The choline acetyltransferase of human placenta.

1. Various methods for the extraction of choline acetyltransferase (acetyl-CoA-choline O-acetyltransferase, EC 2.3.1.6) from immature human placenta (18-28 weeks of gestation) are described. 2. The crude enzyme was found to be stable at -18 degrees and +4 degrees under a variety of conditions. 3. Purification methods, including ammonium sulphate fractionation, gel filtration on various grades of Sephadex and DEAE-Sephadex fractionation, have yielded a preparation of high specific activity.

Neutrophil influx and migration in rabbit airways in response to staphylococcal enterotoxin-A.

Single-cycle lavages performed on the lungs of rabbits 7 hours after they received intravenous staphylococcal enterotoxin-A (SEA) showed that most of the neutrophils were found in the second of 5 fractions of fluid removed from the lungs. To determine whether this finding reflected an accumulation of neutrophils in the airways versus the alveolar regions of the lungs, we counted the neutrophils in histologic sections (20 microns) and estimated the concentration of neutrophils in the epithelial lining fluid of the airways versus the alveoli. Comparisons of the morphometric measurements and the lavage data showed a strong correlation between the 2 methods (r = .90, P < .001), but the lavage technique underestimated the morphometric value by a factor of 7. The histologic examination confirmed that the concentration of neutrophils in the airways was 3-4 times higher than in the alveoli. There were no differences between the alveoli and the airways with respect to the concentration of endogenous proteins and the chemoattractant, interleukin (IL)-8. Lavages performed in additional rabbits 4 and 18 hours after injection of SEA showed that neutrophils most likely entered the alveolar region in response to SEA and migrated into the airways. The single-cycle technique provides a reliable measure of the relative concentration of neutrophils in the airways versus the alveoli.

The Early Reading Screening Instrument: a method for identifying kindergarteners at risk for learning to read.

This study was designed to provide speech-language pathologists and educators with a method for identifying children at risk for reading failure. The Early Reading Screening Instrument (ERSI) was given to 149 end-of-the-year kindergarten children. Half of the sample was tested 1 year later with standardized reading measures. Total ERSI scores from the kindergarten children strongly correlated with reading skills in first grade. Reading comprehension in first grade was the skill most strongly predicted by the subjects' total ERSI scores. The word recognition and invented spelling subtests of the ERSI were the best variables to be selected as predictors of first grade word analysis, word identification, and passage comprehension skills. A split-half reliability study of the ERSI showed that a shortened form could be used with the same degree of predictive validity. The ERSI or similar tools can be used to assess reading readiness in kindergarten children so that we can promptly begin an intervention to prevent or, at the very least, to diminish risk for reading failure. For screening reading readiness across different languages or cultures, the content of the four ERSI subtests can easily be modified.

Continuing education for advanced nurse practitioners: preparing nurse-midwives as clinical preceptors.

Nurses in advance practice roles are increasingly being sought as clinical preceptors for master's students. These nurses have variable experience as clinical teachers. A program was designed to prepare Certified Nurse-Midwives as clinical preceptors for students enrolled in a master's-level nurse-midwifery program. The program focused on techniques to foster clinical decision making, feedback, evaluation, and learning styles. Evaluations were positive and reflected that preceptors benefited from filmed modeling simulations and case study analyses.
