Constructing deletions with defined endpoints in Drosophila.

Chromosomes bearing small deletions are valuable tools in Drosophila genetics. We have investigated a method for efficiently constructing precise chromosomal deficiencies. Two P transposable elements were positioned within a progenitor strain at the sites of the desired deletion endpoints. Deletions spanning the two transposons were recovered at high frequency when P element transposase was expressed in these flies, but only if the flanking P elements were in a cis rather than a trans configuration. Appropriate progenitor strains can now be constructed to delete virtually any chromosomal region by utilizing an extensive collection of lines containing single P element insertions throughout the Drosophila genome.

Phenytoin dosage individualization--five methods compared in the elderly.

Five methods to predict phenytoin dosage have been compared in nine continuous care elderly patients. For each patient three steady-state plasma concentrations were obtained at three different doses. The data were used to estimate the 'optimum' dose for each patient by direct linear plot. The optimum dose for each patient was predicted from each plasma concentration using five dosage prediction methods based on the Michaelis-Menten equation using: (i) the population mean Vmax, (ii) the population mean KM, (iii) the linearized Bayesian method, (iv) the Rambeck nomogram, and (v) two plasma concentration-dose data pairs to estimate both Vmax and KM. The predictive precision was similar for each of methods (i-iv). Ninety-six out of 126 dosage predictions with the five methods were within 25 mg of the optimum dose. Methods (ii) and (iv) tended to overpredict dosage, particularly when used to interpret low plasma phenytoin concentrations.

Synovial fluid concentration of five different cytokines in rheumatic diseases.

Interleukin-1 beta, interleukin-2, tumour necrosis factor alpha, and the interferons, alfa and gamma, were measured concurrently in synovial fluid samples from 68 patients with rheumatic diseases. Mean interleukin-1 beta concentrations (130.3 (SD 22) pg/ml) were higher in synovial fluids from patients with rheumatoid arthritis (RA) than in those from patients with osteoarthritis (27.8(4.5)pg/ml), while measurements in synovial fluids from patients with seronegative spondarthritis were intermediate (72.7 (32) pg/ml). Interleukin-2 and tumour necrosis factor alpha concentrations were lower in the inflammatory arthropathies (RA: 4.5 (0.6) U/ml, 0.39 (0.04) ng/ml; seronegative spondarthritis: 3.1 (0.3) U/ml, 0.33 (0.03) ng/ml respectively) than those in patients with osteoarthritis (5.2 (0.6) U/ml; 0.05 (0.04) ng/ml). Interleukin-2 and tumour necrosis factor alpha concentrations correlated in all groups (r = 0.7), as did the interferons alfa and gamma (r = 0.7). There was no relation between interleukin-1 beta and either interleukin-2 or tumour necrosis factor alpha, or between the interferons and any other cytokine. Several distinct cytokine patterns were noted. Synovial fluids from two non-arthritic subjects were also examined: interleukin-1 beta concentrations were low, but concentrations of the other cytokines were higher than those seen in most arthritic fluids.

The effects of chronic lithium treatment on psychomotor performance related to driving.

A group of 16 psychiatric out-patients in remission, who had been taking lithium carbonate as their sole medication for at least three months, were compared with a control group of 22 healthy volunteers. On a computerised driving simulator which produced measures of reaction time, tracking ability and mistakes made, the patient group had a significantly slower reaction time. Patients should be warned therefore that lithium may affect their ability to drive or operate machinery and that psychomotor impairment and sedation are not synonymous.

Organisation studies: time for change.

Young managers, argues David Thompson, need to understand organisations, but do not at present learn anything like as much as they could from organisation theory. He therefore proposes a revision of the IHSM syllabus in the subject.

Artificial intelligence-guided analysis of cytologic data.

A design for the integration of artificial intelligence (AI) technology with large databases of clinical and objective cytologic data, such as are on file at the University of Chicago, is presented. Among the key features of this approach are the use of a knowledge representation structure based upon an associative network, the use of a Bayesian belief network as a method of managing uncertainty in the system, and the use of neural networks and unsupervised learning algorithms as a means of discovering patterns within this database. Such an automated approach is necessary, given the complexity and interdependence of these data, to gain an understanding of their dependence structure and to assist in their exploration and analysis.

Tissue architectural features for the grading of prostatic carcinoma.

