Two regulatory elements for immunoglobulin kappa light chain gene expression.
By using internal deletions within a rearranged immunoglobulin kappa light chain gene, the presence of an intron regulatory sequence (enhancer) has been confirmed. Its presence is required for high-level transcription from a plasmid after transfection into myeloma cells. Transfection efficiency was monitored by the activity of a deleted H4 histone gene included in the plasmid. The intron element could be moved upstream of the gene in both orientations, fulfilling the definition of an enhancer. By using 5' deletions, a second regulatory element was located upstream of the "TATA" box, between positions -69 and -104. These two elements both are required for efficient kappa chain gene expression.

Cloning of a nitrogen fixation (nif) gene cluster of Azospirillum brasilense.
Homology was detected between the structural genes for the nitrogenase complex of K. pneumoniae (nifHDK genes) and the total DNA of several Azospirillum strains. Bacteriophage lambda gt 7-ara6 was used to construct a gene bank of A. brasilense strain 7000 DNA and a recombinant phage carrying a 6.7 kb Eco RI fragment, termed AbRI, was selected by hybridization with the K. pneumoniae nif probe. Using heteroduplex analysis the extent of the homology of the AbRI fragment and the K. pneumoniae nif genes was found to be approximately 5 kb. Proteins encoded by the AbRI fragment were examined after infection of E. coli minicells.
Regulated expression of an immunoglobulin kappa gene introduced into a mouse lymphoid cell line.
We have introduced a functionally rearranged murine kappa light chain immunoglobulin (Ig) gene into an Abelson murine leukemia virus-transformed lymphoid cell line. Plasmid pSV2gpt-kappa 41, containing the kappa light chain gene from the myeloma MOPC41 and the selectable marker gene gpt, was introduced into 81A-2 cells by the calcium phosphate coprecipitation technique. Cells expressing the gpt gene were selected by growth in medium containing mycophenolic acid. One transfected cell line, kappa-2, was shown to make kappa mRNA and polypeptide chains and to assemble the kappa chain product with gamma 2b heavy chains to form an apparently complete IgG2b. When bacterial lipopolysaccharide was added to the growth medium, levels of kappa mRNA and polypeptide increased, showing regulated expression of the introduced kappa gene.
Surgery in head and neck lymphomas.
Radiation therapy and/or chemotherapy are the treatment modalities of choice in lymphomas. Except for Burkitt's tumor, surgery is usually indicated only when percutaneous biopsy fails to establish the histologic diagnosis. We have described a patient with glomerulonephritis and anasarca secondary to an IgM-producing parapharyngeal lymphoma. After resection of all palpable tumor by an excisional biopsy, the monoclonal gammopathy and secondary nephrosis promptly cleared. With chemotherapy and mantle irradiation, the patient is in remission at three years. It is not now clear whether surgery will become useful as an adjunctive therapy for localized head and neck lymphomas. This will depend on whether local failure rates can be improved solely by using new combinations of irradiation and/or cytotoxic agents, and whether the morbidity of such therapy will outweigh the therapeutic gain.
Noninvasive measurement of systemic arterial pressure in dogs by automatic sphygmomanometry.
Mean, systolic, and diastolic systemic arterial blood pressures were recorded by indirect automatically inflating and deflating sphygmomanometry, using an oscillometric principle in 9 anesthetized dogs whose blood pressures were varied by infusions of sodium nitroprusside and phenylephrine. Blood pressures fluctuated between a diastolic pressure of 40 mm of Hg and a systolic pressure of 210 mm of Hg. A total of 147 blood pressures, measured by indirect sphygmomanometry, were compared with those obtained from a cardiac catheter within the artery of the contralateral limb. The device also indicated heart rate. Correlation coefficients between actual pressures from the cardiac catheter and those registered by the oscillometric device were greater than 0.9 for all pressures, and correlation coefficient for heart rate was 0.827. The lower correlation for heart rate was a consequence of (i) the slow sinus arrhythmia produced in response to phenylephrine and (ii) the device recognizing catacrotic waves as primary pulsations. In the anesthetized dog, this device is reliable, simple to use, and may be of value for monitoring dogs during surgery. Preliminary subjective evaluation indicated that the device is useful for measuring systemic arterial pressures in unanesthetized dogs as well, as long as motion of the limb around which the cuff is placed is minimal.
