Leukocyte kinetics, cell cycle and leukemic proliferation.

A review is presented of the available data in the medical literature and of the author's experience in the study of leukocyte kinetics, starting with the originary stem cell. To facilitate understanding of the process of leukocyte proliferation in leukemias, theoretical and methodological details are given on the techniques for determination of the cell cycle and its perturbations during evolution of the various types of leukemias. Some data on animal leukemias are also discussed. A critical analysis is made of the action of immunosuppressive cytostatic drugs on the leukemic cell proliferation.

Detection of lymphocytic Fc receptors with 125I-labelled protein A.

An attempt is made to improve the methodology for the detection of lymphocytic Fc receptors, using 125I-labelled protein A from Staphylococcus Aureus (125ISpA) and different immunoglobulins G such as human monomer IgG, Fc antiserum and swine Fc+F'c antiserum. The working conditions and sources of error that might affect the results were established. This methodology was applied to the study of leukemia, chronic hepatitis, allergic diseases and normal cases. In the chronic lymphoid leukemia cases the determinations were repeated at different clinico-biological stages. The values obtained in these cases have always been higher than normal. This method may be used for rapid determination of the presence of Fc receptors; its application in the clinic would furnish useful data. It still remains to be established whether determination of these lymphocytic receptors might contribute data that can be corroborated by the response to the immunodepressor treatment applied in some cases.

"Killer"-type phenomena in the immune pathology of some chronic internal diseases. (Scanning electron microscopy studies).

Aspects of cytotoxicity followed by cytolysis have been described by many authors in neoplasias. Using scanning electron microscopy, the authors have investigated the cellular population from 179 patients with various chronic internal diseases such as: chronic hepatitis, diabetes mellitus, systemic lupus erythematosus, scleroderma, chronic lymphocytic leukemia. In some of the forms of disease with severe clinical and biological phenomena, they have detected the presence of "killer" type phenomena, having as target cells either the monocytomacrophage or the T or B lymphocytes. Iconographic aspects are demonstrating the presence of this phenomenon, observed particularly in the peripheral blood. Detection of such phenomena is assumed to provide information on prognosis of the chronic disease investigated and on the effects of therapy at cellular level.

The area of lymphocyte nucleoli, a new method estimating the blastic transformation of stimulated lymphocytes in in vitro cultures.

To estimate the degree of lymphocyte reactivity in patients with internal diseases with immune components, the authors used a new method of investigation, the area of lymphocyte nucleoli (ALN), recently proposed by them. ALN was determined in blood cultures stimulated with phytohemagglutinin (PHA) and sacrificed after three days. The blood was obtained from 120 patients with chronic internal diseases with immune components such as: bronchial asthma, chronic hepatitis, rheumatoid arthritis, systemic lupus erythematosus or chronic lymphocytic leukemia (CLL). The data obtained showed clearly that, compared with the classical test of lymphocyte blastic transformation, ALN--which presented increases even by more than 100% over normal in rheumatoid arthritis and in systemic lupus erythematosus--can supply additional information regarding the degree of reactivity of the cultivated lymphocytes. In CLL, as it was to be expected, ALN values were below the normal.

Cyto-nucleo-nucleolar variables estimating the lymphocyte reactivity in chronic internal diseases with immune component.

To estimate the degree of lymphocyte reactivity, 11 cyto-nucleo-nucleolar variables were determined in the lymphocyte population from 30 healthy blood donors and 103 patients with chronic internal diseases with immune component such as: rheumatoid arthritis, chronic hepatitis, Hodgkin's disease or chronic lymphocytic leukemia. The data obtained showed that all the cyto-nucleo-nucleolar variables investigated were different in chronic hepatitis from all the other groups, both as regards the relative and absolute areas (low for the cell and nucleus and high for the nucleoli), and as regards the ratios of cell/nucleus and nucleus/nucleolus areas.

Cytochemical and cytoenzymatic investigations in the management of leukemias.

The value of certain cytochemical and cytoenzymatic investigations in the management of leukemias is discussed in different types of acute or chronic leukemias. Among the data resulting from cytochemical methods those related to cellular biochemical components such as DNA, RNA, glycogen and lipids are particularly noteworthy. The results of cytoenzymatic investigations have stressed the necessity of knowing the activity of certain enzymes such as peroxidases, alkaline and acid phosphatases, beta-glucuronidase, succinate dehydrogenase, lactate dehydrogenase and glucose-6-phosphate dehydrogenase a.o. The prospective value of enzymes such as dehydrofolate reductase, DNA and RNA polymerases, DNA and RNA-ases a.o. in the management of leukemias is also mentioned.

Mixed tumor: mucinous papillary cystadenocarcinoma associated with arrhenoblastoma.

A case of a 41-year old woman with an enlargement of the inferior part of the abdomen is presented. An echographic examination established the diagnosis of a right ovarian cyst. During the operation, a 14-cm (5 1/2") tumor, presenting a large ruptured cyst and solid mass, was found. From the cystic activity, a viscous fluid was oozing. On the left ovary, a 4 cm (1 1/2") cyst was also found. Within this case report, we underline the unusual association of a mucinous papillary cystadenocarcinoma with an arrhenoblastoma in the right ovary together with the presence of an cystadenoma in the left ovary.

Behaviour of leukocyte acid phosphatase in various chronic diseases.

Leukocyte acid phosphatases were investigated in 146 patients with different chronic diseases. The method of investigation used was that of Kaplow and Burstone slightly modified by the authors in what regards the pH of the incubation medium. Normal or slightly increased scores were observed in the granulocytic series of patients with chronic myeloid leukemia. In patients with rheumatoid arthritis, chronic hepatitis, lupus erythematosus disseminatus and chronic lymphocytic leukemia a moderate enzymatic activity was generally observed in the lymphocyte and more marked in severe forms of disease. A marked increase of the enzyme activity was observed in patients with myeloma. The possibility of a correlation between the intensity of enzyme activity and immunoglobulin formation is discussed.