In research for the development of a computer-aided workstation for the objective grading of prostatic carcinoma, tissue architectural (histometric) features were analyzed in ten cases each of well-differentiated, moderately differentiated and poorly differentiated carcinoma (as subjectively graded by the consensus of a panel of experts). Sections were cut at 4 microns, stained by the Feulgen reaction and digitized by two different video-based photometric systems. Some images were interactively segmented, considering the histometric clues to be studied; others were automatically segmented by an expert system-guided technique. The latter procedure produced good results, with over 90% of the nuclei judged to be correctly segmented in 64% of the fields studied and over 80% in another 24% of the fields. While the number of nuclei per field provided some separation of well-differentiated from other lesions, the number of nuclei per gland distinguished between well-differentiated and moderately differentiated lesions. Simplicial decomposition of the images also provided a measure of the degree of differentiation, as did the "texture" of the nuclear placement, based on two run-length statistics. Combination of the run-length features distinguished the three categories of lesions with statistical significance. The results of this study provided insights into the problems (such as the effect of field boundaries) faced in the design of an computer-aided grading system. They also showed the value of expert system-guided scene segmentation and of such histometric features as the field cellularity and the number of nuclei per gland for the discrimination between lesions of different grades of differentiation.

Independence and status--two different work methods.

This paper describes a study to establish whether there were hidden benefits from the changes occurring in nursing work methods. Of particular interest were the feelings of independence and status of qualified nursing staff operating within two discrete work environments, namely a) primary nursing, b) task allocation nursing. A nine item questionnaire with Likert scale was used as the measure. It was hypothesised that those nurses working with primary nursing would 1) feel more independence and, 2) have a higher sense of status than their counterparts in the traditional task-centred environment.

Changes in food patterns during a low-fat dietary intervention in women.

The Women's Health Trial was initiated by the National Cancer Institute to study the effects of a low-fat diet on the incidence of breast cancer in women at elevated risk for the disease. The purpose of this article is to examine the specific dietary changes that 173 women made while participating in a feasibility intervention program to reduce their fat intake to approximately 20% of total calories over a 12-month period. The intervention program used group sessions to teach nutrition information and behavioral skills necessary to make a life-style dietary change. Four-day food records were collected from participants at the beginning of the study and again at 12 months. Women in the intervention group reduced their total fat intake from a mean of 76 gm (39% of total energy) to 31 gm (22% of total energy), mainly by decreasing their fat intake from milk products, red meats, and fats/oils. These women used cheddar cheese, American cheese, whole milk, butter, mayonnaise, salad dressing, bacon, and hamburgers less frequently, and used diet American cheese, low-fat cottage cheese, and skim milk more frequently. They consumed less fat in their vegetable dishes, and their total caloric intake from fruit increased slightly. In addition, the overall quality of the diets improved, since there was a 20% to 50% increase in the energy-adjusted intake of vitamins and minerals from food sources.

Lacrimal gland dosimetry for the brain imaging agent technetium-99m-HMPAO.

We calculated lacrimal gland dosimetry for the brain imaging agent 99mTc-HMPAO. One hundred thirty-eight patients were studied using a dedicated brain imaging device. Only 11% of the patients showed lacrimal gland uptake. For a 740-MBq (20 mCi) injected dose of 99mTc-HMPAO, the radiation exposure to the lacrimal glands is 10.2 mGy (1.02 rad), or 0.0138 mGy/MBq (0.051 rad/mCi). These values are five times lower than the ones reported by the manufacturer of HMPAO. As the dosimetry calculation is only for subjects showing 99mTc-HMPAO uptake in the lacrimal gland, the average radiation dose to all subjects is considerably lower, 1/10 of the estimated value.

Hodgkin's cell lectin, a lymphocyte adhesion molecule and mitogen.

We have previously shown a novel galactose/N-acetylgalactosamine specific lectin activity (Hodgkin's disease (HD) lectin) on the surface of cultured HD cells (lines L428, its variants, and line L540) to mediate lymphocyte adhesion. We here demonstrate that both surface membrane-bound and secreted HD lectin activities participate in the activation of agglutinated lymphocytes. Among known adhesion molecules expressed by the HD cells, only the intercellular adhesion molecule-1 (ICAM-1) contributed to this activation as an alternative PBL binding site. As yet we have not identified the cellular ligand(s) for the HD lectin on the lymphocyte surface. Pretreatment of lymphocytes with mAb to the accessory molecules CD2, CD3, CD4, CD8, CD11b, or CD11c did not interfere with their response to HD cells. mAb to CD11a (LFA-1), the alleged ligand of ICAM-1, inhibited the ICAM-1 but not the HD lectin-mediated lymphocyte stimulation. Although lymphocyte binding could proceed via either pathway, lymphocyte activation always depended upon factors secreted by the HD cells, one of which we identified as a soluble form of the HD lectin based on its shared properties with the membrane-bound form including immunologic cross-recognition and carbohydrate-binding specificity. Although HD cell-conditioned medium alone stimulated lymphocytes, HD cell plasma membranes could compensate for low concentrations of this medium. In addition, resting lymphocytes, normally unresponsive, were triggered into DNA synthesis by growth medium when cocultured with HD cell membranes. The unique functions of the surface-expressed HD lectin and its soluble counterpart as lymphocyte adhesion molecule and mitogen might be physiologically relevant to the severe immunodeficiencies occurring in patients with HD.

