Generation of hydroxyeicosatetraenoic acids by human inflammatory cells: analysis by thermospray liquid chromatography-mass spectrometry.

Arachidonic acid was converted to a series of hydroxyeicosatetraenoic acids (HETEs) by mixed human inflammatory cells following stimulation with the calcium ionophore A23187. HETEs were purified by a simple one-step extraction procedure followed by HPLC. The HPLC was coupled to a Finnigan quadrupole mass spectrometer using the now commercially available thermospray liquid chromatography-mass spectrometry interface. The HPLC eluant was monitored 'on line' by the mass spectrometer. Soft ionisation occurs, generating intense molecular ion species in the negative ion mode (M - H-:m/z 319) for each of the isomeric HETEs. The (M + H+ - H2O) ion at m/z 303 is the major species in the positive ion spectra of HETEs. Mass spectra were obtained on-line post-HPLC for HETEs formed by the human cells, and the HPLC-MS profile compared with that obtained from standards; species corresponding to the 11-, 9- and 5-HETEs were observed.

The association of rhinoviruses with lower respiratory tract disease in hospitalized patients.

An analysis of 32 hospitalized infants and children from whom rhinoviruses were isolated in our diagnostic laboratories in 1982-83 suggests that these agents are associated with lower respiratory tract disease with focal findings in susceptible patients. In 23 cases, an acute lower respiratory disease was the cause for admission, while nine patients were cultured after new respiratory symptoms developed during hospitalization. Presenting signs and symptoms included cough (23), fever (19), rhinorrhea (19), respiratory distress (14), and decreased feeding (15). Seventeen of 25 chest x-rays showed new focal abnormalities. Twenty-five patients had a significant underlying disease including seven with malignancies, six with respiratory tract abnormalities, and four with congenital heart lesions. Six of the remaining seven patients were less than 2 1/2 months of age. In no cases were significant bacterial or fungal pathogens isolated; two did have concomitant viral isolates. Rhinoviruses in the appropriate clinical setting are associated with significant pulmonary disease.

Bis-trifluoromethylbenzoyl derivatives for steroid analysis by gas chromatography electron capture negative ion chemical ionisation mass spectrometry.

3,5-Bis-trifluoromethylbenzoyl derivatives of eight monohydroxy steroids and eight dihydroxy steroids were synthesised. Under the reaction conditions described, the monohydroxy steroids each gave a single derivative while the dihydroxy steroids, with the exception of corticosterone, showed multiple product formation. The reactivity of hydroxyl groups relative to their stereochemistry is discussed. The electron capture negative ion chemical ionisation mass spectra of these derivatives were recorded. When the derivatised hydroxyl group was alicyclic or aromatic, a molecular ion was normally the base peak in the mass spectrum. Selected ion monitoring of molecular ions indicated that, in certain cases, as little as 1 pg of the parent steroid could be detected. The potential use of this derivative in the quantitative analysis of steroids by gas chromatography-mass spectrometry is discussed.

Effects of heavy metals on Drosophila larvae and a metallothionein cDNA.

Drosophila melanogaster larvae reared on food containing radioactive cadmium retained over 80% of it, mostly in the intestinal epithelium. The majority of this radioactivity was associated with a soluble protein of less than 10,000 molecular weight. Synthesis of this cadmium-binding protein was induced by the metal as demonstrated by incorporation of radioactive cysteine. Most copper ingested by larvae was also found to associate with a low molecular weight, inducible protein, but some of it was found in an insoluble fraction. Zinc was unable to, or very inefficient at, binding or inducing the synthesis of a similar protein. A D. melanogaster cDNA clone was isolated based on its more intense hybridization to copies of RNA sequences from copper-fed larvae than from control larvae. This clone showed strong hybridization to mouse metallothionein-I cDNA at reduced stringency. Its nucleotide sequence includes an open-reading segment which codes for a 40-amino acid protein; this protein was identified as metallothionein based on its similarity to the amino-terminal portion of mammalian and crab metalloproteins. The ten cysteine residues present occur in five pairs of near-vicinal cysteines (Cys-X-Cys). This cDNA sequence hybridized to a 400-nucleotide polyadenylated RNA whose presence in the cells of the alimentary canal of larvae was stimulated by ingestion of cadmium or copper; in other tissues this RNA was present at much lower levels. Mercury, silver, and zinc induced metallothionein to a lesser extent. Whether (any of) the protein(s) discussed above correspond(s) to that coded by this RNA sequence has not yet been determined.

Gas chromatographic-mass spectrometric study of metabolites of C21 and C19 steroids in neonatal porcine testicular microsomes.

