[High level expression and purification of Cysticercosis cellulose annexin32 in Escherichia coli].

In previous work, the cDNA encoding Cysticercus cellulose annexin32 has been cloned. With PCR method, two different restriction Sites were added to each end of the cDNA respectively. Then, the cDNA was inserted into prokaryotic expression vector pJLA-503. After inducing, most foreign protein was expressed in soluble form, which was up to 35% of the total protein of the bacteria. Subsequently, the recombinant Annexin32 was purified with (NH4)2SO4 stepwise precipitation, DEAE-Sepharose FF and Sephacryl S-200 HR chromatography. The final pure protein can been shown as a single band in SDS-PAGE, and the biological activity was verified by Western blot and anticoagulation activity assay.

[Allogeneic peripheral blood stem cell transplantation for treatment of leukemia].

OBJECTIVE: To observe engraftment kinetics, incidence and severity of graft-versus-host disease (GVHD) and clinical outcome in 40 recipients undergoing allogeneic peripheral blood stem cell transplantation (allo-PBSCT). METHODS: 40 leukemia patients with a median age of 35 years underwent allo-PBSCT from June 1997 to May 1999. PBSC were mobilized with G-CSF at a dose of 5 microg/kg S. C. every 12 hours for 5 days. A median of 7.7 (range 2.0 to 16.8) x 10(6) CD(34)(+) cells/kg was infused into the recipients. Busulfan (BU) and cyclophosphamide (CY) were used as conditioning regimen. All patients received cyclosporine A and either methotrexate (n = 34) or methylprednisolone (n = 6) for GVHD prophylaxis. RESULTS: Engraftment of neutrophils and platelets was achieved at a median of day + 13 (9 - 28) and day +12 (7 - 60) respectively. Patients receiving >or=4 x 10(6) CD(34)(+) cells/kg or given G-CSF post transplant had significantly accelerated neutrophil and platelet engraftment. Acute GVHD occurred in 17 of the 40 patients (42.5%), with grade II-IV acute GVHD in 10 patients (25.0%); chronic GVHD developed in 21 out of the 30 evaluable patients (70.0%) with a median follow up of 480 days (range 300 - 1,000). The transplant related mortality is 20.0% and the relapse rate is 10.0%. The probability of leukemia free survival at 3 years is 70.0%. CONCLUSIONS: These results indicate that allo-PBSCT can provide rapid hematopoietic reconstitution without an increased incidence of acute GVHD, but may be associated with high risk of chronic GVHD.

[A multicenter clinical study on clinical effects of meglumine cyclic adenylate in treating patients with chronic pulmonary heart disease].

OBJECTIVE: To observe the clinical effects of meglumine cyclic adenylate (MCA) in treating patients with chronic pulmonary heart disease. METHODS: 785 patients with chronic pulmonary heart disease were divided into two groups: MCA group and control group. Both groups of patients were treated with the same ordinary methods, but MCA was given in the MCA group. The period of treatment was 5-7 days. The degree of improvement in clinical symptoms, signs, ECG, pulmonary ventilation function and arterial blood gas analysis of the two groups before and after treatment were compared. RESULTS: The rates of improvement in clinical symptoms and signs were 92.2% and 73.6% in MCA group and control group respectively. VC increased (426 +/- 123) ml and (313 +/- 145) ml respectively as compared with the values before the treatment. FEV(1) increased (134 +/- 54) ml and (95 +/- 55) ml, PEF increased (0.78 +/- 0.32) L/s and (0.47 +/- 0.32) L/s and PaO(2) increased (23.4 +/- 9.5) mm Hg and (17.3 +/- 10.4) mm Hg respectively. The rates of ECG improvement were 63.7% and 41.2% respectively. There was significant difference between the two groups (P < 0.01 and 0.05). CONCLUSION: Significant therapeutic effects were obtained in treating patients of chronic pulmonary heart disease with respiratory and cardiac failure by using ordinary treatment measures combined with MCA injection.

Torsade de pointes induced by metoclopramide in an elderly woman with preexisting complete left bundle branch block.

