Operative recovery of vasal sperm from anejaculatory men: preliminary report.

Sperm showing good motility and function, as demonstrated by the in vitro sperm penetration assay using hamster ova, were collected from the vas deferens of 3 men with ejaculatory failure. The collection procedure may be repeated over multiple ovulatory cycles. The sperm may be used to attempt artificial insemination or in vitro fertilization.

Eosinophilic gastroenteritis presenting in an adolescent with isolated colonic involvement.

Eosinophilic gastroenteritis most commonly involves the stomach and proximal small intestine with eosinophilic inflammation of either the mucosa, submucosa or serosa. The patient reported here had isolated eosinophilic colitis. The initial presentation with iron deficiency anaemia owing to occult gastrointestinal blood loss emphasises the need to evaluate the entire gastrointestinal tract in patients with eosinophilic gastroenteritis.

Calcium-activated cation channel in rat thyroid follicular cells.

Using the patch-clamp single-channel current recording technique, a cation channel in the contraluminal membrane of rat thyroid follicular cells has been characterized. The channel has a unit conductance of about 35 pS and is equally permeable to sodium and potassium. The pattern of channel opening and closing is independent of the membrane potential. The channel is only operational when the ionized calcium concentration in the fluid which is in contact with the inside of the membrane is at least 1 microM. This conductance pathway can be classified as a calcium dependent non-selective cation channel and could explain stimulant-evoked depolarizations in the thyroid follicular cells.

Aboriginal pregnancies and births in South Australia, 1981-1982.

Information on 555 Aboriginal births which occurred during 1981 and 1982 was forwarded by midwives to the South Australian perinatal statistics unit. Corresponding information was also supplied for all other births in the state. This information showed that Aboriginal women appear to have a higher fertility rate than do other women, particularly in the teenage years. Aboriginal mothers are very young and have a high parity. They appear to receive little antenatal care and there is a greater tendency for their pregnancies to be complicated by medical conditions, such as anaemia, urinary tract infections, cardiac disorders and diabetes. Post-partum haemorrhages and retained placentas are relatively common, as are genital tract infections after delivery. Aboriginal babies are characterized by low birthweights, low Apgar scores, and prematurity. There is an indication that Aboriginal babies may have a high perinatal mortality rate in the country areas of South Australia. It is intended that this information be used as a baseline for evaluating trends in the health status of Aborigines.

Genes and gene products involved in the synthesis of F-pili.

Membrane fractions containing [35-S]methionine labeled proteins synthesized by Flac and Flac tra mutant strains or by lambda tra transducing phages expressed in such strains have been analyzed in order to investigate the pathway for synthesis of the F-pilin subunit and the gene products involved in synthesis of F-pili. Our data indicate that the synthesis of a mature F-pilin subunit requires the expression of at least 2 tra operon genes in addition to the structural gene for F-pilin, traA. In the absence of these activities, traA expression results primarily in the synthesis of a polypeptide, Ap14, with an apparent molecular weight of approximately 14,000. We assume this polypeptide corresponds to the direct product of the traA gene. In the presence of traQ activity, the major detectable product of traA is a polypeptide, Ap7(Q), which migrates with an apparent molecular weight of 7,000, suggesting that traQ product may process or assist in the processing of Ap14. Polypeptide Ap7(Q) is not, however, mature F-pilin, since it reacts poorly with anti-F-pilus-serum. Synthesis of a polypeptide which appears to be antigenically equivalent to F-pilin and which we assume requires a modification of the F-pilin N-terminus, is detected as synthesis of a polypeptide, Ap7*. This protein migrates slightly more slowly than Ap7(Q) on our polyacrylamide gels. Polypeptide Ap7*, can be efficiently precipitated with F-pilus antiserum, and can be detected in both inner membrane and outer membrane fractions under conditions where assembly of F-pili can occur. These data suggest that Ap7* is the mature F-pilin subunit and is assembled from an inner membrane pool. Synthesis of Ap7* appears to require traG activity, but may also be dependent upon additional tra activities.

Chromosomes of human sperm: variability among normal individuals.

The chromosomal constitution of 2468 human sperm cells has been investigated by fusion of human sperm with hamster eggs. The overall frequency of cells with structural aberrations was 7.7%, ranging from 1.9% to 15.8%, and varying significantly among individuals. The highest frequency occurred in sperm from the oldest donor (49 years), who also had had a vasectomy reversal three years prior to sampling. The overall aneuploidy frequency was 1.7%, ranging from 0.6% to 3.1%. In nine out of ten donors from whom blood samples were available the frequency of sperm cells with structural aberrations was higher than that for lymphocytes. Two previously reported donors (Brandriff et al. 1984) were resampled after an interval of 14 and 16 months respectively, and were each found to have similar frequencies of sperm chromosome abnormalities at both sampling times. A father-son pair included in the study had several chromosome breakpoints in common, although no more frequently than unrelated individuals.

Incidence of cancer in surgically treated thyroid nodules based on method of selection.