Microsomal fractions obtained from testes of 3-week-old piglets have been incubated, separately, with progesterone, 17-hydroxyprogesterone, 5-pregnene-3 beta,20 beta-diol, 16 alpha-hydroxypregnenolone, 5-androstene-3 beta,17 alpha-diol and dehydro-epiandrosterone. The metabolites, after derivatization, have been separated by capillary gas chromatography and identified by mass spectrometry. Quantification was by selected ion monitoring. Progesterone was shown to be 17-hydroxylated and also converted into 4,16-androstadien-3-one (androstadienone). The major metabolite of 17-hydroxyprogesterone was 4-androstene-3,17-dione (4-androstenedione), but little, if any, androstadienone was formed, indicating that this particular biosynthesis did not require 17-hydroxylation. The metabolites of 5-pregnene-3 beta, 20 beta-diol were found to be 17-hydroxypregnenolone, 3 beta-hydroxy-5,16-pregnadien-20-one (16-dehydropregnenolone) and 5,16-androstadien-3 beta-ol. Dehydroepiandrosterone and 5-androstene-3 beta,17 alpha-diol were interconvertible but neither steroid acted as a substrate for 16-androstene formation. However, dehydroepiandrosterone was metabolized to a small quantity of 4-androstenedione. Under the conditions used, no metabolites of 16 alpha-hydroxypregnenolone could be detected. The present results, together with those obtained earlier, indicate that the neonatal porcine testis has the capacity to synthesize weak androgens, mainly by the 4-en-3-oxo steroid pathway. Although 16-androstenes cannot be formed from C19 steroids, progesterone served as a substrate and may be converted directly to androstadienone, without being 17-hydroxylated first. The pathway to 5,16-androstadien-3 beta-ol, however, involves 17-hydroxypregnenolone and 16-dehydropregnenolone as intermediates.

Bistrifluoromethylaryl derivatives for drug analysis by gas chromatography electron capture negative ion chemical ionization mass spectrometry. Application to the measurement of (N-dicyclopropylmethyl)amino-2-oxazoline in plasma.

A gas chromatographic mass spectrometric assay for (N-dicyclopropylmethyl) amino-2-oxazoline in plasma with a detection limit of 0.1 ng ml-1 was required. Various fluoroaryl derivatives of this compound (code name S3341) were synthesized and their positive ion chemical ionization and electron capture negative ion chemical ionization mass spectra recorded. While fluorobenzyl derivatives of S3341 could be made by heating with the requisite benzyl bromide and diisopropylethylamine in acetonitrile, initial efforts to synthesize corresponding fluorobenzoyl derivatives using a benzoyl chloride in dry ethyl acetate at 60 degrees C were unsuccessful. Mass spectral data indicated that only a fragment of the oxazoline ring was retained in the reaction product and that an N-(2-chloroethyl)benzamide was formed. However, when diisopropylethylamine was included in the reaction mixture, a benzoyl derivative of the complete molecule was obtained. The mechanisms of these reactions are discussed. The negative ion mass spectrum of the 3,5-bistrifluoromethylbenzoyl derivative of S3341 has a base peak at m/z 420 (the molecular ion) and, when this ion is specifically monitored, an amount of derivative equivalent to 1 pg of S3341 can be detected. This allowed the development of an assay for S3341 in plasma with a precision of 9% (SD) at 0.2 ng ml-1 and a lower limit for quantitative determination of 0.1 ng ml-1.

Effect of structural modification of the hydantoin ring on anticonvulsant activity.

Selectively substituted hydantoins 1 (15 examples), 4-hydroxy-2-imidazolidinones 2 (13 examples), 2-imidazolones 3 (10 examples), 2-imidazolidinones 4 (four examples), vicinal diamines 5 (two examples), and simple amino acid derivatives 6 (four examples) have been prepared and evaluated in the maximal electroshock seizure (MES), subcutaneous pentylenetetrazole seizure threshold (sc Met), and rotorod (Tox) tests. The medium effective doses (ED50) and the medium toxic dose (TD50) for the most active compounds are reported. In general, the most pronounced activity was observed for hydantoins 1 and protected amino acids 6. Within each series of compounds, enhanced anticonvulsant activity was often noted for compounds containing an aromatic group one carbon removed from a nitrogen atom. Among the most active compounds observed were the amino acid derivative N-acetyl-D,L-alanine benzylamide (6d) and the two 2-imidazolones 4-methyl-1-(phenylmethyl)-1,3-dihydro-2H-imidazol-2-one (3e) and 1-phenyl-1,3-dihydro-2H-imidazol-2-one (3g). Compound 6d proved to be slightly more potent in the MES test than phenacemide.

