Serum paraproteins in chronic lymphocytic leukaemia.

The presence of paraproteins in the sera of 10 patients with chronic lymphocytic leukaemia (CLL) was investigated using immunoisoelectric focusing. Monoclonal immunoglobulins were found in nine of these 10 sera. Five sera contained a single monoclonal IgM paraprotein, one serum contained a single monoclonal IgG paraprotein, while three sera contained more than one monoclonal paraprotein--namely, IgM + IgD, IgM + IgG, and IgM + IgD + IgG. The results indicate that the malignant B cells of CLL may be at a later stage of differentiation than previously assumed and serum monoclonal immunoglobulin could be of value as a tumour marker.

The application of isoelectric focusing to routine screening for paraproteinaemia.

We present a scheme for routine screening of patients' sera for monoclonal immunoglobulin by isoelectric focusing in agarose gels. Using this method combined with immunofixation, we have been able to detect monoclonal immunoglobulin in the sera of patients, and to identify the isotype of the immunoglobulin when conventional techniques such as zonal and immunoelectrophoresis have yielded equivocal results.

Inhibition of the hypotensive effect of plasma protein solutions by C1 esterase inhibitor.

The hypotensive effect of stable plasma protein solution (SPPS) is a well-known phenomenon, causing serious problems in surgical patients and patients supported by cardiopulmonary bypass. Kinins are believed to be responsible. A study was made to prevent this hypotension by adding C1 esterase inhibitor to the SPPS and comparing this with 5% human albumin. The results shows a significant difference between the effects of SPPS and SPPS plus C1 esterase inhibitor infusion on bypass. Five percent human albumin did not cause hypotension. Possible sequelae are discussed.

Multiple receptors for brain dopamine.

This study was done to obtain direct in vitro evidence for the possible existence of more than one type of dopaminergic binding site in homogenates of the caudate nucleus from calf brain. Five radioligands for dopaminergic sites were tested. The inhibitions of two agonist radioligands ([3H]dopamine, [3H]apomorphine) by haloperidol, chlorpromazine, or piflutixol were biphasic. The inhibitions of [3H]haloperidol, [3H]spiroperidol, and [3H]dihydroergocryptine binding by dopamine and (-)-norepinephrine were also biphasic. Thus, both 3H-labeled agonists and 3H-labeled antagonists were possibly binding to two high-affinity sites.

Cyclic AMP and protein kinase in the spontaneously hypertensive rat aorta and tissue-cultured aortic smooth muscle cells.

Cyclic AMP levels and protein kinase activity were determined in the aortas of rats with normotension, moderate and severe spontaneous hypertension. While the cyclic AMP levels were reduced in the aortas from rats with moderate and sever hypertension the protein kinase levels were found to be elevated only in the aortas from rats with severe hypertension. We have grown in tissue culture, aortic smooth muscle cells from the normotensive and severely hypertensive rat. Cultured cells from both strains have similar growth patterns and morphology. The differences seen in cyclic AMP and protein kinase levels in the intact aortas are also seen in the aortic smooth muscle cells in cluture.

A novel form of diffusion battery.

A novel form of diffusion battery uses a series of 635-mesh screens instead of the customary cluster of circular tubes or holes. Fifty-five screens are arranged in series so that the amount of aerosol penetrating through different numbers of screens can be measured with a condensation nuclei counter. This "filter stack" must be calibrated because theory is not available to calculate the performance of such screens with sufficient accuracy. For calibration a polydisperse aerosol such as ambient or silver aerosol is used to compare the performance of the screens with that of the cluster-tube and collimated-holes diffusion batteries, whose performance is known from theory. It was observed that a given number of screens is equivalent to a given number of batteries independent of the size or size distribution of the aerosol. This filter stack is very compact and light in weight (0.9 kg). It is particularly useful for measurements in the field.
