Bulk isolation in nonaqueous media of nuclei from lyophilized cells.

Intact lyophilized nuclei are obtainable from a variety of tissues, either in situ or in culture, by freezing at -156 degrees C, drying at -25 degrees C, and mechanical disassociation in glycerol at 2 degrees C. Centrifugal separation of nuclei is accomplished in an 85 : 15 by volume mixture of glycerol and 3-chloro-1,2 propanediol at 2 degrees C. The method gives homogeneous nuclear preparations in high yield with preservation of labile and water-soluble constituents.

Recent measures to further reduce dioxin impact on man and the environment in the Federal Republic of Germany.

During last year's Dioxin symposium at Tampere/Finnland a report was given on the legislative actions of the German Federal Government to reduce dioxin release into the environment (1). Since then, further progress has been made, especially to regulate chemicals and to establish orientation values for PCDD/F in cow's milk. An international dioxin symposium and hearing was held in Berlin in November 1992, which focused on the scientific base for legislative and other governmental action to minimize dioxin burden on man and the environment. Measurements from different media including breast milk indicate a decrease in dioxin release into the environment during the last several years in Germany.

Aging of liver: morphological and biochemical changes.

Light-microscopically there are no significant age-dependent changes in the liver, due to the slight increase in biochemically determined connective tissue components. Electron-microscopical comparison of young and old animals show a larger size and decreased numbers of liver cell mitochondria in older animals. Collagen synthesis in liver is accelerated during growth, as demonstrated by assay of the [3H] proline incorporation rates and of the specific activity of hydroxyproline, corresponding to a decrease in the prolyl hydroxylase activity with age. The neutral salt-soluble collagen fraction may be regarded as a parameter for collagen synthesis, which is activated during the growth period and then remains constant without age and sex differences. Not much is known about the significance of these changes with increasing age or about their initiating factors, although there are parallel findings for liver intoxication and inflammation.

Prolonged effect of an anesthetic dose of ketamine on behavioral despair.

The present study investigated the effect of a single, anesthetic dose of ketamine, a noncompetitive N-methyl-D-aspartate (NMDA) glutamate receptor antagonist, on behavioral despair, an animal model of depression. Separate groups of male Wistar rats injected with an anesthetic dose of ketamine (160 mg/kg ip) and tested 3, 7, or 10 days later showed significantly less immobility in the second of two forced-swim tests compared to saline-injected controls. Ketamine- and saline-treated animals did not differ significantly in the swim tests with respect to other behavioral measures, namely diving, jumping, and head shakes. The present findings point to an ameliorative effect of ketamine on behavioral despair and support the view that NMDA antagonists may have a beneficial effect on depression.

Marine biosurfactants, I. Screening for biosurfactants among crude oil degrading marine microorganisms from the North Sea.

Three bacterial strains of marine origin were isolated during a screening for biosurfactants among n-alkane degrading microorganisms. One strain-identified as Alcaligenes sp. MM1-produced a novel glucose lipid. In the case of Arthrobacter sp. EK 1 the well-known trehalose tetraester was found as major component. From another pure culture classified as Arthrobacter sp. SI 1, extracellular emulsifying agents with properties indicating high molecular weight substances were detected. Furthermore trehalose corynomycolates were found at up to 2 g/l. The isolated biosurfactants showed good interfacial and emulsifying properties.

[Further information on the alkali resistance test].

In 90 patients the alkali resistance (AR) according to Burckhardt was measured on the back of the hand and on the thigh. Subjects with healthy skin (n = 30) reacted in a normal way. Those with healed hand eczema (n = 30) had a diminished AR in both areas tested; subjects with atopic eczema (n = 30) presented an even more diminished AR. Thus, in this broadened scope, simultaneous determination of AR on the back of the hand and on the thigh with the AR-test seems to be valuable in the recognition of an eczematous disposition.
