Excretion of purine derivatives by ruminants: effect of exogenous nucleic acid supply on purine derivative excretion by sheep.

The present study examined the relationship between the supply of exogenous nucleic acid (NA) purines and their recovery as the derivatives hypoxanthine, xanthine, uric acid and allantoin in urine. Six lambs, totally nourished by intragastric infusions of volatile fatty acids (VFA) and casein (i.e. no rumen fermentation), were given by abomasal infusion a microbial NA concentrate at six levels (from zero to 24.5 mmol purines/d). The true digestibility between the abomasum and terminal ileum of the NA purines was measured in a separate experiment using three lambs. The relative proportion of urinary allantoin increased, and that of other derivatives decreased, as the amount of NA infused was increased. The relationship between total excretion of purine derivatives (Y; mmol/d) and exogenous purines absorbed (X; mmol/d) was Y = 0.84X + 0.150W0.75 e-0.25X, where W is body-weight (kg). This implies that the endogenous contribution to the total excretion of derivative decreased as the supply of exogenous purines increased, with an associated progressive replacement of de novo synthesis by exogenous purines. The model also implies that 0.16 of the purines were eliminated through routes other than derivative excretion in urine. Once excretion exceeded 0.6 mmol/kg W0.75 per d, endogenous excretion was effectively zero and thus Y = 0.84 X. In normally fed sheep, derivative excretion should therefore relate to the microbial purines and, hence, microbial protein absorbed according to these models. The changing proportions of allantoin and other derivatives in urine were probably due to changes in the relative importance of endogenous and exogenous purines as precursors.

Polyamine metabolism and uptake during Phaseolus vulgaris lectin, PHA-induced growth of rat small intestine.

Kidney bean lectin, PHA, stimulated the hyperplastic and hypertrophic growth of rat small intestine. This growth was preceded by a rapid accumulation of polyamines in the small intestine. However, since the lectin had little effect on in situ polyamine biosynthesis, most of the polyamines must have been of extracellular origin. To investigate the source of polyamines, both the luminal uptake and basolateral transport of polyamines by the rat small intestine were measured in vivo. Luminal polyamine uptake was apparently by passive diffusion, non-saturable, linearly dependent on concentration and its extent was not stimulated by PHA. In contrast, 14C-polyamines injected intraperitoneally were probably taken up by a transport system(s). Moreover, basolateral polyamine transport was stimulated in a time-dependent manner when small intestinal growth was stimulated by PHA. However, in keeping with the finding of polyamine accumulation prior to demonstrable growth of the tissue, stimulation by PHA of the polyamine transport system also preceded small intestinal growth. Stimulation of polyamine transport by luminal factors is possibly a general mechanism involved in intestinal adaptation.

[Inventory of fresh water snails in Mayotte, Comoro Islands].

No trematodosis is known at the present time in Mayotte despite the close historical relationships between the Mayotte Archipelago and the neighbouring island of Madagascar, which is heavily infested. It is noteworthy that no intermediate host has been described in previous studies in Mayotte. An inventory of fresh water snails was performed from 1985 to 1988, in 53 experimental stations during the rainy and the dry seasons: 3,940 snails of eight different species were collected and further identified. The sole occurrence of Lymnaea natalensis constitutes a new record in Mayotte where there are now 14 different species of fresh water snails. According to the role of Lymnaea natalensis as a vector of Fasciola gigantica, particularly in Madagascar, the veterinary control of cattle importation into Mayotte is of great interest.

The cyclo-oxygenase inhibitors indomethacin and ibuprofen inhibit the insulin-induced stimulation of ribosomal RNA synthesis in L6 myoblasts.

Insulin stimulated total RNA accretion and the incorporation of [3H]uridine into RNA in L6 skeletal-muscle myoblasts. Incorporation of uridine into the rRNA was measured after either separation of 18 S and 28 S rRNA species by agarose-gel electrophoresis or separation of dissociated 40 S and 60 S ribosomal subunits on sucrose density gradients. Both methods showed a stimulation by insulin of uridine incorporation into the RNA of the two subunits. Two non-steroidal anti-inflammatory drugs, indomethacin and ibuprofen, which inhibit the metabolism of arachidonic acid by the cyclo-oxygenase pathway, inhibited the insulin-induced accretion of total cellular RNA and the incorporation of uridine into the RNA of both ribosomal subunits. The effect of insulin was observed both by using a tracer dose of [3H]uridine (5 microM) and in the presence of a high concentration (1 mM) of uridine to minimize possible changes in intracellular precursor pools. Neither insulin nor indomethacin was found to affect the incorporation of uridine into the total intracellular nucleotide pool, or the conversion of uridine into UTP. The ability of inhibitors of arachidonic acid metabolism to prevent insulin-induced increases in RNA metabolism suggests that a prostaglandin or other eicosanoid is involved in the signal mechanism whereby insulin stimulates RNA synthesis.