There is a growing list of drugs implicated in acquired long QT syndrome and torsade de pointes. However, the torsadogenic potential of metoclopramide, a commonly used antiemetic and prokinetic drug, has not been reported in the literature, despite its chemical similarity to procainamide. We report on a 92-year-old woman with preexisting complete left bundle branch block who developed torsade de pointes after intravenous and oral administration of metoclopramide. This patient also developed torsade de pointes when cisapride and erythromycin were given simultaneously. These two episodes were suppressed successfully after discontinuing the offending drugs and administering class IB drugs. This is the first documentation that metoclopramide provokes torsade de pointes clinically. Metoclopramide should be used cautiously in patients with a risk of torsade de pointes.

[The effect of alcohol on c-fos gene expression in rat embryo neuroglial].

OBJECTIVE: This paper is aimed to explore the mechanisms of brain development abnormality induced by alcohol. METHODS: Astrocytes and oligodendrocytes of 19-day rat embryo were exploited and cultured in vitro, and alcohol and its metabolite product (acetaldehyde) were added to DMEMF(12) medium. After different exposure times, c-fos expression of astrocytes and oligodendrocytes was measured by the immunocytochemistry technique. RESULTS: Changes in c-fos gene expression induced by alcohol and acetaldehyde was time and dose dependent. After 1 hr exposure, alcohol and acetaldehyde affected c-fos gene expression in two kinds of neuralglia. C-fos positive expression reached peak value after 2 hr, but recovered after 72 hr and showed special time phase expression. CONCLUSIONS: Alcohol and acetaldehyde cause abnormal increase of c-fos gene expression in astrocytes and oligodendrocytes. This abnormal expression may play an important role in abnormal brain development induced by alcohol.

[The characteristics of multifocal electroretinogram in normal subjects in China].

OBJECTIVE: To test the characteristics of multifocal ERG in normal Chinese subjects and to provide reference values in clinical application. METHODS: The multifocal ERGs by using VERIS 4.0 visual evoked response imaging system were tested for 42 subjects (48 eyes). Their ages ranged from 11 to 67 years (mean, 40.7 years). The stimulative visual angles subtended plus minus 26.6 horizontally and plus minus 22.1 vertically. Burian-Allen contact lens electrode was used to record the responses from 103 retinal locations in 8 min (16 segments). RESULTS: With age increasing, the latencies of N(1) wave and P(1) wave delayed, there were statistically significant differences of the latencies between < 50 year group and greater-than-or-equal 50 year group (t > 2.01, P < 0.05), and the amplitude densifies of N(1) wave and P(1) wave decreased with wide inter-individual variation. The amplitude densities of N(1) wave and P(1) wave were greatest at the central fovea, they were (44.88 plus minus 13.30) nV/deg(2), (70.90 plus minus 17.77) nV/deg(2) respectively, the densities reducing towards more peripheral location. The amplitude sum of N(1) wave at the upper retina were greater than that at the lower retina, and the amplitude densities and sums of N(1) wave and P(1) wave at superior-temporal quadrant were greater than that at inferior-nasal quadrant. CONCLUSION: Our results suggest multifocal ERG be able to reflect the visual function at different retinal locations correctly and objectively and provide normal reference limits.

[Change of cone's motion perception in patients with primary open angle glaucoma].

OBJECTIVE: To assess the change of cone's motion perception in primary open angle glaucoma (POAG). METHODS: Equi-luminance random dots moving test was used to determine the change of motion perception of blue, green and red cones in 51 eyes with POAG. To isolate the three kinds of cones, the blue, green and red visual targets were displayed on the yellow, purple and blue-green background respectively. RESULTS: The direction discrimination thresholds of motion perception from blue, green and red cones were all damaged in POAG compared to that of the normal group, and among them the blue cone was the most obviously damaged. CONCLUSIONS: These findings indicate that cone's motion perception, especially the blue cone's, may reveal pre-clinical visual nerve damage in POAG.

[Is hepatic ultrasonography necessary in the initial check-up of patients with squamous cell carcinoma of the upper respiratory and digestive tract?].

