Autolytic Activity and an Autolysis-Deficient Mutant of Clostridium acetobutylicum.

The optimum conditions for autolysis and autoplast formation in Clostridium acetobutylicum P262 have been defined. Autolysis was optimal at pH 6.3 in 0.04 M sodium phosphate buffer, and the bacterium produced latent and active forms of an autolytic enzyme. The ability of cells to autolyze decreased sharply when cultures entered the stationary phase. Autoplasts were induced by 0.25 to 0.5 M sucrose and were stable in media containing sucrose, CaCl(2), and MgCl(2). A pleiotropic autolysis-deficient mutant (lyt-1) was isolated. The mutant produced less autolysin than did the parent P262 strain, and it had an altered cell wall which was more resistant to both its own and P262 autolysins. The mutant formed long chains of cells, and lysozyme was required for the production of autoplasts. Growth of the P262 strain or the lyt-1 mutant was inhibited by the same concentrations of penicillin, ampicillin, and vancomycin. The lyt-1 mutant strain treated with the minimum growth-inhibitory concentration of penicillin autolyzed upon the addition of wild-type autolysin to the autolysis buffer at the same rate as did the untreated P262 strain. Chloramphenicol did not protect the penicillin-treated lyt-1 cells against autolysis enhanced by exogenous wild-type autolysin.

Effect of oxygen on liquid holding recovery of Bacteroides fragilis.

Liquid holding recovery (LHR) in ultraviolet-irradiated Bacteroides fragilis cells occurred under aerobic conditions but was inhibited by anaerobic conditions. The increase in survival after aerobic LHR resulted in an increase in the shoulder regions of the ultraviolet survival curves. Maximum LHR was obtained after holding the cells for 2 to 3 h. LHR was temperature dependent, and in stationary-phase cells LHR was independent of nutrients. Higher levels of LHR occurred in exponential-phase cells than in stationary-phase cells, and LHR was affected by nutrients in exponential-phase cells. Sublethal concentrations of caffeine and acriflavine inhibited LHR. In addition to LHR, minimal medium recovery also occurred in the concentration of [3H]thymine-containing dimers in the acid-insoluble fraction of the cells. A corresponding increase in [3H]thymine-containing dimers was observed in the acid-soluble fraction after LHR. Although a small proportion of irradiated cells produced filaments, this phenomenon was not directly related to LHR in B. fragilis.

Serum theophylline levels in adult asthmatics: comparison of oral sustained-release and microcrystalline preparations.

Serial serum theophylline levels were measured in 20 inpatient asthmatics given tablets of either microcrystalline or sustained-release theophylline. The sustained-release preparation gave serum levels which were more consistently within the therapeutic range with a significantly higher trough concentration. No increase in adverse effects was encountered.

Carbon-13 magnetic resonance probe of coenzyme and inhibitor binding in liver alcohol dehydrogenase.

Horse liver alcohol dehydrogenase was carboxymethylated at the active site Cys-46 using 90% [1-13C]bromoacetate. The enriched carboxylate resonance was studied in the absence and presence of coenzymes and inhibitors. Previous ambiguities regarding the resonance in the NAD+ complex have now been resolved. However, it is shown that imidazole, an inhibitor introduced during the carboxymethylation, is not removed by the standard gel filtration step frequently employed and must thus bind much more tightly to the enzyme than suspected. Competition experiments involving imidazole and halide inhibitors show that the imidazole is preferentially bound when both are present in equimolar amounts. This suggests that the crystallographically identified anion binding site at the zinc of the carboxymethylated enzyme may require re-evaluation. The electron density at the zinc of the modified enzyme may be better explained as being due to unsuspected binding of imidazole.

Rifampin and bacteriocin resistance in Bacteroides fragilis.

A low-molecular-weight bacteriocin produced by a Bacteroides fragilis strain inhibited ribonucleic acid polymerase activity in crude extracts of a susceptible B. fragilis indicator strain. A total of 10 rifampin-resistant mutants of the indicator strain were isolated. Nine of the rifampin-resistant mutants were resistant to the bacteriocin, and the other mutant was hypersusceptible. The rifampin- and bacteriocin-resistant mutants all adsorbed approximately the same amount of the bacteriocin as the indicator strain. Two of these rifampin- and bacteriocin-resistant mutants were investigated further, and the polymerase activity in crude extracts of the two mutants was not affected by either rifampin or the bacteriocin. The in vitro ribonucleic acid polymerase activity of the hypersusceptible strain was more susceptible to the bacteriocin than the parent indicator strain was. The bacteriocin-producing strain was susceptible to rifampin but was resistant to its own bacteriocin in vivo. The in vitro ribonucleic acid polymerase activity of the producer strain was only slightly affected by 64 arbitrary units of the bacteriocin. Increasing concentrations of the bacteriocin inhibited ribonucleic acid polymerase extracts of the producer strain.