Studies on the anti-inflammatory activity of ebselen. Ebselen interferes with granulocyte oxidative burst by dual inhibition of NADPH oxidase and protein kinase C?

Ebselen (PZ51, 2-phenyl-1,2-benzoisoselenazol-3-(2H)-one) was shown to be an inhibitor of human granulocyte oxidative burst stimulated by phorbol myristate acetate (IC50 25 microM). Estimation of the primary oxygen metabolites of the burst was complicated by the redox chemistry of Ebselen. Ebselen inhibited NADPH-stimulated superoxide generation by a partially purified NADPH oxidase preparation with an IC50 of 0.5-1.0 microM. Ebselen was also shown to inhibit the activity of partially purified Ca2+- and phospholipid-dependent protein kinase C (IC50 ca. 0.5 microM). Phorbol ester-stimulated phosphorylation of protein in intact cells was inhibited by Ebselen (IC50 ca. 50 microM). These pharmacodynamic properties of Ebselen are discussed in terms of its anti-inflammatory activity in vivo. The findings are also discussed in terms of Ebselen's known ability to interact with sulfhydryl components of cells, particularly critical thiol components of the enzymes studied.

Peroxidase-catalyzed oxidation of eugenol: formation of a cytotoxic metabolite(s).

The oxidation of eugenol (4-allyl-2-methoxyphenol) by horseradish peroxidase was studied. Following the initiation of the reaction with hydrogen peroxide, eugenol was oxidized via a one-electron pathway to a phenoxyl radical which subsequently formed a transient, yellow-colored intermediate which was identified as a quinone methide. The eugenol phenoxyl radical was detected using fast-flow electron spin resonance. The radicals and/or quinone methide further reacted to form an insoluble complex polymeric material. The stoichiometry of the disappearance of eugenol versus hydrogen peroxide was approximately 2:1. The addition of glutathione or ascorbate prevented the appearance of the quinone methide and also prevented the disappearance of the parent compound. In the presence of glutathione, a thiyl radical was detected, and increases in oxygen consumption and in the formation of oxidized glutathione were also observed. These results suggested that glutathione reacted with the eugenol phenoxyl radical and reduced it back to the parent compound. Glutathione also reacted directly with the quinone methide resulting in the formation of a eugenol-glutathione conjugate(s). Using 3H-labeled eugenol, extensive covalent binding to protein was observed. Finally, the oxidation products of eugenol/peroxidase were observed to be highly cytotoxic using isolated rat hepatocytes as target cells.

Distortion of spatial selectivity by pattern onset stimulation.

Spatial selectivity of pattern evoked potentials has been thought to provide evidence of lateral inhibition. However, spatial tuning functions may be distorted by pattern onset stimulation that is applied repeatedly to the same area of retina so an after-image is formed. This only applies at low spatial frequencies because of the randomizing effects of eye movements. Low-frequency attenuation may therefore be exaggerated. Pattern reversal stimulation has the opposite effect and this is reflected in the literature by fewer reports of bandpass functions. A new method has therefore been devised to provide the true spatial response function. The spatial phase is reversed after every two consecutive presentations. By combining this paradigm with a correction for the optical transfer function of the eye, the true neural response function is obtained. Ten subjects participated in this study to evaluate the distortion of spatial selectivity in the pattern electroretinogram. The new stimulus paradigm reduced the low spatial frequency attenuation to a barely significant level giving an almost flat amplitude response for the + ve and - ve transients of the pattern electroretinogram for check sizes from 222' to 7' angular subtense. However, correction for optical degradation produces bandpass curves, which closely correspond to those predicted from recent data on receptive fields of primate retinal ganglion cells.

The effect of stimulus contrast on the latency and amplitude of the pattern electroretinogram.

Our studies have verified that a linear relationship exists between the amplitude of the pattern onset ERG and retinal contrast, but indicate that there is no systematic variation in peak latency with contrast. The optical degradation of high spatial frequency patterns cannot therefore provide an explanation for the variation in peak latency with spatial frequency.