A retrospective review of thyroidectomies to rule out cancer at Columbia Presbyterian Medical Center indicated that the maximum cancer incidence achievable in surgical specimens was only 57% if all microfollicular adenomas and thyroid cancers were removed. The size distribution of follicular cancer and microfollicular adenoma was similar and it does not appear to be useful in deciding which microfollicular lesions need to be surgically excised to exclude carcinoma. The incidence of thyroid cancer in patients undergoing thyroidectomy was not influenced by the introduction of fine-needle biopsy (FNB) at Columbia Presbyterian Medical Center. FNB did appear to increase the incidence of cancer at an affiliated community hospital. Coarse-needle biopsy definitely improved the incidence of cancer in patients undergoing thyroidectomy and was superior to FNB and clinical selection alone. A combination of FNB and coarse-needle biopsy when possible yielded a 41% incidence of cancer and appears to be the most useful technique for patient selection.

Hyaluronate binding proteins also bind to fibronectin, laminin and collagen.

Small molecular weight proteins, isolated from the culture medium of embryonic chick heart fibroblasts and 3T3 cell lines by hyaluronate affinity chromatography, bind in order of apparent affinity, to hyaluronate, fibronectin, collagen and laminin. Such proteins isolated from the MSV-transformed 3T3 cell line bind in greater amounts to the nectins and hyaluronate than do similar proteins isolated from heart fibroblasts or 3T3 cells. These small hyaluronate binding proteins are immunologically distinct from other well characterized proteins such as laminin, fibronectin, bovine serum albumin and actin. Their relationship to other small, extracellular proteins and their possible role in structuring of extracellular matrix are discussed.

Abdominal positioning interneurons in crayfish: projections to and synaptic activation by higher CNS centers.

Intracellular recording, stimulation, and Lucifer dye injections were used to characterize abdominal positioning interneurons from the neuropile of the second through sixth abdominal ganglia of the crayfish, Procambarus clarkii. Motor outputs of these cells were recorded with extracellular electrodes placed on various flexion and extension roots along the nerve cord. In an effort to assess the functional relationships between the postural interneurons in the abdomen and those known to exist in the circumesophageal connectives ( CECs ), a stimulus pulse train was delivered to each of the CECs while monitoring the intracellular responses of the impaled interneurons. Abdominal positioning interneurons were grouped into four general categories based on their responses to CEC stimulation: 1) those that projected their axons directly through the CECs ; 2) those that were remotely activated to spiking; 3) those locally activated to produce EPSPs or IPSPs; and 4) those that were not affected by CEC stimulation. The majority of abdominal positioning interneurons encountered in this study evoked flexion (N = 82), with relatively fewer evoking extension (N = 29). A major difference appeared between the two classes. Whereas 39% of the flexion interneurons had axons coursing to the brain, only 7% of the axons of extension interneurons coursed rostrally beyond the thoracic level. Finally, the large majority of those flexion and extension interneurons that lacked processes in the CEC received synaptic inputs at various levels along the lower CNS from other CEC neurons. Thus, control of abdominal positioning involves neurons at all levels in the CNS - some sequentially organized, others forming "through" pathways, but all with multiple input sites.

Metastasizing basal cell carcinoma developing in a gunshot wound in a black man.

Both basal cell carcinomas that arise in blacks and those that metastasize are rare, and development of this tumor is an uncommon, late complication of trauma. We report a case of a 70-year-old black man with a basal cell carcinoma arising in a gunshot wound in a shoulder and metastasizing to axillary lymph nodes. Histologically, the tumor was of the metatypical type.

Synthesis of F-pilin polypeptide in the absence of F traJ product.

The products of a lambda transducing phage (ED lambda 101) which carries a segment of the F tra operon expressing F traA , traL , and traE activity from the lambda leftward promoter were examined using a uv-irradiated host system. After infection of an F- host, products of traE (19,500 Da) and traA (14,000 Da) were detectable among the lambda early proteins synthesized. Infection of an Flac host altered the pattern of polypeptides synthesized by the phage in that the 14,000-Da traA product became barely detectable and was replaced by a polypeptide which migrated at 7000 Da. A derivative of ED lambda 101 carrying the traA1 amber mutation was unable to synthesize either the 14,000-Da polypeptide in F- cells or the 7000-Da polypeptide in Flac cells. The 7000-Da polypeptide derived from ED lambda 101 was synthesized in the absence of traJ product in F- cells coinfected with a second transducing phage which carried a tra operon segment including traQ . It was also a product of ED lambda 134 which expresses genes traA through traH . The 7000-Da polypeptide, like F-pilin, associated primarily with the inner membrane, and could be immunoprecipitated with antiserum prepared against purified F-pili. Analysis of membranes from F- cells infected with ED lambda 101 indicated that the 14,000-Da traA product synthesized under these conditions accumulated in the inner membrane. These results show that both the 14,000-Da traA product might be processed to F-pilin in a traQ -dependent reaction which occurs in or on the inner membrane of the Escherichia coli host. However, the possibility that traQ encodes a regulatory product which affects expression of the traA sequence has not been excluded.
