[Hemodynamic effect of an opioid receptor antagonist diprenorphine in canine endotoxic shock].

The hemodynamic effect of diprenorphine (M5050) in canine endotoxic shock was studied. Mean arterial pressure, left ventricular pressure, left ventricular contractility (dp/dt max), stroke volume, and cardiac output in the endotoxin-shocked dogs were significantly improved by intravenous injection of naloxone (2 mg/kg) and intraventricular microinjection of diprenorphine (40 micrograms), but not by intravenous injection of diprenorphine. It was suggested that the site of action of diprenorphine might be in the central nervous system, and the endogenous opioid substances in the central nervous system may play a more important role in the pathogenesis of septic shock than does that in the peripheral system.

[Effects of gossypol (+,-,+) on phorbol ester and A23187-stimulated prostaglandin synthesis by macrophages].

Previously, gossypol was found to have a wide range of cellular effects. In the present study, the effects of racemic, (-) and (+) gossypol on prostaglandin synthesis by mouse peritoneal macrophages were examined. The synthesis of prostacyclin, thromboxane A2 and prostaglandin F2 alpha was stimulated by phorbol-12-myristate-13-acetate (PMA), A23187 and arachidonic acid. Racemic, (-) and (+) gossypol dose-dependently inhibited PMA or A23187-stimulated prostaglandin synthesis, but failed to affect arachidonic acid stimulated prostaglandin synthesis. It is therefore possible that both (-) gossypol and (+) gossypol inhibit prostaglandin synthesis at the level of arachidonic acid release. Our studies point out two possibilities: 1) the inhibition of (+) and (-) gossypol on prostaglandin synthesis may not be the main mode of the contraceptive action of gossypol; 2) (+) gossypol, which also inhibits prostaglandin synthesis, may have antifertility effects.

[Inhibition of phorbol ester and A23187-stimulated prostaglandins synthesis in macrophages by anisodamine].

The effect of anisodamine on arachidonic acid metabolism in macrophages was studied. The results indicated that the synthesis of PGI2, TXA2 and PGF2 alpha stimulated by phorbol-12-myristate-13-acetate (PMA) and calcium ionophore A23187 was dose and time-dependently inhibited by anisodamine, and preincubation of the cells with anisodamine significantly decreased subsequent PMA and A23187-stimulated synthesis of prostaglandins. An isodamine failed to inhibit exogenous arachidonic acid-stimulated synthesis of prostaglandins. It is suggested that the site of action of anisodamine could be the phospholipase A2 reaction.