PURPOSE OF THE STUDY: The purpose of our study was to determine the position and value of ultrasound scan of the liver in the initial check-up of patients treated for a squamous cell carcinoma of the upper respiratory and digestive tract. MATERIAL AND METHODS: Our study is based on a retrospective review of 267 patients (249 males and 18 females) managed in the E.N.T. Department of Grenoble universitary hospital from 1993 to 1995 for a upper respiratory and digestive tract malignant tumor. No patient has been previously treated. The site of the primary tumor was: the oropharynx (108 cases), the hypopharynx (88 cases), the oral cavity (44 cases), the larynx (20 cases), the rhinopharynx (6 cases) and the cervical oesophagus (1 case). Endoscopic procedure with biopsy was performed for all the patients. Histologic examination revealed an invasive squamous cell carcinoma in all the cases. The complete check up included a ultrasound scan of the liver and a chest X-ray for all the patients. RESULTS: Ultrasound scan of the liver revealed one or several metastases in 4 cases (1.5%). The primary tumor was hypopharyngeal in 3 cases (2 stages III, 1 stage IV) and oropharyngeal in 1 case (stage III). In three cases, carcinoma was poorly differentiated. Ultrasound scan of the liver was doubtful for 8 patients (3%). The primary tumor was oropharyngeal in 6 cases (1 stage I, 3 stages III, 2 stages IV), laryngeal in 1 case (stage III) and hypopharyngeal in case (stage IV). In six cases carcinoma was well differentiated. All the complementary examinations concluded to a benign liver disease, with a mean diagnosis delay of 4 weeks for the 8 patients. The mean follow-up duration of the 8 patients was 22 months (range 9 to 42 months). None presented any metastases during the follow up. CONCLUSION: Our results compared with those of the literature revealed that ultrasound scan of the liver is a few specific examination which may be recommended for hypopharyngeal tumors, or for a large cervical adenopathy (N2 or N3), a poor differentiated tumor wherever the site of the primary tumor is.

[Serum glutathione S-transferase in bromobenzene-induced acute hepato-toxicity in mice].

The activity of serum glutathione S-transferase (GST) was tested in bromobenzene-induced acute hepatotoxicity in mice. The data showed that a lot of small local necrosis in liver biopsy was observed and serum GST activity was increased after 2 h of treatment with bromobenzene (150 mg/kg i.p.) in mice. Serum alanine transaminase (ALT) activity was also gradually enhanced after 8 hours of treatment. The most obvious changes were at 16 hours of bromobenzene treatment, and serum GST activity was firstly restored to normal level after 24 hours of treatment. There were good positive correlations between serum GST, ALT, as well as serum GST and the changes on liver biopsy. Evaluating analysis of the diagnosis test showed that the sensitivity, specificity and accuracy was 89%, 75% and 82% in serum GST test, and 39%, 92% and 64% in serum ALT test respectively. The ROC work curve also demonstrated that serum GST was more sensitive and specific than serum ALT.

[Developmental toxicity of Al(NO3)3 on rat embryos].

The developmental toxicity and its mechanism of Al(NO3)3 on embryos in SD rats were studied. On the 9.5th day of gestation, the embryos were incubated in a whole-embryo culture system with Al(NO3)3 at concentrations of Al3+ from 0.6 to 9.0 micrograms/ml in culture media for 48 hrs. Viable embryos were evaluated by Brown's morphological scoring system, and the diameter of yolk sac, crown-rump, head length and the dry weight of embryos were measured. There was a dose-dependent relations of decreasing embryo development with increasing concentrations of Al3+. Yolk sac diameter, head length, dry weight and heart, forelimb as well as neural tube scores decreased significantly at 1.2 micrograms/ml (P < 0.05). When embryos were exposed to Al3+ at > or = 3.0 micrograms/ml, the embryonic development and morphological differentiation were obviously inhibited(P < 0.05); meanwhile, the incidence of dysmorphogenesis significantly increased, including neural tube defects and dorsiflexion teratogenesis. The results suggested that aluminum might be a developmental toxicant and dysmorphogenesis agent.

[Changes serum level of tumor specific growth factor and their clinical significance in mechanical trauma patients].

