6-trichlorophenol by Azotobacter sp. strain GP1.

A bacterium which utilizes 2,4,6-trichlorophenol (TCP) as a sole source of carbon and energy was isolated from soil. The bacterium, designated strain GP1, was identified as an Azotobacter sp. TCP was the only chlorinated phenol which supported the growth of the bacterium. Resting cells transformed monochlorophenols, 2,6-dichlorophenol, and 2,3,6-trichlorophenol. Phenol and a number of phenolic compounds, including 4-methylphenol, all of the monohydroxybenzoates, and several dihydroxybenzoates, were very good carbon sources for Azotobacter sp. strain GP1. The organism utilized up to 800 mg of TCP per liter; the lag phase and time for degradation, however, were severely prolonged at TCP concentrations above 500 mg/liter. Repeated additions of 200 mg of TCP per liter led to accelerated degradation, with an optimum value of 100 mg of TCP per liter per h. TCP degradation was significantly faster in shaken than in nonshaken cultures. The optimum temperature for degradation was 25 to 30 degrees C. Induction studies, including treatment of the cells with chloramphenicol prior to TCP or phenol addition, revealed that TCP induced TCP degradation but not phenol degradation and that phenol induced only its own utilization. Per mol of TCP, 3 mol of Cl- was released. 2,6-Dichloro-p-benzoquinone was detected in the resting-cell medium of Azotobacter sp. strain GP1. By chemical mutagenesis, mutants blocked in either TCP degradation or phenol degradation were obtained. No mutant defective in the degradation of both phenols was found, indicating separate pathways for the dissimilation of the compounds. In some of the phenol-deficient mutants, pyrocatechol was found to accumulate, and in some of the TCP-deficient mutants, 2,6-dichlorohydroquinone was found to accumulate.

[Study on isoenzymes of Paragonimus iloktsuenensis and Paragonimus westermani by isoelectric focusing].

The isoenzyme patterns of Paragonimus iloktsuenesis and P. westermani were studied by isoelectric focusing on polyacrylamide gels for PGM, MDH, HK, ME, PO and ACP. Interspecific differences were apparent as shown by the isoenzyme profiles of these enzymes examined. MDH was polymorphic with 2 recognizable phenotypes, designated common type and variant type respectively, in P. i. and with 3 phenotypes, designated MDH1, MDH2 and MDH1-2 respectively, in P. w. The result showed that the two species could be distinguished one from another by such species-specific band patterns of isoenzymes. This suggests that P. w. and P. i. have undergone genetically divergent development and P. i. can be regarded as a valid species different from P. w. In the light of MDH polymorphism, some implications of isoenzyme analysis on taxonomy of the genus Paragonimus were discussed.

Mechanisms of photochemically produced turbidity in lens protein solutions.

Calf alpha- and gamma-crystallin were photolyzed in 1-2 mg ml-1 aqueous solutions, using both laser and conventional UV radiation in the 297-320 nm wavelength region. Gamma-crystallin solutions became highly turbid upon UV irradiation, while alpha-crystallin developed no turbidity when irradiated under identical conditions. The photolyzed solutions were analyzed by SDS-PAGE. These gels revealed loss of normal 20 kDa polypeptide, and formation of higher molecular weight peptides, in both alpha- and gamma-crystallin, presumably as a result of photocross-linking reactions and/or protein insolubilization. Thus, although both crystallins underwent photocross-linking, significant turbidity production only occurred in gamma-crystallin. Some possible explanations for these differences are proposed, with one possibility being that most photocross-links in alpha-crystallin occur between subunits of the 1000-kDa oligomer, while in gamma-crystallin the cross-links occur between 20-kDa monomer units. Hence, cross-linking in alpha-crystallin does not affect the average size of particles in solution (or the turbidity), while cross-linking in gamma-crystallin results in a significant increase in average particle size with concomitant increase in turbidity. Another possible explanation is that UV-irradiated gamma-crystallin becomes insoluble (due to charge changes resulting in non-covalent aggregation) while alpha-crystallin does not. Other differences in the photochemical behavior of alpha- vs. gamma-crystallin were noted--gamma-crystallin photolysis rate was about 50% greater than alpha-crystallin. Alpha-crystallin photolysis yielded strong NFK-like fluorescence, while gamma-crystallin did not. One similarity was that photolyzed alpha- and gamma-crystallin lost amino acids His and Trp at about the same rate.(ABSTRACT TRUNCATED AT 250 WORDS)

Epidemiological and aetiological evidence for transmission of Lyme disease by adult Ixodes persulcatus in an endemic area in China.

Involvement of adult Ixodes persulcatus ticks in the transmission of Lyme disease in Hailin County, Heilongjiang Province, China, is reported. In 1986 from April through August adult I. persulcatus was the dominant tick in this endemic area with an infection rate of 43% for the Lyme disease spirochaete, Borrelia burgdorferi. The incidence of Lyme disease cases presenting the symptom of erythema chronicum migrans (ECM) within this area was correlated with the seasonal abundance of adult I. persulcatus and the number of people bitten by ticks. The frequency of ECM formation in all age groups varied and was associated with the frequency of tick bites. In addition, a strain of B. burgdorferi was isolated from a pool of six female I. persulcatus collected from this area. We demonstrate that the seasonal abundance of adult I. persulcatus and its frequent attachment to humans result in the spring and summer transmission of Lyme disease in this endemic area. The role of immature I. persulcatus in Lyme disease transmission is apparently minimal.

Photodamage to calf lenses in vitro by excimer laser radiation at 308, 337, and 350 nm.