Structure of (20R)-20,21-epoxy-19-norpregna-1,3,5(10)-triene-3,17 beta-diol.

C20H26O3, Mr = 314.4, orthorhombic, P2(1)2(1)2(1), a = 10.25 (1), b = 25.36 (2), c = 13.11 (1) A, V = 3410.14 A3, Z = 8, Dx = 1.225 g cm-3, lambda(Mo K alpha) = 0.71069 A, mu = 0.45 cm-1, F(000) = 1352, T = 293 K. The structure was solved by direct methods and refined to R = 0.051 for 2384 reflections with F/signa(F) greater than 5. Both the crystallographically independent molecules are in one 20R configuration and confirm predictions based on mechanistic and stereochemical arguments. In the crystal lattice the molecules form a two-dimensional network of hydrogen bonds approximately perpendicular to c.

The influence of Calicophoron microbothrium on the susceptibility of Bulinus tropicus to Schistosoma bovis.

A total of 480 snails were collected from 3 habitats on the Mau Escarpment, Kenya, and were identified as Bulinus tropicus. Of the 351 snails examined alive in London, 75 were infected with Calicophoron microbothrium, 39 with C. microbothrium and Schistosoma bovis, 1 with S. bovis, 24 with other species of trematodes and 212 were uninfected. Examination of digestive glands of B. tropicus either uninfected or infected with both C. microbothrium and S. bovis demonstrated that it is possible to differentiate between parasite and host enzyme activity using glucose phosphate isomerase. However, malate dehydrogenase enables a much clearer differentiation between the enzyme activity of the schistosome and that of the amphistome. Laboratory snail infection experiments demonstrated that it is possible successfully to infect B. tropicus with S. bovis if the snails have previously been exposed to miracidia of C. microbothrium.

The effect of clenbuterol on basal protein turnover and endogenous nitrogen loss of sheep.

Seven measurements of the effect of clenbuterol on basal nitrogen excretion (UNE), and protein turnover were made in six female sheep. The sheep were sustained by the intraruminal infusion of energy as volatile fatty acids to provide maintenance, but given no protein (N-free) for 12 d (6 d control, 6 d clenbuterol). Clenbuterol reduced UNE by 20%, but only on day 2 of the 6 d subperiod. Protein flux (equivalent to degradation on N-free nutrition), measured on day 6 by the irreversible loss of leucine was significantly increased (12%) by clenbuterol. Amino-N oxidation measured by N excretion was unchanged and, therefore, protein synthesis was also increased. During the 12 d N-free period, the recovery of urinary total N (Kjeldahl) as the sum of urea, ammonia, creatinine and purine derivatives, declined from 87.7 to 74.2%. The form of this missing N was not identified. The effect of clenbuterol of increasing both degradation and synthesis is unlike that reported in the literature for animals receiving protein when, in general, synthesis is unchanged and degradation reduced. This could be due to a different effect of clenbuterol in the N-free state, or to unchanged effects on protein pools other than muscle whose relative contribution to protein metabolism is different in the N-free state.

Improving physical therapy students' clinical problem-solving skills: an analytical questioning model.

The purpose of this study was to assess the effectiveness of a model for teaching problem-solving skills to first-year physical therapy students using the Watson-Glaser Critical Thinking Appraisal (CTA) and participant feedback. We used a pretest-posttest control group design. Subjects were 31 first-year physical therapy students divided into a Control Group (n = 15) and an Experimental Group (n = 16). Students in the Experimental Group used the problem-solving model during a four-week clerkship. No difference was found between the Experimental and Control Groups' performance on the CTA. Subjectively, students in the Experimental Group and their Clinical Instructors found the model to be an effective tool that aided students' understanding of patient assessment and treatment program planning. The need for an objective assessment tool of problem-solving models in physical therapy is discussed.

On-line sample cleanup in the liquid chromatographic analysis of pharmaceuticals for citrate content.

Matrix interferents are removed from pharmaceutical samples via an on-line, automated column switching process in which the difference in hydrophobicity between the analyte and the interferents allows these species to be isolated in different parts of the chromatographic system. In this case, the interferents are trapped on a cleanup column and are flushed to waste as the analyte undergoes additional separation on an analytical column. The utility of this approach is demonstrated by the quantitation of citrate in pharmaceutical samples by ion suppression, reversed-phase liquid chromatography. The performance of this system is statistically equivalent to that of a manual pretreatment method employing disposable, solid-phase extraction cartridges.

Determination of trace levels of oxytocin in pharmaceutical solutions by high-performance liquid chromatography.