Nucleotide sequence of two overlapping myc-related genes in avian carcinoma virus OK10 and their relation to the myc genes of other viruses and the cell.

Avian carcinoma virus OK10 has the genetic structure gag-delta pol-myc-delta env. It shares the transformation-specific myc sequence with three other avian carcinoma viruses (MC29, MH2, CMII) and also with a normal chicken gene proto-myc and the gag, pol, and env elements with non-transforming retroviruses. Unlike the other myc-containing viruses, which synthesize singular myc proteins, OK10 synthesizes two different myc-related proteins of 200 and 57 kDa. Here we have sequenced the myc region of an infectious OK10 provirus to investigate how OK10 synthesizes two different proteins from the same myc domain and to identify characteristic differences between the normal proto-myc gene and the myc-related viral transforming genes. It was found that the 1.6-kilobase myc domain of OK10 is colinear and coterminal with the myc domains of MC29, MH2, and the terminal two exons of proto-myc. It is preceded by the same splice acceptor as the myc sequence of MH2 and as the second proto-myc exon. From this and the known structure of retroviruses, it follows that the OK10 gene encoding the 57-kDa protein is discontinuous with a small 5' exon that includes six gag codons and a large 3' myc exon (delta gag-myc). This gene and the delta gag-myc gene of MH2 are isogenic. The proto-myc-derived intron preceding the myc domain of OK10 is in the same reading frame as the adjacent delta pol and myc domains and, hence, is part of the gag-delta pol-myc gene encoding the 200-kDa protein. Sequence comparisons with proto-myc and MC29 and MH2 indicate that there are no characteristic mutations that set apart the viral myc domains from proto-myc. It is concluded that transforming function of viral myc-related genes correlates with the lack of a viral equivalent of the first proto-myc exon(s) and conjugation of the viral myc domains with large or small retroviral genetic elements rather than with specific point mutations. Because OK10 and MH2 each contain two genes with potential transforming function (namely, delta gag-myc and gag-delta pol-myc or delta gag-mht, respectively), it remains to be determined whether the delta gag-myc genes have transforming function on their own or need helper genes. The possible helper requirement cannot be very specific because the two potential helper genes are very different.

Clinical value of a sensitive immunoradiometric assay for TSH.

We have investigated a sensitive immunoradiometric assay for serum thyroid-stimulating hormone (TSH) in 45 thyrotoxic patients not on treatment, 464 euthyroid patients and 111 hypothyroid patients on replacement therapy. Forty-three thyrotoxic and seven euthyroid patients were found to have an undetectable TSH (less than 0.2 mIU/L). Previous work has shown a very clear separation of thyrotoxic and euthyroid patients using sensitive TSH assays. However, our extended study here has revealed that a significant number of euthyroid patients with undetectable TSH (1.5% in our study) are likely to be found if TSH becomes the initial test for thyroid function. Thirty of the hypothyroid patients on thyroxine were found to have undetectable TSH, but only one showed clinical signs of thyrotoxicosis. Most of the patients, although having raised serum free T4, had serum free T3 levels within the euthyroid range or just slightly elevated.

Use of a diode array detector in investigation of neonatal organic aciduria.

We investigated the use of a diode array detector in conjunction with isocratic cation-exchange liquid chromatography for detection of organic acids in urine. The spectra and retention times of abnormal peaks found on chromatography of urine from patients with methylmalonic aciduria, maple syrup urine disease, and lactic aciduria were recorded and compared with those obtained for group of pure organic acids. Our results show that diode array detection helps considerably in tentatively identifying peaks appearing on an abnormal chromatogram while awaiting confirmation by mass spectrometry.

Identification of 16-dehydropregnenolone as an intermediate in 16-androstene biosynthesis in neonatal porcine testicular microsomes.

The biosynthesis of 16-androstenes has been studied in neonatal porcine testicular microsomes using 17-hydroxypregnenolone and 16-dehydropregnenolone, separately, as substrates. The metabolites formed after microsomal incubation with these substrates were purified, derivatized as O-methyloxime-trimethylsilyl ethers and analysed by capillary gas chromatography-mass spectrometry. In the incubation of 17-hydroxypregnenolone with microsomes, 16-dehydropregnenolone was identified as an intermediate in the biosynthesis of 16-androstenes. Further microsomal incubation of 16-dehydropregnenolone has established the intermediary role of this steroid in the production of 16-androstenes.
