A histopathologic study of the Choyce VIII intraocular lens.

We performed a postmortem ocular examination on a 52-year-old man who had had Choyce VIII intraocular lenses implanted in both eyes. Visual acuity with both lenses was 6/6 (20/20). There were no clinical complications before the patient's death. Marked stretching of the chamber angle occurred at all points of fixation, with ultimate loop fixation into the substance of the ciliary body and, in some instances, erosion close to the greater arterial circle of the ciliary body. Where iris tucking had occurred, the iris tucked over the foot of the lens was extremely thin with the foot and the tucked iris in the pars ciliaris region.

The fate of oxalic acid in the Wistar rat.

1. An established procedure for determining oxalate in human urine has been modified for rat urine. The daily excretion of oxalate by the male Wistar rat is 570-650 microgram. 2. Oxalate excretion in rat urine following i.p. administration of [14C]oxalic acid (1-70 mg/kg) has been studied. The rate and degree of excretion are dose-dependent. 3. The excretion of urinary oxalate by the rat has been quantified after administration of two oxalate-producing xenobiotics, alpha-chlorohydrin and 1,2-dibromo-3-chloropropane. 4. Oxalate inhibits the metabolism of glucose and lactate by isolated rat kidney tubules in vitro.

Inhibition of interferon production and antiviral action in mouse cells by 4,5',8-trimethylpsoralen activated with near ultraviolet light. Brief report.

The DNA of mammalian cells treated with 4,5',8-trimethylpsoralen (Trioxsalen) is crosslinked (preventing transcription) only after photoactivation with near ultraviolet light. Suppression of gene action in cocultivated or fused cultures, including both induction and antiviral action of fibroblast interferon, can consequently be limited to pretreated cells, an advantage over actinomycin treatment.

Kinetics of decay in the expression of interferon-dependent mRNAs responsible for resistance to virus.

We used 5,6-dichloro-beta D-ribofuranosyl-benzimidazole (DRB), a selective and reversible inhibitor of mRNA production, to investigate the regulation of the pathway leading to resistance to viruses in cells treated with interferon (IF). DRB allows initiation of transcription but promotes premature termination of the nucleotide chains, so that it abolishes interferon-dependent protection against viruses. When the DRB is removed, synthesis of complete mRNAs can resume. Mouse L-929 cells were exposed to 100 microM DRB before and during a 1-hr pulse of IF followed by treatment with antibody to IF to prevent cell-to-cell spread of IF after that time. At different intervals thereafter the cells were washed and the DRB was replaced by medium; after further incubation, the cells were infected with vesicular stomatitis virus. Resistance to virus was inversely proportional to the duration of the block imposed by DRB. When the DRB was removed soon after the IF pulse, substantial protection from virus ensued, but none developed when removal of the DRB was deferred for 5-6 hr. Cells exposed to DRB for 5 hr, then pulsed with IF for 1 hr, still mounted a strong antiviral response. The data show that the ability of cells to resist viral infection decays within 5-6 hr after treatment with IF. Whether the decay is due to shutoff of transcription of mRNAs, or to their destruction or degradation, or whether regulation takes place at one or more subsequent steps in the antiviral pathway, remains to be determined.

The diagnosis and treatment of functional disorders of the biliary tract.

An approach to the diagnosis and treatment of patients with presumed functional disorders of the biliary tract (biliary dyskinesia) is described. The current diagnostic criteria are pain compatible with biliary pain in the absence of gallstones and other organic gastrointestinal disease, or other disorders which might produce abdominal pain, together with reproduction of the patient's symptoms by cholecystokinin, or morphine, or both. Other diagnostic methods are described together with their limitations. The results of operation in 38 of 45 patients seen in this Unit during the past six years are presented. The results were poor in 20% of patients, but two-thirds of the group have had good results in the short term.

Operative liver biopsy abnormalities in patients with functional disorders of the biliary tract.

The group of conditions variously termed biliary dyskinesia, acalculous cholecystitis, biliary pain without stones, or functional disorders of the biliary tract, is poorly defined clinically, and no consistent pathological abnormalities have been previously described in patients with this diagnosis. In this paper we report histological abnormalities encountered in operative live biopsies in such patients. The criteria for the diagnosis of a functional biliary tract disorders were: pain typical of biliary pain, negative results of investigations for organic biliary tract or other gastrointestinal disease, and reproduction of the patient's symptoms by cholecystokinin, or morphine, or both. Twenty of 45 patients with a presumptive diagnosis satisfied these criteria, and had a wedge liver biopsy at the time of operation. The 20 liver biopsy specimens were compared in a blind fashion with similar ones taken from patients having diagnostic laparotomies; patients with stones confined to the gallbladder; patients with gallstone pancreatitis; and patients with proven common bile duct stones. The biopsy findings were found to be similar to those in the latter two groups. Thus the abnormalities were similar to those found in partial or intermittent biliary obstruction, and it is suggested that they may be due to intermittent increases in biliary pressure.

Correlation of e antigen, DNA polymerase activity, and Dane particles in chronic benign and chronic active type B hepatitis infections.

The e determinant of hepatitis B surface antigen (HBS Ag) was found in 23 of 42 patients with chronic hepatitis B virus (HBV) infection. Presence of e antigen was associated with increases in DNA polymerase activity and in the number of circulating Dane particles. In the group with detectable e antigen, the average DNA polymerase activity was 367+/-78 counts per minute (cpm; mean+/-standard error [SE]), and the average number of Dane particles counted in electron micrographs was 4.4% of the total HBS Ag. In contrast, e antigen-negative patients had an average DNA polymerase activity of 40+/-6.9 cpm (P less than 0.1) and an average Dane particle count equal to 0.6% of the HBS Ag. The e antigen was detected in 68% of patients who were HBS Ag carriers or had persistent viral hepatitis and 40% of those with chronic active type B hepatitis. Thus, the presence of e antigen correlated with both the chronicity and presence of infectious HBV. However, it did not correlate with the type or severity of liver disease after HBV infection, since e antigen was present in both chronic benign and chronic aggressive hepatitis B infections.

Similarities of two hepatitis A virus strains.

In outbreaks of type A hepatitis in Los Angeles, USA, and Rosario, Argentina, virus particles were isolated from faeces. The geographically diverse strains were identical in appearance and serological reactivity. They differed only in buoyant density, but other workers have also obtained inconsistent results in estimating this. We conclude that the virus strains in the two epidemics were identical.

Salmonella typhimurium infection in guinea pigs: observations on monitoring and control.

A spontaneous outbreak of salmonellosis in a guinea pig colony was caused by Salmonella typhimurium. A simple, comprehensive monitoring procedure, consisting of mass fecal sampling, was developed. Using the monitoring system described, the disease was eliminated by: 1) removal of animals in pens in contact with Salmonella, and 2) strict hygienic procedures. It was not necessary to depopulate the colony in order to control the disease.
