Characterization of human airway epithelial cell leukotriene A4 hydrolase.

We have previously shown that human airway epithelial cells contain leukotriene A4 (LTA4) hydrolase activity. To characterize this activity further, airway epithelial cells, cultured to confluence, were disrupted by sonication and were fractionated at 15,000 and 100,000 x g. Enzymatic activity was assessed by incubating fractions with 15 microM LTA4 at 37 degrees C for 15 min. LTA4 hydrolase activity was present in the 15,000 x g and the 100,000 x g supernatants and was inactivated by heating at 56 degrees C or by pronase, as is the case for neutrophil LTA4 hydrolase. However, the epithelial cell enzyme had a slower time course for product generation and demonstrated a different dose-response relationship to substrate when compared with the neutrophil. Kinetic analysis revealed nonlinear plots for epithelial data, most consistent with an enzyme that has multiple active sites. Immunoblotting, performed with anti-neutrophil LTA4 hydrolase antibody, recognized two bands in epithelial cell 15,000 x g supernatant (M(r) of 69,000 and 110,000-115,000). When resolved by gel filtration chromatography, only the M(r) 69,000 protein had enzymatic activity. Anion exchange chromatography of epithelial cell samples revealed that LTA4 hydrolase and aminopeptidase activity did not co-elute, whereas one of three peaks of aminopeptidase activity did co-elute in chromatograms of neutrophil samples. Immunoblots of proteolytic digests of partially purified M(r) 69,000 protein from epithelial cells and neutrophils revealed different immunoreactive bands. The digest of the M(r) 110,000-115,000 protein revealed no immunoreactive bands. Repeat kinetic analysis on 179-fold purified epithelial LTA4 hydrolase again revealed that it lacked significant aminopeptidase activity and retained its unique kinetic properties.(ABSTRACT TRUNCATED AT 250 WORDS)

Methylamine-treated low density lipoproteins elicit different responses in HepG2 cells and macrophages.

Recent results from this laboratory have demonstrated the existence of labile thiolester bonds in apolipoprotein B (ApoB). Thiolester bonds can be cleaved with nucleophiles such as methylamine, resulting in conformational change. The purpose of this study was to explore whether the cellular interactions would be altered after methylamine treatment of low density lipoproteins (LDL). Human hepatoma cells, HepG2, and human monocyte derived macrophages were used for these studies. Fresh LDL were incubated with methylamine under mild alkaline conditions under N2 and with preservatives for 24 h. The methylamine-treated LDL showed particle size and net charge identical to fresh native LDL. In addition, no oxidative modification of LDL occurred under the experimental conditions. The methylamine-treated LDL were indistinguishable from native LDL in HepG2 cells as judged by binding, degradation, cholesterol accumulation and de novo sterol synthesis. However, methylamine-treated LDL caused an increased accumulation of cholesteryl esters in macrophages which was comparable to the accumulation caused by acetylated LDL. Dual color digital imaging fluorescence microscopy revealed no competition between acetylated and methylamine-treated LDL, suggesting that the excessive uptake of methylamine-treated LDL was not mediated by the 'scavenger' receptor. The increased accumulation of cholesteryl ester in macrophages also did not appear to stem from the classical LDL receptor. These results suggest that a new receptor binding domain is exposed due to the conformational change upon treatment of LDL with methylamine.

[Lysosomal enzyme activity of cultured fetal cells in Chinese and its clinical application].

Lysosomal storage diseases (LSD) are caused by deficient activity of specific lysosomal enzymes. Early diagnosis and selective termination is still the trend of therapy. The purpose of this study was to establish an assay system and investigate the reference range of lysosomal enzyme activity of cultured fetal cells in the Chinese population. Seventy amniotic fluid and 9 chorionic villi samples were collected and cultured in this study. Enzyme activity assay was done by synthesized 4-Mu-binded substrates. The activity was expressed as nmol/mg protein/hour. In cultured amniotic cells, the results showed 14-138 of alpha-glucosidase, 8-133 of alpha-galactosidase, 32-470 of alpha-mannosidase, 101-1121 of alpha-fucosidase, 106-1321 of beta-galactosidase, 15-268 of beta-glucosidase, 11-279 of beta-glucuronidase, 101-1193 of Hexosaminidase A, and 886-6204 of N-acetyl-alpha-glucosaminidase. In cultured chorionic villi samples, it showed 22-335 of alpha-glucosidase, 31-230 of alpha-galactosidase, 47-250 of alpha-mannosidase, 35-218 of alpha-fucosidase, 49-934 of beta-galactosidase, 34-329, of beta-glucosidase, 57-379 of beta-glucuronidase, and 328-3412 of Hexosaminidase A. The enzyme activity was not correlated with the gestation age when sample was obtained. Furthermore, there was no statistical significance among the range of amniotic cells, chorionic villi samples, skin fibroblasts and peripheral leukocytes for each enzyme studied. It is suggested that the synthesis of lysosomal enzymes has been mature since the early fetal state, and the samples obtained as early as 8 weeks of gestation age can be used for early diagnosis of lysosomal storage diseases.