A high-performance liquid chromatographic assay has been developed for quantitating oxytocin in common large volume parenteral intravenous solution matrices. Separation is accomplished by a reversed-phase mechanism using a C18 column. The analyte is detected fluorimetrically after post-column derivatization with fluorescamine. Reaction efficiency is controlled by the use of a reaction buffer which is added separately from the fluorescamine via a dual pump reactor system. Given the low analyte concentration [40 parts per billion (10(9)) or less], the samples are concentrated on-line through the use of a trapping column and switching valve. To improve productivity, pre-concentration and analysis of adjacent samples is timed to occur concurrently. Performance of the assay is characterized by a high degree of accuracy, precision and ruggedness; the system is capable of distinguishing between the analyte, matrix components, impurities and common degradates.

Application of fast LC to pharmaceutical analysis: dopamine.HCl.

A reversed-phase HPLC method for the determination of the dopamine hydrochloride content of pharmaceutical intravenous solutions is developed using short (3-cm) columns packed with 3-micron particles. Total analysis time for the stability indicating assay, which is capable of separating the analyte from all its common degradates and formulation matrix components, is 2 min. The assay is equivalent in performance (accuracy, precision, separation behavior, ruggedness) to a standard method recommended by the United States Pharmacopeia (USP). Common components of conventional HPLC systems are evaluated with respect to their utility for this application of fast LC.

Needle-guided breast biopsy for mammographic abnormalities in 561 patients.

Between 1977 and 1983, 561 consecutive patients underwent 595 surgical biopsies for suspicious mammographic lesions with negative clinical correlation. The procedure consisted of preoperative needle localization, with or without immediate radiologic examination of the biopsy specimen, depending on the presence or absence of microcalcifications in the mammographic lesion. Eighty-four carcinomas were found. Of these, 60 (71%) were infiltrating carcinoma and 24 (29%) were noninvasive carcinoma. The carcinoma yield was 24.2% in the patients with lesions involving foci of microcalcifications and 9% in those lesions without calcifications. Surgical treatment of infiltrating carcinoma consisted of 39 modified radical mastectomies, 10 (25.6%) of which were associated with positive nodes, 16 partial mastectomies with axillary dissection, 3 (18.7%) of which were associated with positive nodes, and 5 wedge resections. Treatment of noninvasive carcinoma consisted of 19 partial mastectomies with axillary dissection and 5 modified radical mastectomies. None of these were associated with positive nodes. Modified radical mastectomy was used with decreasing frequency. Of the 10 patients with infiltrating carcinoma and positive axillary nodes treated by modified radical mastectomy, 7 had one to three involved nodes and 3 had four or more; of those with positive nodes treated by partial mastectomy, 1 had one to three involved nodes and 2 had four or more. These results confirm the correlation between suspicious mammographic non-clinical lesions and breast carcinoma.

Percutaneous catheter drainage of abdominal abscesses.

This paper presents a study of 45 patients who had percutaneous catheter drainage of 52 intra-abdominal abscesses. In 37 patients this was the only treatment necessary; the other 8 needed further surgery to correct the cause of the abscess or treat concurrent surgical disease. The catheter complication rate was 3.8%.

Bulinus tropicus from Central Kenya acting as a host for Schistosoma bovis.

One hundred and twelve snails were collected from two habitats on the Mau Escarpment, Kenya and were provisionally identified as Bulinus tropicus from the characteristics of their shell and soft parts, chromosome number (n = 18), electrophoresis of egg protein on cellulose acetate strip and isoelectric focusing of AcP, GPI, HBDH, MDH and PGM digestive gland enzymes. Of the 55 specimens examined alive in London, 10 were infected with amphistome and schistosome larvae, 9 with amphistome larvae and the remainder were uninfected. The GPI and MDH separations of known infected snails showed two distinct areas of activity: host and parasite. Individual hamsters were exposed to schistosome cercariae emanating from each snail with a double infection (apart from one which died prematurely) and examination of the resulting adult worms showed that all were monomorphic for AcP with a band of enzyme activity at pH 6.45, characteristic of Schistosoma bovis. Examination of eggs found in two infections proved to be S. bovis in shape and size. Exposure of laboratory-bred snails of B. tropicus from the Mau Escarpment and other populations of B. tropicus proved negative. Thus, it is suggested that the presence of the amphistome infection may have a suppressive effect on the immune system of the snail, thereby allowing S. bovis to develop.

Bulinus tropicus, a natural intermediate host for Schistosoma margrebowiei in Lochinvar National Park, Zambia.

Snails, provisionally identified as Bulinus tropicus, on the basis of chromosome number, egg protein profile, AcP and HBDH enzymes of the digestive gland, and radular morphology, from Lochinvar National Park, Zambia have been demonstrated to transmit Schistosoma margrebowiei naturally. The identification of the unpaired male schistosomes was confirmed by PGM and AcP analyses. The observations confirm earlier epidemiological predictions, and add another species of mollusc to the two, B. forskalii and B. scalaris, known to be natural intermediate hosts of S. margrebowiei.
