The participation of nitric oxide in cell free- and its restriction of macrophage-mediated oxidation of low-density lipoprotein.

The potential role of nitric oxide radical (NO .) in macrophage-mediated oxidation and conversion of human low density lipoprotein (LDL) to a high-uptake form was examined by exposing LDL to aerobic solutions of either NO . or 3-morpholino-sydnonimine-hydrochloride (SIN-1, a compound that spontaneously forms NO . and superoxide anion radical) or to mouse peritoneal macrophages in the presence and absence of modulators of cellular NO . synthesis. Incubation with NO . alone caused oxidation of LDL's ubiquinol-10 and accumulation of small amounts of lipid hydroperoxides, but failed to form any high-uptake ligand for endocytosis by macrophages and did not alter the LDL particle charge or the integrity of apoB. Exposure of LDL to SIN-1 resulted in complete consumption of all antioxidants and substantial formation of lipid hydroperoxides, but again had little effect on the lipoprotein particle charge or generation of high-uptake form. Preincubation of macrophages with interferon-gamma increased the cells ability to generate reactive nitrogen metabolites. The extent of cell-mediated oxidation of LDL and the generation of high-uptake LDL was substantial in resident cells in which NO . synthesis was barely detectable, depressed in cells active in NO . synthesis and restored when NO . synthesis was suppressed by the arginine analogue, NMMA. These results suggest that, while together with superoxide anion radical, NO . can oxidize LDL, its synthesis is not required for macrophage-mediated oxidation of LDL in vitro; rather it exerts a protective role in preventing oxidative LDL modification by macrophages.

Dietary supplementation with coenzyme Q10 results in increased levels of ubiquinol-10 within circulating lipoproteins and increased resistance of human low-density lipoprotein to the initiation of lipid peroxidation.

Ubiquinol-10 (CoQH2, the reduced form of coenzyme Q10) is a potent antioxidant present in human low-density lipoprotein (LDL). Supplementation of humans with ubiquinone-10 (CoQ, the oxidized coenzyme) increased the concentrations of CoQH2 in plasma and in all of its lipoproteins. Intake of a single oral dose of 100 or 200 mg CoQ increased the total plasma coenzyme content by 80 or 150%, respectively, within 6 h. Long-term supplementation (three times 100 mg CoQ/day) resulted in 4-fold enrichment of CoQH2 in plasma and LDL with the latter containing 2.8 CoQH2 molecules per LDL particle (on day 11). Approx. 80% of the coenzyme was present as CoQH2 and the CoQH2/CoQ ratio was unaffected by supplementation, indicating that the redox state of coenzyme Q10 is tightly controlled in the blood. Oxidation of LDL containing various [CoQH2] by a mild, steady flux of aqueous peroxyl radicals resulted immediately in very slow formation of lipid hydroperoxides. However, in each case the rate of lipid oxidation increased markedly with the disappearance of 80-90% CoQH2. Moreover, the cumulative radical dose required to reach this 'break point' in lipid oxidation was proportional to the amount of CoQH2 incorporated in vivo into the LDL. Thus, oral supplementation with CoQ increases CoQH2 in the plasma and all lipoproteins thereby increasing the resistance of LDL to radical oxidation.

A calcium model with random absorption: a stochastic approach.

Absorption of calcium, or any mineral, by the body is subject to the random fluctuations typical of diffusion through membranes. In this paper we consider the absorption of calcium from the gut as a white noise process added to the deterministic model of Sen & Mohr (1990, J. theor. Biol. 142, 179-188). The first two moments for the amount of calcium in the extracellular fluid (ECF) have been derived using the Ito Calculus. A confidence interval for the total amount of calcium in the ECF is constructed. The equations for the first two moments of the fraction of dose calcium in the ECF are also given. Suggestions are made for the collection of experimental data in a form which should be helpful in investigating the magnitude of the stochastic effect.

Double-blind, cross-over study comparing prostaglandin E1 and papaverine in patients with vasculogenic impotence.

Intracorporeal injection of vasoactive agents is the treatment of choice for many patients with organic impotence. However, some patients with vasculogenic impotence respond poorly to injections of papaverine or papaverine/phentolamine. This study was conducted to determine if patients with vasculogenic impotence who failed to respond to papaverine might respond to prostaglandin E1 and thus be salvaged from possible prosthetic surgery. A total of 54 patients with vasculogenic impotence were administered intracorporeal prostaglandin E1 (20 micrograms) and intracorporeal papaverine (60 mg) randomized in a double-blind fashion and crossed over one week later. Forty-six percent of patients receiving prostaglandin E1 produced a satisfactory erection compared with 14 percent of patients with a similar response to papaverine. The difference between these results was highly significantly by the McNemar test. The number of side effects were similar for both drugs. Prostaglandin E1 might be considered the intracorporeal agent of choice for patients with significant vasculogenic impotence.

Predictors of differential response to cognitive, experiential, and self-directed psychotherapeutic procedures.

