Immunochemical demonstration of different antigens in single cells in paraffin-embedded histological sections.

A new modification of the double staining immunochemical technique is described, which is capable of demonstrating the presence of several antigens in single cells at the light microscopical level. This method was employed by the authors in order to detect the simultaneous occurrence of different types of cytoplasmic immunoglobulin chains in neoplastic plasma cell precursors in paraffin-embedded lymph-node specimens from patients with B-cell malignancies.

[Association of primary malignant neoplasms in the same patient].

The Authors have examined the problem concerning the presence of "Double malignancies" in the same patient, on the basis of a review of the literature and of an investigation of their own case material (nine patients: six with double solid tumours and three with a solid tumour and an associated haematologic neoplasia). The Authors discuss the time sequence of the two neoplastic processes and the immunological pattern (T and B lymphocytes) of all cases.

Cytobiological and clinical aspects of tissue mast cell leukaemia.

The authors describe the results of a series of cytochemical, autoradiographic, cytophotometric and immunological investigations carried out in a case of tissue mast cell leukaemia. Leukaemic mast cells showed certain distinctive cytochemical features, amongst which an intense periodic acid-Schiff (PAS) reaction, sensitive to amylase digestion, strong naphthol AS-D chloroacetate esterase (NASDCE), intense lactate dehydrogenase (LD) activity. Proliferative activity, determined autoradiographically with 3H-dT, was considerably low and was mainly confined to the larger cells. Also uridine and leucine incorporation were markedly reduced. Microdensitometry disclosed that the mast cell population was mainly arrested in the G1 phase. Because of previous attempts to destroy selectively neoplastic tissue mast cells with sheep antihuman IgE serum, a search for surface bound IgE was carried out, but gave a negative result. Possible therapeutic approaches are considered in the light of previous clinical experience and on the basis of the results of the kinetic and metabolic studies.

Autoradiographic studies on lymph node populations from non-Hodgkin B lymphomas.

The authors have investigated the cytokinetic behaviour of 23 cases of non-Hodgkin B cell lymphomas. In all cases diagnosis was established on the basis of histological, cytoimmunological and cytochemical data. The study, carried out with the aid of autoradiographic techniques, indicates that the various histological subclasses are characterized by a different metabolic and kinetic behaviour: the aggressiveness of the malignant lymphoma is closely related to the rate of growth of the lymphomatous cell populations, as assessed by the mitotic index and the labelling index with 3H-thymidine. The study of the synthesis and metabolism of RNA provides not only useful information on the energy requirements necessary for the neoplastic growth, but, when employed in association with immunohistochemical techniques for the detection of intracytoplasmic Ig synthesis, it defines more accurately the functional characteristics and the differentiation potentialities of the neoplastic populations in different cases.

[Etiopathogenetical, clinical and therapeutic considerations on meningo-encephalic localizations in acute lymphatic leukemias: personal contribution].

The aetiology, pathogenesis, clinical forms and therapeutic attempts carried out in connection with the central nervous system localizations in acute lymphoblastic leukaemia, on the basis of an extensive review of the literature and of personal experience are examined. The particular role which central nervous system leukaemia plays on the subsequent clinical course of the disease is stressed and the efforts made, both prophylactically and therapeutically, in order to decrease the incidence of this complication, which invariably has a deteriorating influence of the progress of the disease are described.

[Anatomo-clinical studies and therapeutic results in 2 cases of essential mixed cryoglobulinemia with renal insufficiency].

Two cases of essential mixed cryoglobulinaemia selected from a personal series on account of their clinical features (hyperviscosity syndrome in one case and frank renal involvement in the other) are presented. Extensive seroimmunological examinations were carried out, along with histopathological studies designed to reveal significant changes in organs and tissues, particularly the skin and kidneys. Reference is briefly made to the debate pathogenesis of the immunological phenomenon that probably lies at the root of the disease. A short account is also given of attempts at treatment with corticosteroids, immunosuppressors and plasmapheresis.

[Development of terminal leukemia in a patient with Hodgkin's disease].

A morphological, cytochemical, genetic and autoradiographic examination of the myeloblasts observed in the terminal phase of a patient with Hodgkin's disease is reported. Comparison is made with the literature data in an assessment of the possible aetiopathogenesis of this association, which has become increasingly frequent in recent years in coincidence with the more widespread employment of more radical radio- and polychemotherapeutical management.
