Applications of heavy isotope tracers to clinical studies: progesterone urinary production rate determination in the menstrual cycle and pregnancy.

A method for the in vivo determination of progesterone production rate using progesterone labeled with deuterium as the tracer and gas chromatography-mass spectrometry for assessing isotope dilution in a urinary metabolite is described. By measuring the ratio of labeled to unlabeled free progesterone in a 24-h urine sample, the problem of identifying a unique metabolite appears to have been avoided. Median progesterone production rates on day 7 of the follicular phase and day 7 of the luteal phase were 3.5 mumol/24 h and 72.6 mumol/24 h, respectively. Where repeated determinations were performed through the luteal phase in two nonconceptual cycles, the familiar midluteal plateau of progesterone production was noted. The MCR of progesterone calculated from luteal phase data was 2051 1/24 h (SD, 275 1/24 h). When determined in pregnancy, progesterone production varied from 250 mumol/24 h at 6 weeks in a singleton pregnancy, to 1565 mumol/24 h at 35 weeks in a triplet pregnancy. Repeated determinations before term in a twin pregnancy detected no evidence of decreasing progesterone production before labor. The value of urinary pregnanediol and plasma progesterone to indirectly monitor progesterone production were confirmed.

Integrating severely adaptive handicapped seventh-grade students into constructive relationships with nonhandicapped peers in science class.

The effects of cooperative and individualistic learning experiences were compared on interactions and relationships between severely adaptive handicapped (not adapting to regular classroom demands) and nonhandicapped seventh-grade students in science classes. We investigated the predictions that (a) requiring cooperation between the two groups of students would result in increased rejection of the severely adaptive handicapped students and (b) cooperation would promote increased interpersonal attraction regardless of the heterogeneity of the students. Forty-eight suburban junior-high-school, seventh-grade students (41 nonhandicapped, 9 severely adaptive handicapped) were assigned to conditions on a stratified random basis controlling for handicap, sex, and ability level. They participated in the study for 10 days, 55 minutes per day. Results indicated that cooperative learning procedures, compared with individualistic ones, promoted more interaction and interpersonal attraction between severely adaptive handicapped and nonhandicapped students.

Border disease in a flock of Minnesota sheep.

A flock problem affecting newborn lambs on a Minnesota farm was investigated. The lambs had hairy fleece and rhythmic tremors, and were unthrifty. Histologic examination revealed hypomyelinization of the CNS and hypertrophy of primary follicles in the skin. A virus antigenically related to bovine viral diarrhea virus was isolated. Serum neutralization showed that a large number of flock ewes had much higher titers to bovine viral diarrhea virus than did control animals.

Tumor associated antigens of bovine cancer eye.

A study was conducted to determine the presence of surface antigens on cultured cells of bovine ocular squamous cell carcinoma (BOSCC) by serological testing. Serum from cattle with plaque and malignant stages of disease were tested for reactivity with surface antigens of cultured autologous and allogeneic cells. Indirect immune fluorescence assays were conducted to determine the presence of antibodies towards these tumor cells. It was found that sera tested possessed antibodies to the cultured tumor cells. Further, no surface reactivity was observed when these sera were tested for reactivity with normal epithelial cells. Reactive sera were analyzed by absorption tests with autologous and allogeneic cells. Absorbed sera showed no reactivity to surface antigens or allogeneic cells. This study suggests that there might be a shared antigen among the cells from plaque and malignant lesions.

Bovine monocyte-induced tumor cell cytotoxicosis.

Human monocytes have been shown to be highly cytotoxic for tumor cells in vitro. Th determine whether these observations could be extended to the bovine species, monocyte-induced tumor cell cytotoxicosis was examined in vitro. Target cells were initiated from benign hyperplastic corneal plaque lesions or ocular squamous cell carcinoma lesions in cattle. The monocyte-induced tumor cell cytotoxicosis, as measured by target cell [3H] thymidine incorporation, was highly significant (P less than 0.001) in corneal plaque cells at target: effector cell ratios of 1:10, 1:5, and 1:1. The [3H] thymidine incorporation was also highly significant (P less than 0.001) in squamous cell carcinoma cells at target:effector cell ratios of 1:10 and 1:5, but of less significance (P less than 0.01) at 1:1. Further, the ability of bovine monocytes to kill target cells was enhanced by culturing effector cells for 3 days before assay. These data show that bovine monocytes are cytotoxic for tumor cells in vitro and indicated that bovine monocytes may be of importance in the control of neoplastic proliferation in vivo.

Levamisole potentiation of antigen specific lymphocyte blastogenic response in Brucella abortus exposed but nonresponsive cattle.

Incubation of Brucella abortus (field strain) infected and strain 19 vaccinated bovine peripheral blood lymphocytes with B. abortus antigen and levamisole caused a consistently significant increase in [3H] thymidine uptake when compared to cultures without levamisole. Levamisole did not potentiate B. abortus-induced blastogenic response of lymphocytes from non-exposed cattle. A dose response study showed that 10 micrograms/culture induced maximum potentiation of B. abortus-induced lymphocyte stimulation. Using the 10 micrograms/well concentration of levamisole, further studies were conducted to determine the net potentiation of the blastogenic responses in lymphocytes from B. abortus (field strain) infected cattle. B. abortus strain 19 vaccinated but nonresponsive and non-exposed cattle. Levamisole significantly potentiated the B. abortus-induced lymphocyte blastogenesis in lymphocytes from unresponsive cattle.

Effect of cooperative, competitive, and individualistic experiences on self-esteem of handicapped and nonhandicapped students.

Compared are the effects of cooperative, competitive, and individualistic learning experiences on the self-esteem and perceived personal acceptance by the teacher of nonhandicapped and severely handicapped students in a mainstream setting. Thirty students (18 nonhandicapped and 12 trainable retarded) were randomly assigned to conditions stratified on the basis of handicap and sex. They participated in a bowling class which met for one hour per week for eight weeks. The results indicate that those in the cooperative condition had higher self-esteem and perceived more personal acceptance from the teacher than did the students in the competitive and individualistic conditions. Handicapped students reported higher self-esteem than nonhandicapped students.

Development of a migration inhibitory factor assay under agarose of bovine mononuclear leukocytes, using an antigen of Brucella abortus.

A study was conducted to develop a migration inhibitory factor assay under agarose of bovine mononuclear leukocytes, with an antigen of Brucella abortus. Different concentrations of mononuclear leukocytes were prepared by the Ficoll-Hypaque technique from the blood of nonvaccinated calves and from calves previously vaccinated with strain 19. Concentrations of 0.5, 1, 2, 3, 4, and 5 x 10(6) leukocytes were suspended in RPMI-1640 medium and various dilutions (20, 10, 1, and 0.1 microgram) of B abortus-soluble antigen, dispensed in triplicate wells cut in 1% agarose containing minimal essential medium and 10% bovine fetal serum. These agarose plates were incubated for 4-, 8-, 12-, 16-, 20-, and 24-hour periods and then were fixed; leukocytes were stained with Wright's stain. Migration distances were measured, and statistical analyses of the data revealed a concentration of 2 x 10(6) cells/well and an antigen concentration of 10 microgram/well. An incubation period of 20 hours was optimal for the assay.