Lipid peroxide levels in type II hyperlipoproteinemic subjects.

The purpose of this study was to evaluate, under the most rigorous precautionary measures against in vitro oxidation, whether the baseline lipid peroxide levels in hypercholesterolemic subjects were higher than those in normolipidemic subjects. Two methods were employed: thiobarbituric acid (TBA), and hemoglobin-methylene blue (HbMB). Blood was collected into EDTA tube and centrifuged at 4 degrees C for 30 min to collect plasma, then protected from in vitro oxidation with preservatives and N2. Serum was from blood samples allowed to clot at 20 degrees C for 1 h, then protected from oxidation. Determination of lipid peroxide was carried out within 2 h of blood collection. Results from 35 hypercholesterolemic and 34 control subjects showed that lipid peroxide levels obtained from both methods were significantly higher in serum than in plasma for both groups, suggesting a greater rate of lipid peroxidation occurred in serum during clot formation. However, no significant difference in lipid peroxide levels was found between patients and controls in either serum or plasma by either assay method. No correlation existed between lipid peroxide values and plasma cholesterol or LDL-cholesterol levels. These results suggest that the mechanism for a higher tendency towards atherosclerosis in hypercholesterolemic subjects is not related to baseline levels of plasma lipid peroxide.

Prevalence of atretic corpora lutea in ovaries of bovine fetuses.

Both ovaries from 88 bovine fetuses in the fifth month or later of gestation were studied histologically to determine the prevalence, origin, and time of appearance of atretic corpora lutea (ACL). Ovaries from 36 (41%) fetuses had ACL; fetuses < 6 months of gestation did not have ACL. Six fetuses had more than 25 ACL, but there was no apparent relationship between fetal age and number of ACL. Formation of ACL involved disintegration of the stratum granulosum of secondary follicles, concomitant with proliferation and invasion by vascularized elements of the theca. Fully developed ACL consisted of a large primary oocyte surrounded by a prominent zona pellucida and encased in a well-vascularized, largely thecal, fibrocellular wall. They measured approximately 0.5 to 1.0 mm in diameter. Empty, collapsed zona pellucidas were seen in many of the degenerating ACL.

Receptor-dependent and -independent catabolism of low-density lipoprotein in a kindred with familial hypobetalipoproteinemia.

Three affected members of a kindred with asymptomatic hypobetalipoproteinemia (HBL) were injected intravenously with 125I-labeled native low-density lipoproteins (LDL) and 131I-labeled cyclohexanedione (CHD)-treated LDL. Plasma and urine radioactivity data were collected for 15 days at regular intervals. A compartmental model using the SAAM program was built to fit simultaneously 125I and 131I plasma radioactivity decay and urine excretion data. This model allows precise calculation of the kinetic parameters of both receptor-independent (NR) and receptor-dependent (R) pathways. Compared with normal subjects, HBL patients show a 90% increased fractional catabolic rate (FCR) of LDL by both routes, more marked for the R pathway (215% increase), and an approximately 50% reduced production rate (PR). Structural analysis did not show significant abnormalities of apolipoprotein (apo) B in HBL patients compared with normal. These data suggest that the very reduced, LDL-apo B plasma levels result from a combination of two processes: (1) an increased activity of all catabolic routes, and (2) a reduced "synthesis" rate. The latter may result from a decreased conversion of very-low-density lipoprotein (VLDL) to LDL secondary to an increased direct removal of large VLDL, suggested by apo C-II and C-III turnover studies previously reported.

[How painful is intensive insulin therapy?].

Injection-pain was assessed in 39 type-1 diabetic patients in relation to an insulin injection (with an insulin-pen) into the upper arm, the abdomen, and the thigh and in another 10 patients, pain associated with finger-pricking for blood-glucose self-monitoring was assessed. Pain perception was recorded on a visual analogue scale. According to the results, injections were relatively painless with an average (median) pain-score of less than 10% of maximum pain. Injection pains varied in relation to the injection sites (upper arm greater than thigh greater than abdomen). Least painful (n = 44) versus most painful (n = 39) injections were distributed as follows: upper arm 20% vs. 46%, abdomen 43% vs. 23%, thigh 36% vs. 31% (chi 2 7.26, df2, p less than 0.05). Finger-pricking was no more painful than injecting insulin. It is concluded that insulin injecting (e.g. with insulin pens) and blood glucose self-monitoring in general is not very painful. However, if this positive conclusion also applies to long-term performance of intensive insulin therapy remains to be proven.

