Activity of Schistosoma mansoni over a ten-hour period in vitro, and its modification by oxamniquine.

The activity of pairs of adult Schistosoma mansoni maintained in a continuous flow culture system was monitored for ten hours using the ultrasound method of Brown, Norman, Bell and Chavasse (1978). The results of 75 experiments were collated by computer to demonstrate the average effect of oxamniquine on worm activity in vitro at concentrations from 0.2 microgram/ml to saturation. The averaging technique indicates that oxamniquine concentrations down to 0.2 microgram/ml modify worm activity in vitro.

Schistosoma mansoni: ultrasonically detectable motor activity responses to 5-hydroxytryptamine in vitro.

The ultrasonic method compared favourably with other methods for monitoring schistosome motor activity in vitro, being able to detect and differentiate between the activity responses of Schistosoma mansoni to 5-hydroxytryptamine at concentrations down to 1 X (10(-6) M. This was taken to indicate suitable sensitivity for the application of the ultrasonic method to other drug studies.

Lack of immunoglobulin E response to longstanding strongyloidiasis.

Patients harbouring worm infections usually have very marked elevations of serum immunoglobulin E (IgE) concentration. However, in a group of twenty ex-Far East prisoners of war with strongyloidiasis only two (10%) had raised levels due to the worm, and these were only moderately elevated. This poor IgE response may be related to the very longstanding nature of the infection (at least 30 years). The patients may have a form of immunological incompetence, or may have become immunologically tolerant to the parasite.

Albumin permeability of the peritubular capillaries in rat renal cortex.

1. Two series of experiments were carried out to determine the permeability of the renal cortical peritubular capillaries to albumin under control conditions in the concentrating kidney and after an infusion of hypo-osmotic fluid amounting to 6% body weight. 2. In the first series of these experiments the turnover of the interstitial albumin pool of the renal cortex was studied. Specifically, the mean transit time of albumin molecules from arterial plasma to renal lymph was measured in nine rats of each group. Mean values of 39.0 +/- 2.8 and 30.6 +/- 2.2 (S.E. of mean) min were found for the control and infused groups, respectively. 3. In the second series of experiments the interstitial distribution volume of plasma albumin in renal cortex was determined in twenty-three control and twenty-two infused rats. Mean values of the extravascular distribution volumes were 1.66 +/- 0.21 and 1.37 +/- 0.18 (S.E. of mean) microliter/100 mg tissue, respectively. 4. The unidirectional clearance of albumin from the capillary to the interstitium in the control and infused groups, respectively, was calculated to be 7.1 +/- 1.0 and 7.5 +/- 0.9 (S.E. of mean) ml. sec-1 10(-4) in 100 g cortical tissue. 5. For the reabsorbing surface of the peritubular capillaries in the renal cortex, a lower bound was calculated for sigma, the reflexion coefficient of albumin. The reflexion coefficient was found to be higher than 0.998 under both experimental conditions.

Metrifonate in urinary schistosomiasis. A field trial in northern Nigeria.

A field trial of metrifonate in the form of Bilarcil tablets (Bayer) was carried out in 39 northern Nigerian schoolchildren heavily infected with Schistosoma haematobium. Three oral doses of 7.5 mg/kg were given at four-week intervals. The drug proved effective (mean reduction in egg counts eight weeks after completion of treatment 94.5%) and was excellently tolerated. Its use on a larger scale is suggested.