OBJECTIVE: To investigate the post-traumatic changes in the serum level of tumor specific growth factor (TSGF) and their relationship with Injury Severity Score and to inquire into the possible mechanism and clinical significance. METHODS: Fifty patients with mechanical trauma were eligible and included. According to the standard of Injury Severity Score (ISS), the patients were divided into there groups: mild degree group, (ISS < or = 16, n = 31) severe degree group (16 < ISS < 25, n = 13) extreme severe degree group (ISS > or = 25, n = 6). The blood samples of all patients were obtained within 24 hours, and those of 25 dynamically observed patients were obtained on days 3, 5, 7, 9 and 12. RESULTS: The serum level of TSGF was significantly higher in patients with trauma in different groups (P < 0.05). The dynamical observation showed that the index demonstrated curve changes with increase in early stage and decrease later. the climax of TSGF appeared on 7 day in all 3 groups. The TSGF serum levels in extremely severe degree and severe degree groups > mild degree group on days 5, 7, 9 and 12 day (P < 0.05). CONCLUSION: TSGF has relationship with the inflammatory response and recovery of trauma.

[An experimental study on bone marrow purging in vitro for patients with chronic myelogenous leukemia].

OBJECTIVE: To investigate the effect of IL-2, IFN and bcr-abl AS-ODN on bone marrow purging in vitro for patients with chronic myelogenous leukemia (CML). METHODS: By amplifying in vitro clone forming culture, and purging the bone marrow of 5 CML patients through different combined use of IL-2, IFN and bcr-abl AS-ODN, we compared the effects of 4 combined regimens. RESULTS: It was found that the suppressive effects of the 4th regimen combining IL-2 (800 U/ml), IFN (800 u/ml) and bcr-abl AS-ODN (30 micrograms/ml) on leukemia cell clone forming unit (CFU) were significantly different from the suppressive effects of the other 3 regimens (P < 0.01). With the use of the 4th regimen, the GM-CFU and L-CFU survival rates were 26.91% and 3.49% respectively (P < 0.01). CONCLUSION: The combined regimen of IL-2 (800 U/ml), IFN (800 u/ml) and bcr-abl AS-ODN (30 micrograms/ml) has good effect on selective CML bone marrow purging in vitro and can be used in clinical purging of CML bone marrow.

[One stage otoplasty treatment for congenital malformations of the auricle and middle ear].

OBJECTIVE: To investigate the application of one stage otoplasty for congenital atresia of the external acoustic canal and malformations of the middle ear and the auricle. METHOD: Patients with the ear malformations were given surgical reconstruction of one stage otoplasty. The auricle was reconstructed with the rib which was encapsulated with the superthin temporal flap. According to the malformations of the middle ear in patients, I style tympanoplasty and III style tympanoplasty were carried out respectively. All patients were performed myringoplasty with temporal fascia and reconstructed the external acoustic canal with full thickness skin-grafting. RESULT: A long term follow-up (4-6 years) demonstrated that 11 ears were survival of which 8 ears figuration were ideal. The hearing improvement was observed in all patients. CONCLUSION: One stage otoplasty is effective for treatment of the congenital malformations of the external and middle ear.

[The measurement of multifocal electroretinography].

PURPOSE: Discussing the testing procedure of multifocal electroretinography(ERG). METHODS: The testing procedure of 131 multifocal ERG records was reviewed. RESULTS: Some variable factors existed in the measurement of multifocal ERG. These factors included the subject's co-operation, the mounting of electrode, the fixation of tested eye and adjustment of focus. The good records of multifocal ERG depend upon the skill of the operator. CONCLUSION: The testing procedure of multifocal ERG must be standardized so as to ensure the reliability and repeatability of the results.

Potentiation of docetaxel antitumor activity by batimastat against mouse forestomach carcinoma.