Rat apolipoprotein B differs in solubility properties from human apolipoprotein B.

Apolipoprotein B (ApoB) in general is known as an insoluble protein in aqueous buffers without the initial aid of denaturing agents. However, following the total delipidization of rat plasma low-density lipoproteins, a considerable portion (28.2 +/- 3.0%) of rat ApoB was found directly soluble in an aqueous buffer, N-ethylmorpholine acetate (pH 7.3) as demonstrated by SDS-polyacrylamide gel electrophoresis, immunoblotting and electron microscopic analysis. On the other hand, this was not observed for human ApoB. This solubility difference may suggest some structural differences that exist between rat ApoB and human ApoB.

Physicochemical studies of two major subpopulations of low density lipoproteins by differential scanning calorimetry and n.m.r. spectroscopy.

Two subpopulations, layer 2 (density 1.025-1.029 g/ml) and layer 3 (density 1.032-1.043 g/ml) of low density lipoproteins (LDL) were isolated from fresh human plasma of normal lipidaemic subjects by density gradient ultracentrifugation. Chemical analyses demonstrated the ratios of triglyceride/cholesterol ester decreased with increasing densities of subfractions. These subfractions together with triglyceride-rich lipoproteins (layer 1, density less than 1.019 g/ml) were subjected to physicochemical studies by differential scanning calorimetry (d.s.c.) and nuclear magnetic resonance (n.m.r.) spectroscopy. The average transition temperature (Tt) of layer 2 was 34.20 +/- 0.83 degrees C and that of layer 3 was 37.25 +/- 0.35 degrees C. In addition, many of the layer 3, but not layer 2 and layer 1, samples showed structural alteration and gave rise to an average Tt of 39.18 +/- 1.24 degrees C. The structural alteration could be detected with polarizing light microscopy showing birefringent spherulites at body temperature. The peak Tt values obtained by d.s.c. were in good agreement with those by n.m.r. spectroscopy. These results demonstrate the physicochemical heterogeneity within the LDL density region and suggest that layer 3 subpopulation is much more labile than the others.

Intramolecular thiolester linkages in apolipoprotein B.

Intramolecular thiolester bonds in apolipoprotein B (ApoB) were studied using [14C]methylamine (MA) to cleave the thiolester and [3H]- or [14C]iodoacetate (IA) to titrate the newly generated sulfhydryls. Covalent incorporation of [14C]MA and [3H]carboxylmethyl group into the previously carboxymethylated LDL or the reduced and carboxymethylated ApoB was observed and both radioactivities coincided with ApoB-100 band on SDS-polyacrylamide gel electrophoresis. The [14C]MA-labeled ApoB was completely trypsinized and cross-linked to the activated thiol Sepharose 4B beads. The peptides were eluted with DTT and the free -SH groups blocked with IA then separated on FPLC. Two fractions contained [14C]MA. Sequence analyses showed that these labeled peptides contained Cys-51 and Cys-3734, respectively. Evidence suggests that the thiolester is formed between Cys-51 and gamma-Glu-54 for one, and Cys-3734 and beta-Asp-3737 for the other, with Lys and a hydrophobic amino acid, Val/Leu, in between. This is the first evidence for the presence of intramolecular thiolester linkages in ApoB. The presence of high energy, labile thiolester bonds may explain many of the unusual properties of ApoB and LDL.

Concentrations and compositions of plasma lipoprotein subfractions of Lpb5-Lpu1 homozygous and heterozygous swine with hypercholesterolemia.

