Utilization of acetate by Methanomonas emthanooxidans.

Methanomonas methanooxidans incorporates both carbon atoms of acetate into the glutamate and aspartate families of amino acids during growth on methane; carbon dioxide is also evolved from both carbon atoms of acetate. The distribution of carboxyl-labeled acetate incorporated into convalently bound glutamate is consistent with the operation of the tricarboxylic acid cycle in this species, and the presence of alpha-ketoglutarate dehydrogenase was demonstrated in cell-free extracts.

Specificity of a catabolic pathway--a lesson learned from indirect assays.

Studies with purified orcinol hydroxylase suggest that, contrary to previous conclusions, the enzymes of the orcinol pathway cannot transform analogous compounds to common metabolites. The substrate analogues of orcinol uncouple electron flow from reduced nicotinamide adenine dinucleotide to oxygen from the hydroxylation reaction catalyzed by orcinol hydroxylase.

Automatic assessment of respiration during growth in stirred fermentors.

An apparatus for the continuous and automatic measurement of respiration during growth of micro-organisms in stirred aerated culture is described. The effluent atmosphere from the culture vessels is passed through commercially available oxygen and carbon dioxide gas analyzers, and their electrical output is fed to a multipoint recorder. The apparatus has been used to measure the respiration of Escherichia coli, Pseudomonas aeruginosa, Bacillus thuringiensis, and Methylococcus capsulatus during growth on complex and defined media. In addition, pH values and dissolved oxygen concentrations were measured. Growth of P. thuringiensis in glucose-limited media showed unexpected interruptions in the oxygen consumption curves which resembled diauxic growth; growth of B. thuringiensis in complex media showed similar discontinuities of respiration. These results are explained as the sequential utilization of preferred substrates which, in the case of P. aeruginosa were provided as transient intermediates of glucose.

Occurrence of poly-beta-hydroxybutyrate in the Azotobacteriaceae.

Poly-beta-hydroxybutyrate (PHB) from various representative strains of the genera Azotobacter, Beijerinckia, and Derxia was isolated and characterized. During growth in shake culture, with glucose as a carbon and energy source, and molecular nitrogen as a nitrogen source, increase in dry weight appeared linear, and PHB formed a constant percentage of the dry weight. In a medium containing 1% (w/v) glucose, PHB declined with the onset of the stationary phase of growth; with 2% (w/v) glucose, an increase in PHB content during stationary phase was noted in the case of some strains, before a subsequent decline. The decrease in PHB as a percentage of dry cellular weight (not of total amount present in the culture) during growth of some strains with 2% as opposed to 1% (w/v) glucose may be ascribed to a greater production of capsular polysaccharide. PHB content could not be used as a taxonomic criterion. Strain differences were as great as or greater than species differences. The only strain of Beijerinckia fluminensis obtained contained PHB, but it could not be grown on the nitrogen-free medium used. Two species of the genus Azotomonas, reported to be aerobic, nonsymbiotic nitrogen-fixers, did not grow on the nitrogen-free medium used and did not produce PHB during growth with a combined nitrogen source.

The route of ethanol formation in Zymomonas mobilis.

1. Enzymic evidence supporting the operation of the Entner-Doudoroff pathway in the anaerobic conversion of glucose into ethanol and carbon dioxide by Zymomonas mobilis is presented. 2. Cell extracts catalysed the formation of equimolar amounts of pyruvate and glyceraldehyde 3-phosphate from 6-phosphogluconate. Evidence that 3-deoxy-2-oxo-6-phosphogluconate is an intermediate in this conversion was obtained. 3. Cell extracts of the organism contained the following enzymes: glucose 6-phosphate dehydrogenase (active with NAD and NADP), ethanol dehydrogenase (active with NAD), glyceraldehyde 3-phosphate dehydrogenase (active with NAD), hexokinase, gluconokinase, glucose dehydrogenase and pyruvate decarboxylase. Extracts also catalysed the overall conversion of glycerate 3-phosphate into pyruvate in the presence of ADP. 4. Gluconate dehydrogenase, fructose 1,6-diphosphate aldolase and NAD-NADP transhydrogenase were not detected. 5. It is suggested that NAD is the physiological electron carrier in the balanced oxidation-reduction involved in ethanol formation.

Sucrose utilization by Zymomonas mobilis: formation of a levan.

1. Molar growth-yield coefficients of Zymomonas mobilis for glucose, fructose, glucose plus fructose, and sucrose are reported. Yield coefficients for sucrose are appreciably lower than those for the equivalent concentrations of glucose plus fructose. 2. Only 2.6% of [U-(14)C]glucose supplied in the growth medium is incorporated into cell substance by Z. mobilis utilizing glucose as the energy source. 3. During growth on sucrose a levan is formed. It has been characterized and shown to resemble other bacterial levans. 4. Levan formation from sucrose could be demonstrated with both washed cell suspensions and cell extracts of Z. mobilis. 5. Sucrose phosphorylase could not be demonstrated in extracts of the organism.