Characterization and mode of action of a bacteriocin produced by a Bacteroides fragilis strain.

A Bacteroides fragilis strain produces a low-molecular-weight (13,500 to 18,700), proteinaceous bacteriocin during the stationary growth phase. The extracellular bacteriocin is not inducible by ultraviolet light or mitomycin C and is stable between pH 7.5 and 8.2. The majority of the bacteriocin is thermolabile, but a small proportion (3%) of the bacteriocin is stable after autoclaving at 121 degrees C for 15 min. Killing of sensitive bacteroides cells follows single-hit kinetics, and the interaction of a single molecule of bacteriocin with a target cell occurs in two stages. The killing of susceptible cells is affected by temperature and the growth state of the susceptible cells. The bacteriocin is unusual in that the primary event in its mode of action is the inhibition of RNA synthesis. The bacteriocin inhibits RNA synthesis immediately but has no effect on DNA synthesis or intracellular ATP levels. Protein synthesis is inhibited after a delay of 20 min, presumably as a result of the initial inhibition of RNA synthesis.

Exercise electrocardiography and myocardial perfusion imaging in the diagnosis of coronary artery disease: preliminary report.

Exercise electrocardiography and rest/exercise myocardial perfusion imaging with thallium-201 were performed in 43 patients with typical angina or atypical chest pain; the results were correlated with those of coronary arteriography. Exercise electrocardiography sensitivity was 65%, specificity was 78%, predictive value for a positive result was 73% and for a negative result was 93%. The low sensitivity of the exercise electrocardiogram was mainly due to the number (13 of 43, 30%) of inconclusive results (no ST-segment change on the electrocardiogram, but failure to attain the target rate), most of which were in the group with typical angina. The predictive value of exercise electrocardiography for both a positive and negative result was excellent in typical angina. In patients with atypical chest pain, the negative predictive value was high (90%) but the positive predictive value was very low (50%). The sensitivity of myocardial perfusion imaging was 71%, specificity was 59%, positive predictive value was 52% and negative predictive value 89%. The low specificity of this test is related to the number of false-positive results obtained, most of which occurred in the group with atypical pain. When the results of exercise electrocardiography and myocardial perfusion imaging are combined, the sensitivity is increased but specificity is unacceptably low. However, myocardial perfusion imaging in patients with an inconclusive result from exercise electrocardiography (most of them in the group with typical angina) showed a sensitivity of 80%, specificity of 88%, positive predictive value of 80% and negative predictive value of 100%.

Usefulness of three additional electrocardiographic chest leads (V7, V8, and V9) in the diagnosis of acute myocardial infarction.

Additional electrocardiocardiographic chest leads (V7, V8, and V9) were used in 117 persons consecutively admitted to a coronary care unit. Among the 46 (39%) with a proven acute myocardial infarction the electrocardiograms (ECGs) of 9 (20%) showed ST-segment elevation or abnormal Q-waves, or both, in the three additional leads. In six of the nine, such changes were associated with signs of anterolateral or inferior wall infarction (in three each) on the standard 12-lead ECG, but in the other three (7% of the 46) electrocardiographic changes diagnostic of acute myocardial infarction were found only on the additional chest leads; the last three had characteristic changes in serum enzyme concentrations. This study showed that additional chest leads are helpful in detecting myocardial injury of necrosis in areas of the heart not properly reflected on the standard 12-lead ECG.

Sources of fast neutrons for radiotherapy.

There is currently great interest in the use of fast neutrons for radiotherapy. Although it is too early for the long-term effects to be known, the results so far are encouraging. Three types of fast neutron-producing devices are available: neutron generators, compact cyclotrons and large cyclotrons. For the requirements of radiotherapists the most suitable and practical source currently available is the reaction 9Be(d,n) in conjunction with a large cyclotron capable of accelerating deuterons to energies greater than 35 MeV.