Calf lenses in vitro were irradiated using an excimer laser at wavelengths of 308, 337, and 350 nm for times ranging from 10 minutes to 5 hours. The laser power was 2.0 watts (W) at 308 nm, 0.2 W at 337 nm, and 2.0 W at 350 nm. During irradiations, the visible light transmission (632.8 nm) of the lenses was measured and found to be decreased markedly with 308- or 337-nm irradiation. No change in visible light transmission was observed with irradiation at 350 nm. Irradiated lenses were also compared with dark control lenses by photographic record. Lenses exposed to 308-nm ultraviolet (UV) radiation for 10-30 minutes showed significant yellow-brown pigmentation and colorless opacification compared with dark controls. Lenses exposed to 337-nm UV light showed primarily colorless opacity with little pigment production. Lenses exposed to 350-nm radiation for up to 1 hour were visibly indistinguishable from dark controls. After photolysis, the lenses were separated into water-soluble and insoluble fractions and analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Compared with dark controls, UV-exposed lenses (308 or 337 nm) showed loss of 20-30-kilodalton (kD) material and production of higher molecular weight material at 40-60 kD and greater than 100 kD. There was no evidence of such changes after 350-nm exposure. The data gave the following order for the degree of photodamage: 308 nm approximately 5 x 337 nm greater than 20 x 350 nm. An action spectrum for lens damage is presented.

Transformation by v-sis occurs by an internal autoactivation mechanism.

Transformation by the v-sis oncogene appears to require an interaction of its protein product, p28v-sis, with the receptor for the platelet-derived growth factor (PDGF). However, this interaction may not occur at the cell surface as predicted by the autocrine hypothesis because phenotypic transformation was not reversed by incubation of SSV-NRK cells with antisera to PDGF and because morphological transformation did not occur when nontransformed NRK cells were cultured continuously with p28v-sis. A mutant of the wild-type v-sis gene was constructed that encodes a v-sis protein targeted for retention within the endoplasmic reticulum and Golgi. NRK cells expressing the mutant v-sis gene did not secrete any detectable v-sis protein but were as fully transformed as wild-type v-sis transfectants. The results support a mechanism of transformation by v-sis in which internal activation of the PDGF receptor occurs before expression of either p28v-sis or the PDGF receptor at the cell surface.

[Free calcium in platelets and the serum in essential hypertension].

Intracellular free calcium of platelets was studied in 45 patients with essential hypertension and in 6 patient's normotensive offspring. Intracellular free calcium of platelets in hypertensive patients was significantly higher than those in normotensive subjects (223 +/- 26 vs 170 +/- 28 nmol/lL, P less than 0.001). There was a positive correlation between intracellular free calcium of platelets and blood pressure (r = 0.653 approximately 0.729, P less than 0.001). No increase of intracellular free calcium of platelets was found in patient's normotensive offspring. 33 patients also had serum free calcium measured simultaneously. There was no difference between patients and the normotensive control (1.10 +/- 0.15 vs 1.09 +/- 0.10 mmol/L, P greater than 0.05). However, a positive correlation between serum free calcium and blood pressure (r = 0.363, P less than 0.05) and a weak positive correlation between serum free calcium and intracellular free calcium of platelets (r = 0.337, r 0.05 = 0.344) were found. These results indicated that intracellular free calcium was involved in the regulation of blood pressure, it was postulated that the intracellular free calcium of smooth muscle cell in the peripheral arteriole with resistance might have the similar changes seen in platelets. The changes of intracellular free calcium of platelets might be valuable in the prevention and treatment of hypertension and its vascular complications.

Clinical manifestations and epidemiological characteristics of Lyme disease in Hailin county, Heilongjiang Province, China.

Clinical manifestations and epidemiological characteristics of Lyme disease in Hailin county, Heilongjiang Province, China have been reported. The clinical picture of erythema chronicum migrans (ECM) is variable. ECM in the form of annular erythematous patch is uncommon. It is an extensive and indurated lesion. In some instances, a vesicle or necrosis appears in the center of the lesion. Secondary erythema may present in some patients. The neurologic abnormalities consist of meningitis, facial palsy, and polyneuritis. Cardiac abnormalities are rare. In addition, there were cases with lymphadenosis benigna cutis (LABC), which had heretofore only been reported in Europe. The attack rate of ECM is 8.4%. There was a significant sex difference, and most cases occurred in May and June. All patients had a history of tick bite. The prevalence rates of neurologic abnormalities and arthritis were 4.6% and 6.6%, respectively. Three strains of spirochete that are closely related to Borrelia burgdorferi were isolated from Ixodes persulcatus ticks and facial palsy patients. From the above results it is concluded that a focus of Lyme disease exists in this region.

Protective effects of ginsenosides on anoxia/reoxygenation of cultured rat myocytes and on reperfusion injuries against lipid peroxidation.

Lactic acid dehydrogenases (LDH) release from cultured rat myocytes during anoxia and reoxygenation were significantly reduced by ginsenosides (G) and its components Rb+Ro, both in 83 micrograms/ml, but not by Rg. G in higher concentration (250 micrograms/ml) enhanced the LDH release. G and Rb+Ro both 8.3 micrograms/ml in the perfusate, also significantly reduced CPK release from isolated rat heart. Rb component 50 mg/kg protected rat myocardial infarct in vivo against lipid peroxidation signified by significant reduction of malondialdehyde (MDA) and increase of superoxide dismutase (SOD) in rat myocardium. It was concluded that G in adequate dose can protect myocyte anoxia and reoxygenation as well as reperfusion damage in isolated rat heart, and the mechanism of protection may partly attribute to its action on oxygen free radical formation and lipid peroxidation. The active components of myocardial protection are Rb+Ro, and not Rg.
