A practical technique for preparation of antiserum to canine factor VIII-related antigen.

Identification and measurement of canine factor VIII-related antigen (VIIIR:Ag) has many clinical and research applications, including differential diagnosis of hemophilia A and von Willebrand's disease, use as a marker for specific cell types, and elucidation of the structure of this factor VIII component. We have developed a practical method for producing antibodies to canine VIIIR:Ag that uses 2% agarose filtration for purification and identifies the antigen by correlation with the elution of the peak A280 fraction in the void volume. Antisera for electroimmunoassay (EIA) can be produced in less than four weeks from simple starting materials and with commonly available laboratory equipment. This technique would be useful for either clinical veterinary or comparative research laboratories.

The molecular structure of 11-methylbenz[a]anthracene: an almost planar substituted benz[a]anthracene.

The molecular structure of the almost inactive 11-methylbenz[a]anthracene has been determined by single-crystal X-ray diffraction analysis and refined to a final discrepancy index R of 0.036 over 955 independent reflections. With the benzo-ring A inclined at only 1.8 degrees to the furthest anthracene ring D, the benz[a]anthracene nucleus is much more nearly planar than in other (and carcinogenically more active) benz[a]anthracenes with bay-methyl substituents. Several carbon atoms in rings D and A deviate by approximately 0.05 A from the mean plane through the ring-carbon atoms of the bay-ring B. The shortest carbon-carbon bonds are C5-C6 = 1.341(5) A (at the K-region), C3-C4 = 1.355(5) and C8-C9 = 1.360(5) A, while C10-C11 next to the substituent is also quite short at 1.367(5) A.

Molecular structure of 5,10-dimethoxybenzo[j]fluoranthene.

The molecular and crystal structure of the synthetic 5,10-dimethoxy derivative of the carcinogen benzo[j]fluoranthene has been determined by direct methods from X-ray diffractometric data and refined to an R index 0.041 over 2788 independent reflections. The benzo ring is inclined at approximately 3 degrees to the almost planar fluoranthene moiety (carbon atoms have r.m.s. deviation of 0.02 A from the carbon-atom plane); the methoxy carbon atoms lie within 0.2 A of the molecular plane. Corresponding bond lengths (e.s.d. 0.004 A for carbon-oxygen and carbon-carbon) and angles lie within 3 sigma for the two independent molecules, with mean dimensions of methoxy groups: C-C = 1.422, C-O = 1.372 A, C-O-C = 117.4 degrees.

Efficiency and precision of electroimmunoassay for canine factor VIII-related antigen.

Factors that improved the efficiency, precision, and sensitivity of the electroimmunoassay for canine factor VIII-related antigen were investigated. These included rapid electrophoresis, increased rocket heights, deletion of the gel-washing step, and doubling of the sample capacity per plate. The modified assay was reliable and accurate, could be completed within 3 hours, and permitted twice as many determinations. Other experimental variables that affected the assay were the type of agarose and sample diluent, storage conditions of samples before assay, source of antibody, and use of 2 mM calcium lactate.

Intrarenal abscess. Report of 14 cases.

Six of 14 patients with renal abscess had prior history of urinary tract infection; initial symptoms included fever and flank pain in 12. A drip-infusion intravenous pyelogram was the most sensitive radiologic test, but selective renal arteriography was most specific. Urine cultures were positive in all 14 patients; blood cultures were positive in nine. Six patients were treated with antibiotics alone and eight required surgery. Of the eight, five had pus-filled cavities, one had multiple stones, one had a renal infarct, and one had a resolving abscess. Of six treated with antibiotics alone, one died of unrelated complications and five have demonstrated no pathological renal condition after three to six years.

A crevice corrosion cell configuration.

An occluded corrosion cell configuration and polarization program were developed in order to measure the crevice corrosion resistance of implant alloys. 316 L stainless steel was used in establishing that the crevice cell configuration was convenient, reproducible, free of unwanted crevices, and suitable for laboratory measurements requiring relatively short times.

Shapes of carcinogenic benz[a]anthracenes: the crystal and molecular structure of 1-methylbenz[a]anthracene.

The crystal structure of 1-methylbenz[a]lanthracene, which is weakly carcinogenic, has been determined by application of direct methods to single-crystal X-ray diffractometric data and refined by least squares to R = 0.09 over 845 independent reflections. Crystals are monoclinic, space group P2(1), with a = 8.491(2), b = 7.138(2), c = 10.500(2)ABEta = 95.06(01), Z = 2. As in other benz[a]anthracenes, the K-region bond C(5)-C(6) is short [1.34(1)A]. The distinctive bay geometry, with a methyl group opposite to a hydrogen, H(12), peri to another hydrogen, H(11), has a long bond C(13)--C(18) = 1.47(1)A in the bay, and the angular benz-ring is inclined at 16.5 degrees to the mean plane of the anthracene fragment. The methyl carbon atom is 0.79 A out of the mean molecular plane (or 0.19 A out of the plane of the benz-ring) and the 1.50 A long C(1)-methyl bond makes angles of 117 degrees and 125 degrees at C(1).