Pigs with two mutant epitopes, Lpb5 of apolipoprotein B (apoB) and Lpu1 of a yet undefined apolipoprotein, specified by a haplotype Lpb5-Lpu1 and fed a cholesterol-free low fat diet show hypercholesterolemia. The purpose of this study was to establish whether a direct relationship exists between the swine lipoprotein concentration/composition and the genotype for the Lpb5-Lpu1 haplotype; i.e., homozygote versus heterozygote. Lipoproteins of fasted plasma from hypercholesterolemic swine, homozygous (HmHC) and heterozygous (HtHC) for Lpb5-Lpu1, and from normolipidemic (NL) pigs of other Lpb-Lpu haplotypes were separated into five layers by density gradient ultracentrifugation. Layer 1 contained particles of d less than 1.019 g/ml and layer 5 particles of d greater than 1.073 g/ml. Layers 2, 3, and 4 represented subfractions of low density lipoproteins (LDL). The plasma total cholesterol (TC) of the HmHC group (300 +/- 84 mg/dl) was different (P less than 0.05) from the HtHC group (200 +/- 80 mg/dl) and in both HmHC and HtHC, TC was significantly higher (P less than 0.0005 and P less than 0.005, respectively) than that of the NL group (69 +/- 14 mg/dl). The elevation in plasma TC was due to the increased TC in layers 2 and 3: a 13- and 7-fold increase in HmHC and a 7- and 4-fold increase in HtHC in layers 2 and 3, respectively. Parallel increases in unesterified cholesterol were observed in these two layers. Marked increases in apoB were also observed in layers 2 and 3 of HmHC and intermediate increases in apoB in the same two layers of HtHC.(ABSTRACT TRUNCATED AT 250 WORDS)

Leukotriene A4 hydrolase activity of human airway epithelial cells.

Human tracheal epithelial cells were incubated with LTA4 and metabolic products were identified in extracted supernatants by high pressure liquid chromatography, ultraviolet spectroscopy, and gas chromatography-mass spectrometry. In the presence of epithelial cells, LTA4 was converted to LTB4, but not to LTC4 or LTD4. Maximum LTB4 was released at an LTA4 concentration of 3 microM and had occurred by 30 min. LTB4 release was increased in the presence of albumin, but was not affected by extracellular calcium or A23187. This LTA4 hydrolase activity had a slower time course and could not be clearly inactivated by repeated exposure to substrate as is the case for previously described LTA4 hydrolase enzymes. This hydrolase appears to have novel biochemical characteristics.

Presence of covalently attached fatty acids in rat apolipoprotein B via thiolester linkages.

Thiolester linked lipids in rat apolipoprotein B (ApoB) were examined by incubating reduced and carboxymethylated ApoB in 6 M urea buffer with [14C]methylamine at pH 8.5, 30 degrees C. It was observed that [14C]methylamine was covalently incorporated into ApoB, and there was a [14C]methylamine modified product which was lipid in nature. After extraction with organic solvents, the [14C]methylamine labeled product showed its Rf on TLC to be similar to that of the synthetic N-methyl fatty acyl amide. After purification on TLC and transesterification with 3 N methanolic HC1, methyl esters of C16:0, C18:0 and C18:1 fatty acids were identified by gas-liquid chromatography. These results suggest that rat ApoB, similar to human ApoB, contained covalently linked fatty acids through the high energy, labile thiolester bonds.

Lack of effect of probucol on atheroma formation in cholesterol-fed rabbits kept at comparable plasma cholesterol levels.

Rabbits were fed cholesterol for 14 weeks to study the effect of probucol on atheroma formation. Three groups of animals were investigated: group CHOL was fed 1% cholesterol and served as control for group P + CHOL. fed 1% cholesterol and 1% probucol from the onset till the end of the experiment: group CHOL + P received 1% cholesterol throughout the experiment and 1% probucol during the last 4 weeks only. Plasma cholesterol concentrations were monitored at frequent intervals and were modulated by dietary perturbations so that the areas under the curve expressing plasma cholesterol changes with time, were similar in probucol and non-treated rabbits. The efficacy of long-term probucol treatment was evidenced by a significant reduction in plasma apolipoprotein A-I throughout the experiment and lower plasma TBARs during the first 6 weeks, when the hypocholesterolemic effect of probucol was also seen. Two weeks prior to the termination of the experiment, the rabbits were injected with rabbit plasma labeled with [3H]cholesteryl linoleyl ether [( 3H]CLE). Aortic atheromatosis was quantified by determination of total and cholesteryl ester (CE). The aortic cholesterol content was related to the arch, thoracic and abdominal segments, to the surface area of each segment or its dry defatted weight. Total and esterified cholesterol were highest in the aortic arch in all 3 groups when related to any of the above mentioned parameters. No statistically significant difference in aortic total cholesterol and CE content was seen among the three groups studied. The [3H]CLE recovered in the aortic segment correlated with the CE content and the [3H]CLE (dpm)/mg CE in all segments was similar. No statistically significant difference in the [3H]CLE recovered in the aortic segments among the 3 groups was seen. We conclude that in cholesterol-fed rabbits, in which the plasma cholesterol levels were maintained at comparable levels, probucol treatment did not affect plasma CE influx into the aorta and did not attenuate development of aortic atherosclerosis.