Group cognitive therapy (CT), focused expressive psychotherapy (FEP; a form of group experiential psychotherapy), and supportive, self-directed therapy (S/SD) were compared among 63 patients with major depressive disorder (MDD). Variation among patients' coping styles (externalization) and defensiveness (resistance potential) was used in a prospective test of hypothesized differential treatment-patient interactions. Results suggest that patient characteristics can be used differentially to assign psychotherapy types. Externalizing depressed patients improved more than nonexternalizing depressed patients in CT, whereas nonexternalizing (internalizing) patients improved most in S/SD. Conversely, high defensive (resistant) patients improved more in S/SD than in either FEP or CT, whereas low defensive patients improved more in CT than in S/SD.

Problem solving in elderly sophisticated and naive monkeys.

Elderly (greater than 23 years) rhesus monkeys, some of whom had previous learning test experience, were tested on a standardized battery of discrimination and concept-formation tasks, and their performance was compared with that of test-naive middle-aged (15 years) and juvenile (2 years) monkeys. Naive elderly monkeys performed more poorly on virtually all tasks compared to their younger counterparts. In contrast, test-experienced elderly monkeys outperformed both middle-aged and juvenile monkeys on those problem types with which they had limited previous experience, despite the fact that 8-10 years had passed since their prior training. However, test-experienced elderly monkeys did not display superior performance on new tests that approximated visual acuity or assessed delayed discrimination capabilities.

A kinetic model for calcium distribution.

Models for the distribution of minerals in the body are of interest as they allow researchers to trace the effect of a dose on mineral levels in plasma, storage and other compartments. Limited models are available in the literature for tracing the distribution of a calcium dose through a short time period. We propose a more general kinetic model which includes both limited absorption through the gut and loss of calcium via excretion. This new method has the advantages of giving reasonable results over moderate time periods, and allowing the extrapolation of calcium levels in extracellular fluid and storage. We fit the model to published data in order to obtain typical parameter values. These values are then used to analyze the implications of the model regarding the effect of calcium dose on calcium levels in various compartments.

[Laboratory studies of vibration transmission in motor vehicle seats].

Operator seats (Models 050 and 500, Möve) were examined with vertical sinusoidal whole-body vibration (WBV) in the frequency range from 1 to 40 Hz (1.5 and 3.0 ms-2rms) and with simulated stochastic vibration of combines for the harvest of sugarcane. Six male subjects volunteered for the experiments. The WBV transmitted from the seat mounting base to the seat cushion and head were compared with data of a hard experimental seat. Interindividual differences cannot be explained by different body masses only. The iolation and main resonance of the two operator seats differed. Additional resonant peaks occurred above 6 Hz. They can cause unfavourable conditions at certain applications. The operator seats tested are probably suited for the reduction of WBV exposure on combines for the harvest of sugarcane.

[Laboratory studies on the subjective evaluation of motor vehicle seats].

Operator seats (Models 050 and 500, Möve) were examined with vertical sinusoidal whole-body vibration (WBV) in the frequency range from 1 to 40 Hz (1.5 and 3.0 ms-2 rms) and with simulated stochastic vibration of combines for the harvest of sugarcane. The subjective assessments of operator seats by cross-modality matching, paired comparisons, and questionnaires were compared with data of a hard experimental seat. Transmitted WBV and subjective evaluation correlated partially only. Suggestions are derived for the improvement of the seats tested.

[Behaviour of serum concentrations of glucose, immunoreactive insulin and potassium ions in newborns after long term or crash treatments with Partusisten or Partusisten in combination with Isoptin (author's transl)].

The investigation was done on 50 infants born of metabolic healthy mothers. A group of 10 women received short term treatment with Partusisten and Isoptin. Another 10 were treated over the same period with Partusisten only. A further group of 15 received long term treatment (more than 7 days) with Partusisten and Isoptin and a final group of 15 mothers received placebos. Serum concentrations of glucose, immunoreactive insulin and potassium, together with acid-base-status and other important blood chemistry were determined in mothers and newborns directly postpartum. Blood sugar, immunoreactive insulin and potassium were further determined in the newborns 30 minutes, 60 minutes, 2 hours and 6 hours after birth. The results showed that a temporary relative hypoglycaemia occured in the newborns in short term as well as long term treatment with Partusisten or Partusisten in combination with Isoptin. Temporary derangements in insulin-glucose equilibrium were also seen in newborns from mothers receiving long term treatment. It was also demonstrated that approximately 1 hour post-partum, blood sugar and immunoreactive insulin of the newborn attained values compatible with normal carbohydrate metabolism. From these results it is indicated that infants born of mothers, who received tocolytic treatments with Partusisten or Partusisten in combination with Isoptin, should be given sodiumbicarbonate and glucose directly after birth.

[Growth hormone in the newborn infant during the first 6 days of life].

Human Growth Hormone (HGH) of the newborn during its first days of life is approximately 61 ng/ml, then it falls between the third and fourth day. This fall in serum concentration principally occurs when the infant stops losing weight and starts regaining it. Experiences show that HGH of newborn does not stem from the mother.