Enzymatic cleavage products of the alpha subunit of the receptor for immunoglobulin E.
The high-affinity membrane receptor for immunoglobulin E on mast cells and on a tumor analogue, rat basophilic leukemia cells, consists of two polypeptide chains: an alpha chain of Mr congruent to 50,000 and a beta chain of Mr congruent to 30,000. In this study we reacted alpha chains purified from tumor cells with proteolytic and glycolytic enzymes and compared the products by using differential labeling procedures. A variety of proteolytic enzymes cleave the chain into two similar-sized fragments, alpha 1 and alpha 2. The alpha 1 fragment behaves as if it were slightly larger on electrophoresis through polyacrylamide gels and is rich in carbohydrate as determined by incorporation of [14C]glucosamine. Less incorporation is observed into alpha 2 but it, like alpha 1, binds to concanavalin A. Labeling of the surface proteins on intact cells by lactoperoxidase-catalyzed iodination leads to preferential, perhaps exclusive, labeling of the alpha 2 fragment. The relative proportion of incorporated 3H-labeled amino acids and radioactive Bolton--Hunter reagent suggests that the polypeptide portions of alpha 1 and alpha 2 are similar in size. From these and other data we propose that the alpha chain may be U-shaped. Results from endoglycosidase digestions show that the receptor as isolated is heterogeneous because of variable glycosylation.
Parotid CT sialography.
Computed tomographic (CT) scanning of the parotid gland during the injection of water-soluble contrast medium allows precise mapping of the gland's parenchyma and of any filling defects or extrinsic masses. Thirty-six patients were examined in this way. The most valuable information that was gained was the differentiation of extrinsic and intrinsic tumors and an accurate appraisal of the location of the facial nerve with respect to intrinsic parotid lesions. In the majority of patients the diagnosis of benign or malignant tumor is suggested by the patterns of tumor growth. Inflammatory disease shows diminution in the amount of parotid parenchyma in relationship to interstitial tissues, but the appearance is not sufficiently characteristic for definitive diagnosis.

Colorado court involvement in chemical spill clean-up activities.
Judicial involvement was utilized to force the owners of a pesticide formulation plant to decontaminate property that had been covered with toxic pesticides having the potential to contaminate both surface and groundwater supplies in the East Denver metropolitan area. This case represented the first use of the Colorado state court system in dealing with a hazardous waste "spill." In this case, judicial intervention was unsatisfactory because of the delays involved. Other courses of action will be considered in future cases of a similar nature.
CT of cervical lymph node cancer.
This study seeks to establish whether CT can accurately detect cancer in cervical lymph nodes. Retrospective correlation of CT scans and surgical findings in 51 patients with laryngeal cancer undergoing radical neck dissection suggested that the node-bearing areas in the neck can be usefully evaluated with CT scans. An additional 10 patients studied prospectively with carefully correlated CT scans and radical neck dissections were used to evaluate CT specific criteria for cancer in individual nodes. In 13 patients CT correctly predicted surgically confirmed nonpalpable nodal metastases in six. It suggested false-positive nodes in two. These criteria are now being tested in an ongoing evaluation of the clinical value of CT in the management of head and neck tumors.
Pulmonary core biopsy by percutaneous needle aspiration.
Percutaneous aspiration needle biopsy for pulmonary lesions has been practised for many years but has not until recently gained popularity. Further proof of its simplicity, safety and accuracy for both central and peripheral lesions is given in this retrospective study of 181 biopsies performed in a community hospital. The diagnostic yield was higher in neoplastic (87.5%) than in nonneoplastic (68.4%) lesions and higher in malignant (91%) than in benign (58.3%) neoplasms. The larger the lesion the higher was the success rate--95.6% if the lesion was larger than 3 cm. The complication rate was low (14.9%) and there were no deaths. There was no difference in the success rate or the incidence of complications between biopsy done for central or peripheral lesions. The complication rate did not differ between large and small lesions, localized or diffuse. The sample could be used for cell block histologic sectioning, which gave the same diagnostic accuracy as cytologic examination. There were no false-positive cytologic diagnoses. During the procedure the operator (a radiologist in all cases) was able to predict the diagnosis in cases of hamartoma and neurofibroma.
