Some aspects of adoption in selected samples of adult adoptees.

Various aspects of adoption--psychopathology, age at placement, agency versus independent placement, best age to tell child he is adopted, etc.--are studied in three groups of adoptees: biological parent schizophrenic or manic-depressive; adoptive parent schizoprenic or manic-depressive; biological parent with no psychiatric history. Findings raise questions about a number of common assumptions regarding adoptees.

Analytical methods for the determination of cannabinoids in biological media.

A pharmacokinetic study of the blood plasma levels in man of delta9-tetrahydrocannabinol, 11-hydroxy-delta9-tetrahydrocannabinol and cannabinol has been carried out by means of combined gas chromatographic-mass spectral analysis. In some cases comparison of the data was obtained on the same sample using thin layer chromatography of radiolabeled samples and electron capture gas-liquid chromatography. For the mass spectral studies appropriately deuterium labeled analogs of the previously named compounds were used both as internal standards and as a carrier for the relatively small amounts of nonlabeled drug present in plasma. Blood samples were obtainted at periodic intervals up to 24 hours from volunteers receiving 4-5 mg delta9-THC intravenously. After extraction and "clean-up" by Sephadex chromatography, the extracts were concentrated and subjected to glc-ms in the electron impact (ei) mode or alternatively with a chemical ionization (ci) source, in which case preliminary chromatography could be omitted. In all cases calibration curves were obtained from replicate analyses of spiked plasma containing the internal standard and various quantities of the cannabinoid under analysis. A typical biphasic elimination of the drug was observed with rapid elimination of delta9-THC from the blood over a period of 40 min followed by a much slower elimination up to 24 hours. The experimental data show that 11-hydroxy-delta9-THC is found in the plasma in quantities only about one-twentieth to one-twenty-fifth the values found for delta9-THC. Cannabinol was not found in significant quantities. Good agreement was obtained between the mass spectral analyses and the thin layer chromatography or electron capture gas-liquid chromatographic procedures.

The offspring of schizophrenics. Fetal and neonatal deaths.

This is the first report in a projected series on neurological development among the offspring of schizophernics. An increased incidence of fetal and neonatal deaths is noted among offspring of schizophrenics when compared to a closely matched control group, using data collected prospectively. In many cases, no apparent reason for death was determined; in some cases, major neurological malformations were found. It is necessary to consider three possible explantions for these findings--genotype of the child, adverse intrauterine environment, and medication toxicity.

Parent-child relationships and psychopathological disorder in the child.

This report uses naturally occurring adoptions to tease apart the effects of heredity and rearing on the development of psychopathological disorder in the child. The quality of relationship between the child and his adoptive parents was assessed in four groups of subjects, and the degree of illness in the child was correlated with the quality of parent-child relationship. Quality of rearing and hereditary input both affect the development of psychopathological disorder, but the amount of variance explained by rearing tends to be low.

Placental passage of antibodies to Dengue virus in persons living in a region of hyperendemic Dengue virus infection.

Maternal and umbilical cord levels of antibodies to dengue 2 and 3 viruses were compared in 54 matched pairs of specimens from subjects in the Dominican Republic. Dengue infection appeared to be hyperendemic; pregnant women experienced a 6% weekly infection rate. Titers of antibody to dengue 2 and 3 viruses were considerably higher in cord sera than in maternal sera (P smaller than 0.001 and P smaller 0.01, respectively), especially when maternal levels were low. When maternal and cord sera were compared at equal titers of dengue 2 virus, heterotypic antibodies to dengue 1 and St. Louis encephalitis viruses were higher in the cord. These findings could not be directly related to the higher levels of IgG found in cord sera or to the presence of elevated concentrations of IgM. It is concluded that antibody with increased cross-reactivity preferentially crosses the placenta. The presence of such cross-reactive antibody does not appear to sensitize infants to dengue hemorrhagic fever/shock syndrome.

Isolation and identification of a new metabolite of naltrexone in human blood and urine.

A new metabolite of naltrexone was isolated from urine and erythrocytes of patients taking 100 mg naltrexone daily. Its tentative structure of 2-hydroxy-3-methoxy naltrexol was derived by analysis of the purified compound using gas chromatography combined with mass spectrometry, high resolution mass spectrometry and infrared spectrometry. Long bioavailability of the new metabolite is indicated by its slower excretion rate into the urine than naltrexone and beta-naltrexol. The rather high concentration of the new metabolite in the erythrocytes and its solubility in non-polar solvents like benzene demonstrate the ability of this compound to penetrate membranes. These features of the new metabolite suggest that it may contribute to naltrexone's long time course of action.

Pyoderma Gangrenosum and rheumatoid arthritis.

Two patients had pyoderma gangrenosum and rheumatoid arthritis. Biopsy specimens of the cutaneous ulcers in the patients showed a necrotizing vasculitis. Complement (C3) and immunoglobulins were not detected in the skin lesions. There was no important impairment of cell-mediated immunity observed.
