Use of cross-correlation function to detect patient motion during SPECT imaging.

We have developed a procedure to detect patient motion during a tomographic acquisition. The method uses frame-to-frame cross-correlation functions of the summed profiles in the vertical and horizontal directions of the planar images. The quantitative output derived from examination of the variation of the change in the pixel value, corresponding to the maximum of the cross-correlation function at each view, provides an effective and nonsubjective means of performing quality control on the presence and amount of movement during a single photon emission computed tomographic scan. In contrast to cine mode and sinogram display, easy to interpret hard copy can be generated through this procedure.

Evaluation of distance-weighted SPECT reconstruction for skeletal scintigraphy.

In 15 patients who had undergone skeletal scintigraphy for evaluation of low back pain, single-photon emission computed tomography (SPECT) reconstruction techniques of distance-weighted filtered backprojection (new method) and non-distance-weighted filtered backprojection (standard method) were compared. Mid-spine sagittal views differing only by reconstruction methods were evaluated by trained observers and by a quantitative profile technique. When asked to compare image quality, the observers demonstrated a clear preference for images reconstructed by the new method. Profile quantitation showed significantly greater contrast in more superficial structures with the new method.

Enhanced formation of benzo(a)pyrene:DNA adducts in monocytes of patients with a presumed predisposition to lung cancer.

Blood monocytes from 45 selected patients with lung cancer and 30 healthy controls were incubated with [G-3H]-benzo(a)pyrene for 30 h, and the formation of covalently bound DNA adducts was determined. The lung cancer patients were either relatively young (below 46 yr), nonsmokers, or had at least one first degree relative with lung cancer. Therefore, they might be considered cancer prone. The DNA adducts were significantly elevated in 22 patients with early age cancer (4.34 fmol/micrograms of DNA; P less than 0.04). In 12 familial cases, the slight elevation (2.77 fmol/micrograms of DNA) was not statistically significant in comparison to healthy controls. Benzo(a)pyrene:DNA adduct levels did not differ significantly between smokers and nonsmokers. Eight of 9 lung cancer patients with DNA adducts equal or above 4.5 fmol/micrograms of DNA but only 16 of 36 with adducts below this value had either oat cell or squamous cell cancer (P less than 0.05). The observed enhanced formation of covalent DNA adducts in blood monocytes exposed to a carcinogenic polycyclic hydrocarbon may be genetically determined and could play a role in the development of lung cancer at an early age.

Effect of paraquat intoxication and ambroxol treatment on hydrogen peroxide production and lipid peroxidation in selected organs of rat.

Paraquat (Pq) is a herbicide which is very toxic to all animals and to man. It generates free radicals and leads to acute or chronic lung injury and usually to death. So far, the role of lipid peroxidation of cell membranes in the mechanism of its toxicity has not been proved satisfactorily and therefore in the present study we examined the concentration of hydrogen peroxide (H2O2) and various lipid peroxidation products (LPP) such as conjugated dienes (CD), lipid hydroperoxides (LH), malonyldialdehyde (MDA) and Schiff bases in selected organs of the rat given a single intraperitoneal dose 35 mg kg-1 Pq. We also evaluated the influence of a mucolytic and probably antioxidant drug, ambroxol, on Pq-induced changes in the concentration of H2O2 and LPP. Paraquat increased the hepatic concentration of H2O2, CD, LH and MDA by approximately fourfold. Though the dose of Pq was nearly twice the LD50 dose, we did not notice any changes in the concentration of these substances in the critical organ, lung or heart and kidney. Ambroxol alleviated the increase of H2O2 in the liver but did not reduce the concentration of LPP. Moreover, the drug administered alone induced lipid peroxidation in the liver. Our results indicate that Pq dose not induce H2O2 production and lipid peroxidation in the lung but it increases the concentration of H2O2 and LPP in the liver. Ambroxol inhibits the Pq-induced increase in the concentration of H2O2 in the liver without protecting it against lipid peroxidation. Moreover, the drug alone may act as a pro-oxidant.

Monitoring of molecular and cytogenetic damage in lymphocytes from three persons with polycystic kidney disease.

BACKGROUND: Much interest has been generated in the studies that would help to understand whether there is a causal association between disease and various types of molecular or cytogenetic damage detected in human cells. MATERIALS AND METHODS: The aims of this study were to elicit the possible association between DNA and cytogenetic damage induced in lymphocytes of three members of a family with autosomal dominant polycystic kidney disease (ADPKD). The predictability to develop cancer or to sensitive response to environmental exposure of the young girl at the age of 19, her brother (9 years old) and a maternal aunt at the age of 41 were sought. Cytogenetic studies, analysis of DNA damage by single cell gel electrophoresis assay (SCGE known as a Comet assay), and analysis of p21ras protein level in blood plasma were carried out on their lymphocytes. RESULTS: The analysis for presence of chromosome aberrations in the first mitosis and sister chromatid exchanges in the second mitosis revealed elevated levels of cytogenetic biomarkers when compared to the mean values observed in the reference group in environmental biological monitoring studies. Results of sister chromatid exchanges (SCE) and percent of cells with elevated number of exchanges (high frequency cells) that were significantly higher in two probands had demonstrated susceptibility to or possibility of environmental exposure (pesticides, smoking). The results of this study show that the lymphocytes of two persons revealed increased sensitivity to 0.5 Gy dose of gamma radiation expressed in the increased, although statistically insignificant, damage detected on the molecular level after cell irradiation. CONCLUSIONS: The latter might be associated with a specific aberration present in the cells of these persons. But final conclusions can be arrived at when an application of FISH technique is completed.