OBJECTIVE: To improve the treatment of tumors, we studied the combined effects of docetaxel and batimastat (BB-94) on mouse forestomach carcinoma (MFC), and compared them with doxorubicin. METHODS AND RESULTS: In vitro, growth curve analysis, MTT assay and clonogenic assay used to determine the cytotoxic effect of docetaxel or/and BB-94 on MFC cell showed that docetaxel but not BB-94 had a significant cytotoxicity, and the effect of docetaxel wasn't enhanced by BB-94. In early stage MFC tumor model, obvious antitumor effect of docetaxel or doxorubicin given i.v. at maximum tolerated dose (MTD, docetaxel: 20 mg/kg; doxorubicin: 6 mg/kg) every 4 days for 3 injections (q4d x 3), even that of BB-94 (30 mg/kg i. p. qd x 20) was observed. Tumor growth inhibition was greater for docetaxel-batimastat (96.0%) than for doxorubicin-batimastat (88.0%), docetaxel (89.0%), doxorubicin (68.0%) and BB-94 (33.0%), and the effect of docetaxel could be potentiated by BB-94. Docetaxel also showed activity against advanced stage MFC tumor in dose-dependent manner, and was more effective at MTD than doxorubicin with 4/5 regressions, 46.5 days tumor growth delay and 2.8log(10) tumor cell kill. CONCLUSION: Our results suggest that in the MFC model with dose and schedule used, docetaxel is an effective cytotoxic new drug against MFC tumor and BB-94 enchances the antitumor activity of docetaxel.

[Effect of radix angelicae sinensis decoction for supplementing blood on inhibiting the increase of endothelial cell monolayer permeability induced by hypoosmotic solution].

OBJECTIVE: To study effect of radix Angelicae sinensis decoction for supplementing blood (RASDSB) on inhibiting the increase of endothelial cell monolayer permeability induced by hypoosmotic solution. METHODS: The endothelial cells isolated from newborn bovine aorta were cultured on polycarbonate microporous filter membrane to develop compact endothelial monolayer. Fluid filtration coefficient (Kf), filtration volume (Jv) and osmotic reflective coefficient (sigma) to protein of the endothelial monolayer were measured treated by hypoosmotic solution (changing concentration of serum in M199 solution from 20% to 2%) for 120 min or by hypoosmotic solution containing 10(-4) g x ml(-1) RASDSB for 120 min after perfused Hanks balanced salt solution containing 5 g x L(-1) albumin. RESULTS: Kf and Jv of the endothelial monolayer treated by hypoosmotic solution increased and sigma of that decreased. RASDSB could inhibit above-mentioned change. Morphological analysis demonstrated that RASDSB could inhibit widening of intercellular distance and enlargement of cellular area in the endothelial monolayer induced by hypoosmolality. CONCLUSION: Hypoosmotic solution could increase endothelial cell monolayer permeability and RASDSB could inhibit the increase.

The human hematopoietic stem cell compartment is heterogeneous for CXCR4 expression.

The chemokine stromal derived factor-1alpha (SDF-1alpha) has been implicated recently in the chemotaxis of primitive human hematopoietic cells, suggesting that pluripotent human stem cells express the SDF-1alpha receptor, CXCR4. By using flow cytometry and confocal microscopy, we have identified and isolated primitive subsets of human CXCR4(+) and CXCR4(-) cells. Distinctions in the progenitor content and response to SDF-1alpha in vitro indicate that CXCR4(+) and CXCR4(-) cells represent discrete populations of primitive blood cells. The i.v. transplantation of these subfractions into immune-deficient mice established that both possess comparable engraftment capacity in vivo. Human myeloid, lymphoid, and primitive CD34(+) CXCR4(+) cells were present in chimeric animals transplanted with either subset, indicating that CXCR4(+) and CXCR4(-) stem cells have equivalent proliferative and differentiative abilities. Our study indicates that the human stem cell compartment is heterogeneous for CXCR4 expression, suggesting that the relationship between CXCR4 expression and stem cell repopulating function is not obligatory.

Sequence and analysis of chromosome 1 of the plant Arabidopsis thaliana.

The genome of the flowering plant Arabidopsis thaliana has five chromosomes. Here we report the sequence of the largest, chromosome 1, in two contigs of around 14.2 and 14.6 megabases. The contigs extend from the telomeres to the centromeric borders, regions rich in transposons, retrotransposons and repetitive elements such as the 180-base-pair repeat. The chromosome represents 25% of the genome and contains about 6,850 open reading frames, 236 transfer RNAs (tRNAs) and 12 small nuclear RNAs. There are two clusters of tRNA genes at different places on the chromosome. One consists of 27 tRNA(Pro) genes and the other contains 27 tandem repeats of tRNA(Tyr)-tRNA(Tyr)-tRNA(Ser) genes. Chromosome 1 contains about 300 gene families with clustered duplications. There are also many repeat elements, representing 8% of the sequence.
