Measurement of tissue disulfide groups by Alcian blue 8GX and atomic absorption spectrophotometry.

The primary purpose of this study was to quantify tissue disulfides with Alcian blue 8GX using atomic absorption spectrophotometry (ASS) to measure the copper moiety of the Alcian blue. The secondary purpose was to determine whether aqueous or alcoholic formalin was a better fixative for this procedure. Sections of mouse renal cortex fixed in 10% neutral formalin (NBF) or in ethanol/formalin/acetic acid (AFA) were stained in 0.5% Alcian blue 8GX in 0.8 M MgCl2 with prior thiosulfation (+Th) or without prior thiosulfation (-Th). The relative contents in the tissues of the copper of Alcian blue, measured by AAS, may be summarized as follows: -Th NBF, 1.15 micrograms Cu/mg tissue; -Th AFA, 1.35 micrograms Cu/mg tissue; +Th NBF, 1.99 micrograms Cu/mg tissue; +Th AFA, 3.25 micrograms Cu/mg tissue. These data indicate that 1) disulfide may be measured by this procedure, and 2) AFA is the preferred fixative for measurement of both disulfides and native sulfates.

Reducing severe diurnal bruxism in two profoundly retarded females.

Several diurnal audible teeth grinding (bruxism) was found to affect 21.5% of a profoundly retarded population. However, no previous research has treated bruxism in retarded individuals. In the current study a multiple baseline across subjects design was used to assess the effectiveness of contingent "icing," brief contingent tactile applications of ice, as a treatment for bruxism. Three 15-minute treatment periods and two 5-minute generalization periods were conducted 5 days per week. One resident displayed a 95% reduction in the percentage of intervals during which bruxism occurred during treatment periods and a 67% reduction during generalization periods. The other resident displayed a 94% reduction in the percentage of intervals during which bruxism occurred during treatment periods and a 53% reduction during generalization periods.

Phase I study of L-alanosine using a daily x 3 schedule.

L-Alanosine is an antitumor antibiotic that inhibits adenine synthesis. It showed significant activity in animal tumor systems. Using a daily x 3 dose schedule every 3 weeks, we performed a phase I study to determine toxicity in man. Doses of 8-375 mg/m2/day x 3 were administered to 49 patients in 117 courses. The dose-limiting toxic effect was mucositis. Vomiting, infrequent myelosuppression, fever, headache, malaise, and blood pressure changes were detected at higher dose levels. No antitumor activity was noted. Toxicity with this regimen is acceptable. A phase II study with doses of 250 mg/m2/day x 3 every 3 weeks is feasible.

Enterobius vermicularis infestation of the female genital tract causing generalised peritonitis. Case report.

Enterobius (Oxyuris) vermicularis is the only nematode which infests man. It inhabits the large intestine and is of low pathogenicity. The ova are occasionally found in ectopic sites within the peritoneal cavity, usually as an asymptomatic granuloma. We describe the first patient to present with generalised peritonitis caused by rupture of a tubo-ovarian abscess containing the ova of Enterobius vermicularis.

Autologous bone marrow transplantation in relapsed adult acute leukemia.

From March, 1976 to February, 1979, 28 cases of adult acute leukemia of which 24 were evaluable were treated in irreversible relapse with high dose chemotherapy (piperazinedione) and supra-lethal total body irradiation (TBI) in conjunction with autologous bone marrow transplantation (ABMT). The marrow cells grafted were collected and stored in liquid nitrogen at the time of remission. In 12 patients the marrow cells were fractionated using discontinuous albumin gradients in an attempt to separate normal cells from residual leukemic cells. Twelve patients achieved complete remission (CR); in 9 additional patients signs of engraftment were evident but death occurred before achievement of CR. Seven of 12 AML patients, which were treated with bone marrow transplantation as first treatment of their relapse, achieved CR. Four of 5 patients with ALL, whose bone marrows were collected during first remission, reached CR. The median CR duration was 4+ months and the median survival of the patients reaching CR was 6+ months. Autologous bone marrow transplantation offers a good chance of CR (66%) when marrow is collected during first remission and used as first treatment for AML in third relapse and ALL in second relapse.

Ventricular aneurysm: false or True? An important distinction.

A case of ventricular false aneurysm withe the typical clinical, radiological, electrocardiographic, and angiographic features of this entity is presented. The distinction between false ventricular aneurysm and true aneurysm is discussed. This distinction is important because of the propensity of false aneurysms to rupture. An early diastolic murmur was present prior to, but not after, resection of the aneurysmal sac. A theory as to the origin of this murmur is offered.

Drug 'administration' via breast milk.

In the current popularity of breast-feeding, certain pitfalls for the infant tend to be overlooked. Physicians should be particularly aware with respect to which drugs and toxins can be transmitted by the lactating mother, and which cannot. In aid of such awareness, the authors tabulate the excretion characteristics and compatibility with breast-feeding of more than 200 common drugs and substances.
